    Forced desorption of nanoparticles from an oil-water interface.

    While nanoparticle adsorption to fluid interfaces has been studied from a fundamental standpoint and exploited in application, the reverse process, that is, desorption and disassembly, remains relatively unexplored. Here we demonstrate the forced desorption of gold nanoparticles capped with amphiphilic ligands from an oil-water interface. A monolayer of nanoparticles is allowed to spontaneously form by adsorption from an aqueous suspension onto a drop of oil and is subsequently compressed by decreasing the drop volume. The surface pressure is monitored by pendant drop tensiometry throughout the process. Upon compression, the nanoparticles are mechanically forced out of the interface into the aqueous phase. An optical method is developed to measure the nanoparticle area density in situ. We show that desorption occurs at a coverage that corresponds to close packing of the ligand-capped particles, suggesting that ligand-induced repulsion plays a crucial role in this process. © 2011 American Chemical Society

    Macrophage-derived upd3 Cytokine causes impaired glucose homeostasis and reduced lifespan in drosophila fed a lipid-rich diet

    Long-term consumption of fatty foods is associated with obesity, macrophage activation and inflammation, metabolic imbalance, and a reduced lifespan. We took advantage of Drosophila genetics to investigate the role of macrophages and the pathway(s) that govern their response to dietary stress. Flies fed a lipid-rich diet presented with increased fat storage, systemic activation of JAK-STAT signaling, reduced insulin sensitivity, hyperglycemia, and a shorter lifespan. Drosophila macrophages produced the JAK-STAT-activating cytokine upd3, in a scavenger-receptor (crq) and JNK-dependent manner. Genetic depletion of macrophages or macrophage-specific silencing of upd3 decreased JAK-STAT activation and rescued insulin sensitivity and the lifespan of Drosophila, but did not decrease fat storage. NF-κB signaling made no contribution to the phenotype observed. These results identify an evolutionarily conserved “scavenger receptor-JNK-type 1 cytokine” cassette in macrophages, which controls glucose metabolism and reduces lifespan in Drosophila maintained on a lipid-rich diet via activation of the JAK-STAT pathway

    Modelling localised fracture of reinforced concrete structures

    This paper presents a robust finite element procedure for simulating the localised fracture of reinforced concrete members. In this new model the concrete member is modelled as an assembly of plain concrete, reinforcing steel bar and bond-link elements. The 4-node quadrilateral elements are used for 2D modelling of plain concrete elements, in which the extended finite element method is adopted to simulate the formation and growth of individual cracks. The reinforcing steel bars are modelled by using a 3-node beam-column element. 2-node bond-link elements are employed for modelling the interaction between plain concrete and reinforcing steel bar elements. It is evident that the nonlinear procedure proposed in this paper can properly model the formation and propagation of individual localised cracks within the reinforced concrete structures. The model presented in this paper enables the researchers and designers to access the integrity of reinforced concrete members under extreme loading conditions by using mesh independent extended finite element method.The support of the Engineering and Physical Sciences Research Council of Great Britain under Grant No. EP/I031553/1

    Metabolic flexibility as a major predictor of spatial distribution in microbial communities

