    Immunofluorescent Localization of RuBPCase in Degraded C\u3csub\u3e4\u3c/sub\u3e Grass Tissue

    Digestion-resistant tissues found in C4 grasses may allow soluble protein to escape rumen degradation. The objective of this study was to use immunofluorescent localization to follow loss of ribulose 1,5-bisphosphate carboxylase/oxygenase (RuBPCase) from switchgrass (Panicum virgatum L.) and big bluestem (Andropogon gerardii Vitman) parenchyma bundle sheath cells (BSC) during rumen degradation. Fluorescent signal was observed from switchgrass and big bluestem BSC through 24 and 16 h in situ digestion, respectively, and from BSC associated with both intact tissue fragments, and isolated vascular bundles in omasal digesta and fecal material from steers (Bos tarus L.) grazing switchgrass. Immunofluorescent localization demonstrated that in certain C4 grasses (i) parenchyma BSC can protect RuBPCase from degradation through 24-h in situ incubation, (ii) BSC containing RuBPCase can exit the rumen prior to degradation, and (iii) protein protected by BSC can escape degradation in the whole gastrointestinal tract and be excreted

    Upregulation of PTEN in Glioma Cells by Cord Blood Mesenchymal Stem Cells Inhibits Migration via Downregulation of the PI3K/Akt Pathway

    PTEN (phosphatase and tensin homologue deleted on chromosome ten) is a tumor suppressor gene implicated in a wide variety of human cancers, including glioblastoma. PTEN is a major negative regulator of the PI3K/Akt signaling pathway. Most human gliomas show high levels of activated Akt, whereas less than half of these tumors carry PTEN mutations or homozygous deletions. The unique ability of mesenchymal stem cells to track down tumor cells makes them as potential therapeutic agents. Based on this capability, new therapeutic approaches have been developed using mesenchymal stem cells to cure glioblastoma. However, molecular mechanisms of interactions between glioma cells and stem cells are still unknown.In order to study the mechanisms by which migration of glioma cells can be inhibited by the upregulation of the PTEN gene, we studied two glioma cell lines (SNB19 and U251) and two glioma xenograft cell lines (4910 and 5310) alone and in co-culture with human umbilical cord blood-derived mesenchymal stem cells (hUCBSC). Co-cultures of glioma cells showed increased expression of PTEN as evaluated by immunofluorescence and immunoblotting assays. Upregulation of PTEN gene is correlated with the downregulation of many genes including Akt, JUN, MAPK14, PDK2, PI3K, PTK2, RAS and RAF1 as revealed by cDNA microarray analysis. These results have been confirmed by reverse-transcription based PCR analysis of PTEN and Akt genes. Upregulation of PTEN resulted in the inhibition of migration capability of glioma cells under in vitro conditions. Also, wound healing capability of glioma cells was significantly inhibited in co-culture with hUCBSC. Under in vivo conditions, intracranial tumor growth was inhibited by hUCBSC in nude mice. Further, hUCBSC upregulated PTEN and decreased the levels of XIAP and Akt, which are responsible for the inhibition of tumor growth in the mouse brain.Our studies indicated that upregulation of PTEN by hUCBSC in glioma cells and in the nude mice tumors downregulated Akt and PI3K signaling pathway molecules. This resulted in the inhibition of migration as well as wound healing property of the glioma cells. Taken together, our results suggest hUCBSC as a therapeutic agent in treating malignant gliomas

    Search for W~1Z~2\widetilde{W}_1\widetilde{Z}_2 Production via Trilepton Final States in ppˉp\bar{p} collisions at s=1.8\sqrt{s}=1.8 TeV

    We have searched for associated production of the lightest chargino, W~1\widetilde{W}_1, and next-to-lightest neutralino, Z~2\widetilde{Z}_2, of the Minimal Supersymmetric Standard Model in ppˉp\bar{p} collisions at \mbox{s\sqrt{s} = 1.8 TeV} using the \D0 detector at the Fermilab Tevatron collider. Data corresponding to an integrated luminosity of 12.5±0.7\pm 0.7 \ipb were examined for events containing three isolated leptons. No evidence for W~1Z~2\widetilde{W}_1\widetilde{Z}_2 pair production was found. Limits on σ(W~1Z~2)\sigma(\widetilde{W}_1\widetilde{Z}_2)Br(W~1lνZ~1)(\widetilde{W}_1\to l\nu\widetilde{Z}_1)Br(Z~2llˉZ~1)(\widetilde{Z}_2\to l\bar{l}\widetilde{Z}_1) are presented.Comment: 17 pages (13 + 1 page table + 3 pages figures). 3 PostScript figures will follow in a UUEncoded, gzip'd, tar file.     Measurement of the WW Boson Mass

