    Effects of climate and plant phenology on recruitment of moose at the southern extent of their range

    Climate plays a fundamental role in limiting the range of a species, is a key factor in the dynamics of large herbivores, and is thought to be involved in declines of moose populations in recent decades. We examined effects of climate and growing-season phenology on recruitment (8–9 months old) of young Shiras moose (Alces alces shirasi) over three decades, from 18 herds, across a large geographic area encompassing much of the southern extent of their range. Recruitment declined in 8 of 18 herds during 1980–2009, whereas others did not exhibit a temporal trend (none showed a positive trend). During those three decades, seasonal temperatures increased, spring–summer precipitation decreased, and spring occurred earlier, became shorter in duration, and green-up occurred faster. Recruitment was influenced negatively by warm temperatures during the year before young were born, but only for herds with declining recruitment. Dry spring–summers of the previous year and rapid rates of spring green-up in the year of birth had similar negative influences across declining and stable herds. Those patterns indicate both direct (yeart) and delayed (yeart−1) effects of weather and plant phenology on recruitment of young, which we hypothesize was mediated through effects on maternal nutritional condition. Suppressed nutrition could have been induced by (1) increased thermoregulatory costs associated with warming temperatures and (2) shortened duration of availability of high-quality forage in spring. Progressive reductions in net energetic gain for species that are sensitive to climate may continue to hamper individual fitness and population dynamics

    Effects of Heparin and Enoxaparin on APP Processing and Aβ Production in Primary Cortical Neurons from Tg2576 Mice

    BACKGROUND Alzheimer's disease (AD) is caused by accumulation of Aβ, which is produced through sequential cleavage of β-amyloid precursor protein (APP) by the β-site APP cleaving enzyme (BACE1) and γ-secretase. Enoxaparin, a low molecular weight form of the glycosaminoglycan (GAG) heparin, has been reported to lower Aβ plaque deposition and improve cognitive function in AD transgenic mice. METHODOLOGY/PRINCIPAL FINDINGS We examined whether heparin and enoxaparin influence APP processing and inhibit Aβ production in primary cortical cell cultures. Heparin and enoxaparin were incubated with primary cortical cells derived from Tg2576 mice, and the level of APP and proteolytic products of APP (sAPPα, C99, C83 and Aβ) was measured by western blotting. Treatment of the cells with heparin or enoxaparin had no significant effect on the level of total APP. However, both GAGs decreased the level of C99 and C83, and inhibited sAPPα and Aβ secretion. Heparin also decreased the level of β-secretase (BACE1) and α-secretase (ADAM10). In contrast, heparin had no effect on the level of ADAM17. CONCLUSIONS/SIGNIFICANCE The data indicate that heparin and enoxaparin decrease APP processing via both α- and β-secretase pathways.     Survival of White-Tailed Deer Fawns in the Grasslands of the Northern Great Plains

    Environmental factors, such as forest characteristics, have been linked to fawn survival in eastern and southern white-tailed deer ( Odocoileus virginianus) populations. In the Great Plains, less is known about how intrinsic and habitat factors influence fawn survival. During 2007-2009, we captured and radiocollared 81 fawns in north-central South Dakota and recorded 23 mortalities, of which 18 died before 1 September. Predation accounted for 52.2% of mortality; remaining mortality included human (hunting, vehicle, and farm accident; 26.1%) and hypothermia (21.7%). Coyotes (Canis latrans) accounted for 83.3% of predation on fawns. We used known-fate analysis in Program MARK to estimate summer (15 May-31 Aug) survival rates and investigated the influence of intrinsic and habitat variables on survival. We developed 2 a priori model sets, including intrinsic variables and a test of annual variation in survival (model set 1) and habitat variables (model set 2). Model set 1 indicated that summer survival varied among years (2007-2009); annual survival rates were 0.94 (SE = 0.06, n = 22), 0.78 (SE = 0.09, n = 27), and 0.54 (SE = 0.10, n = 32), respectively. Model set 2 indicated that survival was further influenced by patch density of cover habitats (Conservation Reserve Program [CRP]-grasslands, forested cover, and wetlands). Mean CRPgrassland and wetland patch density (no. patches/100 ha) were greater (P \u3c 0.001) in home-range areas of surviving fawns (xcRPPD = 1.81, SE = 0.10, n = 63; XWe,PD = 1.75, SE = 0.14, n = 63, respectively) than in home-range areas of fawns that died (xcRPPD = 0.16, SE = 0.04, n = 18; XWe,PD = 1.28, SE = 0.10, n = 18, respectively). Mean forested cover patch density was less (P \u3c 0.001) in home-range areas of surviving fawns (fycpn = 0.77, SE = 0.10, n = 63) than in home-range areas of fawns that died (XF CPD = 1.49, SE = 0.21, n = 18). Our results indicate that management activities should focus on CRP-grassland and wetland habitats in order to maintain or improve fawn survival in the northern Great Plains, rather than forested cover composed primarily of tree plantings and shelterbelts

    Conventional Fourier transform IR (FTIR) spectroscopes cannot be used to perform real time in situ IR reflection absorption spectroscopy at monolayer sensitivity for high deposition rates (a couple of tens to hundreds of nm/s) which can be obtained when using an expanding thermal deposition plasma. Therefore a new anal. tool has been developed. The tool is based on a fast optical scanner in combination with conventional grating technol. This results in a loss of spectral range with respect to FTIR spectroscopes, but a significant gain is obtained in time resoln. For the combination used this makes it possible to measure at time resoln. as low as 1.3 ms and resoln. of 24 cm-1 at 1000 cm-1. The absorption sensitivity for single reflection at the best time resoln. is approx. 10-2, but can be improved by using signal enhancement techniques. Here attenuated total reflection is used and the best sensitivity obtained is approx. 10-3, which is close to monolayer sensitivity for various absorption bands in the IR spectrum of silicon oxide films. Monolayer sensitivity can be obtained by averaging multiple spectra, however this will cause the time resoln. to decreas

    Cognitive improvement in Alzheimer\u27s disease (AD) patients treated with intravenous immunoglobulin (IvIg) has been attributed to its antibodies to amyloid beta (A beta) We compared the concentrations of specific antibodies to soluble A beta 1-42 conformations, namely A beta 1-42 monomer and A beta 1-42 soluble oligomers, between three IvIg preparations. Gamunex, Gammagard. and Flebogamma. To determine specific antibody Concentrations to these A beta 1-42 conformations. nonspecific binding of the IvIg preparations to the An reverse sequence, A beta 42-1. was subtracted These antibodies were measured in untreated IvIg preparations and also after they were treated to dissociate antibody-antigen complexes. because this procedure has been reported to increase the detectable levels of serum anti-A beta antibodies. Antibody levels to A beta 1-42 monomer were significantly higher in untreated Gamunex than in the other two IvIg preparations, and antibody-antigen dissociation increased the measured anti-A beta monomer concentrations in Gamunex and Gammagard Dissociated Gamunex and Gammagard had higher anti-A beta monomer levels than Flebogamma. Generally similar results were found for antibodies to soluble A beta 1-42 oligomers. with the exception that after antibody-antigen dissociation, only Gammagard had significantly higher antibody levels than Flebogamma. These differences in antibody concentrations to A beta 1-42 conformations (particularly to A beta 1-42 soluble oligomers, thought to be the most neurotoxic conformation of soluble A beta) and the increased availability of these antibodies after antibody-antigen complex dissociation have important implications for IvIg treatment of AD patients