    A better understand the ecology of microbes and their role in the global ecosystem could be achieved if traditional ecological theories can be applied to microbes. In ecology organisms are defined as specialists or generalists according to the breadth of their niche. Spatial distribution is often used as a proxy measure of niche breadth; generalists have broad niches and a wide spatial distribution and specialists a narrow niche and spatial distribution. Previous studies suggest that microbial distribution patterns are contrary to this idea; a microbial generalist genus (Desulfobulbus) has a limited spatial distribution while a specialist genus (Methanosaeta) has a cosmopolitan distribution. Therefore, we hypothesise that this counter-intuitive distribution within generalist and specialist microbial genera is a common microbial characteristic. Using molecular fingerprinting the distribution of four microbial genera, two generalists, Desulfobulbus and the methanogenic archaea Methanosarcina, and two specialists, Methanosaeta and the sulfate-reducing bacteria Desulfobacter were analysed in sediment samples from along a UK estuary. Detected genotypes of both generalist genera showed a distinct spatial distribution, significantly correlated with geographic distance between sites. Genotypes of both specialist genera showed no significant differential spatial distribution. These data support the hypothesis that the spatial distribution of specialist and generalist microbes does not match that seen with specialist and generalist large organisms. It may be that generalist microbes, while having a wider potential niche, are constrained, possibly by intrageneric competition, to exploit only a small part of that potential niche while specialists, with far fewer constraints to their niche, are more capable of filling their potential niche more effectively, perhaps by avoiding intrageneric competition. We suggest that these counter-intuitive distribution patterns may be a common feature of microbes in general and represent a distinct microbial principle in ecology, which is a real challenge if we are to develop a truly inclusive ecology

    Nodular lymphocyte predominant Hodgkin lymphoma behaves as a distinct clinical entity with good outcome: evidence from 14-year followup in the West of Scotland Cancer Network

    Clinically and biologically, nodular lymphocyte predominant Hodgkin lymphoma (NLPHL) has much more in common with germinal-center derived B-cell non-Hodgkin lymphoma (NHL) than with classical Hodgkin lymphoma (cHL). Management of NLPHL remains controversial. In a 14-year multicenter series, 69 cases were analyzed, and the median follow-up was 53 months (range 11–165.) B-symptoms were present in only 4.3% of patients, and 81.1% of patients had stage I/II disease. Treatment was with radiotherapy (53.6%), chemotherapy (21.7%), combined modality (17.4%), and observation (7.2%). In all, 10.1% of patients relapsed and 2.9% of patients developed high-grade transformation to DLBCL. All relapses and transformations were salvageable. No patient died of their disease. The 5-year relapse-free survival was 92%, transformation-free survival 98.4%, and overall survival 100%. We conclude that NLPHL behaves as a distinct clinical entity, often presenting at an early stage without risk factors. It has an excellent outcome. It may be possible, in early-stage disease, to reduce the intensity of therapy in NLPHL, to single-modality radiotherapy, without affecting OS

    Deep Active Inference for Partially Observable MDPs

    Deep active inference has been proposed as a scalable approach to perception and action that deals with large policy and state spaces. However, current models are limited to fully observable domains. In this paper, we describe a deep active inference model that can learn successful policies directly from high-dimensional sensory inputs. The deep learning architecture optimizes a variant of the expected free energy and encodes the continuous state representation by means of a variational autoencoder. We show, in the OpenAI benchmark, that our approach has comparable or better performance than deep Q-learning, a state-of-the-art deep reinforcement learning algorithm.Comment: 1st International Workshop on Active inference, European Conference on Machine Learning (ECML/PCKDD 2020

    Changes in chemical composition and bioassay assessment of nutritional potentials of almond fruit waste as an alternative feedstuff for livestock

    Changes in chemical composition upon processing and bioassay assessment of nutritional potentials of almond fruit waste as an alternative feedstuff were conducted using day-oldcockerels.Proximate analyses revealed that AFW contained valuable nutrients, carbohydrate/dry matter, protein, fat, fiber, mineral matter (ash). The concentration of some of these nutrients increased as the raw AFW was treated. The raw AFW gave on analysis quantitatively highconcentrations of the chemical compounds, tannins, hydrocyanides, phytic acid and other unquantified chemical compounds like oxalates assessed qualitatively. Subjecting raw AFW to lactic fermentation and enzymes treatments reduced the levels of the phytochemicals. Bioassay of the treated and untreated AFW using day-old cockerels and considering performance parameters showed that treated AFW improved feed intake, body weight gain and feed conversion ratio even better than the results obtained on these indices on the reference diet (p0.05). Untreated AFW elicited misperformance and high mortality of the test birds. It was concluded that the nutritional value of AFW, if could be used as an alternative feedstuff for animals, must be given adequate treatments