    A measurement of the mass of the WW boson is presented based on a sample of 5982 WeνW \rightarrow e \nu decays observed in ppp\overline{p} collisions at s\sqrt{s} = 1.8~TeV with the D\O\ detector during the 1992--1993 run. From a fit to the transverse mass spectrum, combined with measurements of the ZZ boson mass, the WW boson mass is measured to be MW=80.350±0.140(stat.)±0.165(syst.)±0.160(scale)GeV/c2M_W = 80.350 \pm 0.140 (stat.) \pm 0.165 (syst.) \pm 0.160 (scale) GeV/c^2.Comment: 12 pages, LaTex, style Revtex, including 3 postscript figures (submitted to PRL

    Probing BFKL Dynamics in the Dijet Cross Section at Large Rapidity Intervals in ppbar Collisions at sqrt{s}=1800 and 630 GeV

    Inclusive dijet production at large pseudorapidity intervals (delta_eta) between the two jets has been suggested as a regime for observing BFKL dynamics. We have measured the dijet cross section for large delta_eta in ppbar collisions at sqrt{s}=1800 and 630 GeV using the DO detector. The partonic cross section increases strongly with the size of delta_eta. The observed growth is even stronger than expected on the basis of BFKL resummation in the leading logarithmic approximation. The growth of the partonic cross section can be accommodated with an effective BFKL intercept of a_{BFKL}(20GeV)=1.65+/-0.07.Comment: Published in Physical Review Letter

    Search for Top Squark Pair Production in the Dielectron Channel

    This report describes the first search for top squark pair production in the channel stop_1 stopbar_1 -> b bbar chargino_1 chargino_1 -> ee+jets+MEt using 74.9 +- 8.9 pb^-1 of data collected using the D0 detector. A 95% confidence level upper limit on sigma*B is presented. The limit is above the theoretical expectation for sigma*B for this process, but does show the sensitivity of the current D0 data set to a particular topology for new physics.Comment: Five pages, including three figures, submitted to PRD Brief Report

    Direct Measurement of the Top Quark Mass at D0

    We determine the top quark mass m_t using t-tbar pairs produced in the D0 detector by \sqrt{s} = 1.8 TeV p-pbar collisions in a 125 pb^-1 exposure at the Fermilab Tevatron. We make a two constraint fit to m_t in t-tbar -> b W^+bbar W^- final states with one W boson decaying to q-qbar and the other to e-nu or mu-nu. Likelihood fits to the data yield m_t(l+jets) = 173.3 +- 5.6 (stat) +- 5.5 (syst) GeV/c^2. When this result is combined with an analysis of events in which both W bosons decay into leptons, we obtain m_t = 172.1 +- 5.2 (stat) +- 4.9 (syst) GeV/c^2. An alternate analysis, using three constraint fits to fixed top quark masses, gives m_t(l+jets) = 176.0 +- 7.9 (stat) +- 4.8 (syst) GeV/C^2, consistent with the above result.     Search for a Fourth Generation Charge -1/3 Quark via Flavor Changing Neutral Current Decay

    We report on a search for pair production of a fourth generation charge -1/3 quark (b') in pbar p collisions at sqrt(s) = 1.8 TeV at the Fermilab Tevatron using an integrated luminosity of 93 pb^-1. Both quarks are assumed to decay via flavor changing neutral currents (FCNC). The search uses the signatures gamma + 3 jets + mu-tag and 2 gamma + 2 jets. We see no significant excess of events over the expected background. We place an upper limit on the production cross section times branching fraction that is well below theoretical expectations for a b' quark decaying exclusively via FCNC for b' quark masses up to m(Z) + m(b).Comment: Eleven pages, two postscript figures, submitted to Physical Review Letter

    The D0 collaboration has performed a measurement of the top quark mass based on six candidate events for the process t tbar -> b W+ bbar W-, where the W bosons decay to e nu or mu nu. This sample was collected during an exposure of the D0 detector to an integrated luminosity of 125 pb^-1 of sqrt(s)=1.8 TeV p-pbar collisions. We obtain mt = 168.4 +- 12.3 (stat) +- 3.7 (sys) GeV/c^2, consistent with the measurement obtained using single-lepton events. Combination of the single-lepton and dilepton results yields mt = 172.0 +- 7.5 GeV/c^2.Comment: 12 pages, 3 figure

    Search for Electroweak Production of Single Top Quarks in ppbar Collisions

    We present a search for electroweak production of single top quarks in the electron+jets and muon+jets decay channels. The measurements use ~90 pb^-1 of data from Run 1 of the Fermilab Tevatron collider, collected at 1.8 TeV with the DZero detector between 1992 and 1995. We use events that include a tagging muon, implying the presence of a b jet, to set an upper limit at the 95% confidence level on the cross section for the s-channel process ppbar->tb+X of 39 pb.
