    Razvoj edukacionih mini cnc mašina alatki sa sistemom upravljanja otvorene arhitekture

    Rezime: Ovaj rad predstavlja pregled istraživanja na temu razvijenih edukacionih mini CNC mašna alatki na bazi upravljanja otvorene arhitekture, na PC platformi i na kompaktnim računarskim platformama. Razmatraju se sledeće varijante edukacionih desktop mašina alatki: mini CNC troosna mašina alatka sa tri translatorne ose, troosne mašine alatke sa jednom obrtnom osom i dve translatorne ose i virtuelna 5- osna mašina alatka sa integrisanim upravljanjem. Pokazane su prve razvijene varijante ovih mašina, njihovo konfigurisanje, sistem za programiranje i upravljanje i verifikacija rad

    Rezni alati predstavljaju važan segment u finalnoj obradi drveta. Rezni alati svojom složenom geometrijom omogućuju da se, u jednom prolazu, naprave složene površine na izratku koje su ranijih godina mogle da se kreiraju u više prolaza, korišćenjem manje složenih alata, uz obavezno prisustvo vrlo iskusnih operatera na mašinama. Izrada profilnih koturastih glodala pored konvencionalnih mašina alatki zahteva i petoosnu CNC brusilicu koja profiliše sečivo alata koje je složenog oblika. U ovom radu posebna pažnja posvećena je strategiji 3+2 osne obrade na novoj CNC brusilici uzimajući u obzir činjenicu da je na istoj napravljen veliki broj profilnih koturastih glodala čime je potvrđena industrijska primena nove CNC brusilice i uspostavljene tehnologije

    Genotype–phenotype correlation in contactin-associated protein-like 2 (CNTNAP-2) developmental disorder

    Contactin-associated protein-like 2 (CNTNAP2) gene encodes for CASPR2, a presynaptic type 1 transmembrane protein, involved in cell–cell adhesion and synaptic interactions. Biallelic CNTNAP2 loss has been associated with “Pitt-Hopkins-like syndrome-1” (MIM#610042), while the pathogenic role of heterozygous variants remains controversial. We report 22 novel patients harboring mono- (n = 2) and bi-allelic (n = 20) CNTNAP2 variants and carried out a literature review to characterize the genotype–phenotype correlation. Patients (M:F 14:8) were aged between 3 and 19 years and affected by global developmental delay (GDD) (n = 21), moderate to profound intellectual disability (n = 17) and epilepsy (n = 21). Seizures mainly started in the first two years of life (median 22.5 months). Antiseizure medications were successful in controlling the seizures in about two-thirds of the patients. Autism spectrum disorder (ASD) and/or other neuropsychiatric comorbidities were present in nine patients (40.9%). Nonspecific midline brain anomalies were noted in most patients while focal signal abnormalities in the temporal lobes were noted in three subjects. Genotype–phenotype correlation was performed by also including 50 previously published patients (15 mono- and 35 bi-allelic variants). Overall, GDD (p < 0.0001), epilepsy (p < 0.0001), hyporeflexia (p = 0.012), ASD (p = 0.009), language impairment (p = 0.020) and severe cognitive impairment (p = 0.031) were significantly associated with the presence of biallelic versus monoallelic variants. We have defined the main features associated with biallelic CNTNAP2 variants, as severe cognitive impairment, epilepsy and behavioral abnormalities. We propose CASPR2-deficiency neurodevelopmental disorder as an exclusively recessive disease while the contribution of heterozygous variants is less likely to follow an autosomal dominant inheritance pattern

    Global Analysis of Dynamical Decision-Making Models through Local Computation around the Hidden Saddle

    Bistable dynamical switches are frequently encountered in mathematical modeling of biological systems because binary decisions are at the core of many cellular processes. Bistable switches present two stable steady-states, each of them corresponding to a distinct decision. In response to a transient signal, the system can flip back and forth between these two stable steady-states, switching between both decisions. Understanding which parameters and states affect this switch between stable states may shed light on the mechanisms underlying the decision-making process. Yet, answering such a question involves analyzing the global dynamical (i.e., transient) behavior of a nonlinear, possibly high dimensional model. In this paper, we show how a local analysis at a particular equilibrium point of bistable systems is highly relevant to understand the global properties of the switching system. The local analysis is performed at the saddle point, an often disregarded equilibrium point of bistable models but which is shown to be a key ruler of the decision-making process. Results are illustrated on three previously published models of biological switches: two models of apoptosis, the programmed cell death and one model of long-term potentiation, a phenomenon underlying synaptic plasticity
