    Untersuchung von Z-Scheiben-Proteinen in quergestreiften Muskeln mittels 2-D-Elektrophorese

    Ziel dieser Arbeit war es, Faktoren bzw. Mechanismen zu identifizieren, die an der Regulation der MLP-abhängigen Signaltransduktion im Herzen beteiligt sind. Dafür wurde 1. das homozygote MLP knock-out (KO) Mausmodel bezüglich kardialer Proteomveränderungen untersucht und 2. posttranslationale Modifikationen in MLP identifiziert und charakterisiert. 2-D-DIGE-Analysen an ein, vier und 12 Wochen alten MLP KO und wt Herzen ergaben im MLP KO Veränderungen in 200 Proteinspots. Im kardialen MLP wurden zwei konservierte Phosphorylierungsstellen, an S95 und S46, identifiziert. PKC epsilon phosphoryliert MLP in vitro an S46. ERK1/2 phosphoryliert und Calcineurin (CnA) dephosphoryliert MLP an S95. Die MLP S95 Phosphorylierung wird durch den CnA- und MAPK-Signalweg reguliert. In hypertrophen Hundeherzen mit diastolischer Herzinsuffizienz war die S95 Phosphorylierung um 40% vermindert, was vermutlich auf eine verstärkte CnA-vermittelte Dephosphorylierung zurückzuführen ist

    FastAMI -- a Monte Carlo Approach to the Adjustment for Chance in Clustering Comparison Metrics

    Clustering is at the very core of machine learning, and its applications proliferate with the increasing availability of data. However, as datasets grow, comparing clusterings with an adjustment for chance becomes computationally difficult, preventing unbiased ground-truth comparisons and solution selection. We propose FastAMI, a Monte Carlo-based method to efficiently approximate the Adjusted Mutual Information (AMI) and extend it to the Standardized Mutual Information (SMI). The approach is compared with the exact calculation and a recently developed variant of the AMI based on pairwise permutations, using both synthetic and real data. In contrast to the exact calculation our method is fast enough to enable these adjusted information-theoretic comparisons for large datasets while maintaining considerably more accurate results than the pairwise approach.Comment: Accepted at AAAI 202

    Evaluation of Postsurgical Hyperalgesia and Sensitization After Open Inguinal Hernia Repair: A Useful Model for Neuropathic Pain?

    Cutaneous mechanical hyperalgesia can be induced in healthy volunteers in early phase analgesic studies to model central sensitization, a key mechanism of persistent pain. However, such hyperalgesia is short-lived (a matter of hours), and is used only for assessing only single drug doses. In contrast, postsurgical peri-incisional hyperalgesia may be more persistent and hence be a more useful model for the assessment of the efficacy of new analgesics. We undertook quantitative sensory testing in 18 patients at peri-incisional and nonoperated sites before open inguinal hernia repair and up to the 24th postsurgical week. The spatial extent of punctate hyperalgesia and brush allodynia at the peri-incisional site were greatest at weeks 2 and 4, but had resolved by week 24. Heat allodynia, suggestive of local inflammation or peripheral sensitization, was not observed; instead, there were deficits in cold and heat sensory detection that persisted until week 24. The findings suggest that central sensitization contributes significantly to mechanical hyperalgesia at the peri-incisional site. The prolonged duration of hyperalgesia would be advantageous as a pain model, but there was considerable variability of mechanical hyperalgesia in the cohort; the challenges of recruitment may limit its use to small, early phase analgesic studies. PERSPECTIVE: Peri-incisional mechanical hyperalgesia persists for ≥4 weeks after open inguinal hernia repair and reflects central sensitization; this may have usefulness as a model of chronic pain to assess the potential of antineuropathic analgesics.Unrestricted educational grant from GlaxoSmithKline U