    Assessing the language of chat for teamwork dialogue

    In technology enhanced language learning, many pedagogical activities involve students in online discussion such as synchronous chat, in order to help them practice their language skills. Besides developing the language competency of students, it is also crucial to nurture their teamwork competencies for today's global and complex environment. Language communication is an important glue of teamwork. In order to assess the language of chat for teamwork dimensions, several text mining methods are pos sible. However, difficulties arise such as pre-processing being a black box and classification approaches and algorithms being dependent on the context. To address these issues, the study will evaluate and explain preprocessing and classification methods used to analyze teamwork dialogue from a dataset of chat data. Analytics methods evaluated in this study provide a direction for assessing the language of chat for teamwork dialogue and can help extend the work of technology enhanced language learning to n ot only focus on academic competency, but on the communication aspect too

    An Iterative CT Reconstruction Algorithm for Fast Fluid Flow Imaging

    The study of fluid flow through solid matter by computed tomography (CT) imaging has many applications, ranging from petroleum and aquifer engineering to biomedical, manufacturing, and environmental research. To avoid motion artifacts, current experiments are often limited to slow fluid flow dynamics. This severely limits the applicability of the technique. In this paper, a new iterative CT reconstruction algorithm for improved a temporal/spatial resolution in the imaging of fluid flow through solid matter is introduced. The proposed algorithm exploits prior knowledge in two ways. First, the time-varying object is assumed to consist of stationary (the solid matter) and dynamic regions (the fluid flow). Second, the attenuation curve of a particular voxel in the dynamic region is modeled by a piecewise constant function over time, which is in accordance with the actual advancing fluid/air boundary. Quantitative and qualitative results on different simulation experiments and a real neutron tomography data set show that, in comparison with the state-of-the-art algorithms, the proposed algorithm allows reconstruction from substantially fewer projections per rotation without image quality loss. Therefore, the temporal resolution can be substantially increased, and thus fluid flow experiments with faster dynamics can be performed

    Distinct emphysema subtypes defined by quantitative CT analysis are associated with specific pulmonary matrix metalloproteinases.

    BACKGROUND: Emphysema is characterised by distinct pathological sub-types, but little is known about the divergent underlying aetiology. Matrix-metalloproteinases (MMPs) are proteolytic enzymes that can degrade the extracellular matrix and have been identified as potentially important in the development of emphysema. However, the relationship between MMPs and emphysema sub-type is unknown. We investigated the role of MMPs and their inhibitors in the development of emphysema sub-types by quantifying levels and determining relationships with these sub-types in mild-moderate COPD patients and ex/current smokers with preserved lung function. METHODS: Twenty-four mild-moderate COPD and 8 ex/current smokers with preserved lung function underwent high resolution CT and distinct emphysema sub-types were quantified using novel local histogram-based assessment of lung density. We analysed levels of MMPs and tissue inhibitors of MMPs (TIMPs) in bronchoalveolar lavage (BAL) and assessed their relationship with these emphysema sub-types. RESULTS: The most prevalent emphysema subtypes in COPD subjects were mild and moderate centrilobular (CLE) emphysema, while only small amounts of severe centrilobular emphysema, paraseptal emphysema (PSE) and panlobular emphysema (PLE) were present. MMP-3, and -10 associated with all emphysema sub-types other than mild CLE, while MMP-7 and -8 had associations with moderate and severe CLE and PSE. MMP-9 also had associations with moderate CLE and paraseptal emphysema. Mild CLE occurred in substantial quantities irrespective of whether airflow obstruction was present and did not show any associations with MMPs. CONCLUSION: Multiple MMPs are directly associated with emphysema sub-types identified by CT imaging, apart from mild CLE. This suggests that MMPs play a significant role in the tissue destruction seen in the more severe sub-types of emphysema, whereas early emphysematous change may be driven by a different mechanism.
