    Development of hydroxyapatite-coated nonwovens for efficient isolation of somatic stem cells from adipose tissues

    Adipose-derived stem cells (ASCs) are an attractive cell source for cell therapy. Despite the increasing number of clinical applications, the methodology for ASC isolation is not optimized for every individual. In this study, we developed an effective material to stabilize explant cultures from small-fragment adipose tissues. Methods: Polypropylene/polyethylene nonwoven sheets were coated with hydroxyapatite (HA) particles. Adipose fragments were then placed on these sheets, and their ability to trap tissue was monitored during explant culture. The yield and properties of the cells were compared to those of cells isolated by conventional collagenase digestion. Results: Hydroxyapatite-coated nonwovens immediately trapped adipose fragments when placed on the sheets. The adhesion was stable even in culture media, leading to cell migration and proliferation from the tissue along with the nonwoven fibers. A higher fiber density further enhanced cell growth. Although cells on nonwoven explants could not be fully collected with cell dissociation enzymes, the cell yield was significantly higher than that of conventional monolayer culture without impacting stem cell properties. Conclusions: Hydroxyapatite-coated nonwovens are useful for the effective primary explant culture of connective tissues without enzymatic cell dissociation

    Structural basis for dimer formation of human condensin structural maintenance of chromosome proteins and its implications for single-stranded DNA recognition

    Eukaryotic structural maintenance of chromosome proteins (SMC) are major components of cohesin and condensins that regulate chromosome structure and dynamics during cell cycle. We here determine the crystal structure of human condensin SMC hinge heterodimer with ∼30 residues of coiled coils. The structure, in conjunction with the hydrogen exchange mass spectrometry analyses, revealed the structural basis for the specific heterodimer formation of eukaryotic SMC and that the coiled coils from two different hinges protrude in the same direction, providing a unique binding surface conducive for binding to single-stranded DNA. The characteristic hydrogen exchange profiles of peptides constituted regions especially across the hinge-hinge dimerization interface, further suggesting the structural alterations upon single-stranded DNA binding and the presence of a half-opened state of hinge heterodimer. This structural change potentially relates to the DNA loading mechanism of SMC, in which the hinge domain functions as an entrance gate as previously proposed for cohesin.     Development of early neutropenic fever, with or without bacterial infection, is still a significant complication after reduced-intensity stem cell transplantation

    AbstractLittle information is available on the clinical characteristics of infectious complications that occur in the early period after reduced-intensity stem cell transplantation (RIST). We retrospectively investigated the clinical features of neutropenic fever and infectious episodes within 30 days after RIST in 76 patients who had received fluoroquinolones as part of their antibacterial prophylaxis. Preparative regimens included cladribine 0.66 mg/kg or fludarabine 180 mg/m2 plus busulfan 8 mg/kg. All but 1 patient survived 30 days after transplantation, and 75 patients (99%) became neutropenic within a median duration of 9 days. Neutropenic fever was observed in 29 patients (38%), and bacterial infection was confirmed in 15 (20%) of these, including bacteremia (n = 13), bacteremia plus pneumonia (n = 1), and urinary tract infection (n = 1). The causative organisms were gram-positive (n = 9) and gram-negative organisms (n = 7), with a mortality rate of 6%. Neither viral nor fungal infection was documented. Multivariate analysis showed that the presence of neutropenia at the initiation of preparative regimens was an independent risk factor for subsequent documented bacterial infections (P = .026; 95% confidence interval, 1.25–35.1). We conclude that neutropenic fever and bacteremia remain common complications in RIST

    Factors increasing nursing students’ satisfaction with practical training in the perioperative period, with a focus on the relationship between training situations and the guidance of perioperative nurses and teachers

    To clarify the levels of satisfaction of nursing students with perioperative practical training and the factors influencing these levels, we conducted an anonymous self-questionnaire survey of 83 nursing students who had participated in perioperative practical training. The survey included questions about training situations, levels of satisfaction with the training, and evaluation of the training and teaching system. Analysis of the impacts of the perioperative practical training on the students’ learning showed that students who had observed the surgery through to completion showed significantly greater interest in operating room nursing, and that nurses’ guidance in open surgery was evaluated significantly more highly than their guidance in endoscopic surgery. Deepening of learning by observing surgery, receiving teachers’ guidance, and receiving guidance from nurses during surgery affected the students’ levels of satisfaction with perioperative practical training.  To increase these levels of satisfaction, continuing surgical nursing education by teachers and nurses is needed. Before surgery, we need to improve students’ preparation for training by prior learning; during surgery, we need to tell students the progress of the ongoing surgery and the roles of nurses as needed. In addition, practical experience in nursing skills enables students to deepen their understanding of surgical nursing. These factors will increase the satisfaction levels of nursing students.看護学生の手術室実習における実習満足度とその影響要因を明らかにすることを目的とし、手術室実習 を行った看護学生83名に無記名自記式質問紙調査を行った。調査内容は実習状況、実習満足度のほか、 実習および指導体制への評価とした。手術室実習が看護学生の学びに与える影響を分析した結果、実習 状況別では手術終了まで見学した学生は興味が有意に高く、切開手術群は鏡視下手術群に比して手術室 看護師の指導への評価が有意に高かった。手術室実習満足度には手術見学による学びの深まり、教員の 指導、手術中の看護師の指導が影響していることが明らかになった。  手術室実習の満足度を高めるためには、教員と看護師による継続的な手術看護教育が求められる。術 前は事前学習により実習準備状態を高める工夫を行い、術中は手術の進行状況や看護師の役割を随時伝 えることや看護実践の経験により手術看護の理解を深めることが満足度の向上につながる

    General characterization of Antarctic micrometeorites collected by the 39th Japanese Antarctic Research Expedition: Consortium studies of JARE AMMs (III)

    From November 1998 to January 1999,the 39th Japanese Antarctic Research Expedition (JARE-39) undertook Japanese first large-scale collection of Antarctic micrometeorites (AMMs), with sizes larger than 10μm, at the Meteorite Ice Field around the Yamato Mountains in Antarctica (at three different locations, for a total of 24 collection sites). The number of collected AMMs larger than 40μm is estimated to be about 5000. Here we present the general characterization (i.e., micro-morphology and surface chemical composition using SEM/EDS) of ∿810 AMMs chosen from 5 of the 24 sites. Additionally, the mineral composition of 61 out of 810 AMMs was determined by Synchrotron X-ray radiation. Preliminary results on mineralogical and chemical compositions show similarities with that of previous studies, even though a pronounced alteration of some AMMs is noticed. A correlation is found between the Mg/Si ratio at the sample\u27s surfaces of unmelted AMMs and the age of snow/ice in which the AMMs are embedded
