    Polyamines are present at high levels in proliferating cells, including cancerous cells and protozoan parasites and the inhibition of their synthesis has been exploited in antiproliferative strategies. Inhibition of the malaria parasite’s polyamine biosynthetic pathway causes cytostatic arrest in the trophozoite stage but does not cure in vivo infections in the murine model of malaria. This is possibly due to exogenous polyamine salvage from the host, which replenishes the intracellular polyamine pool. This implies that disruption of polyamine metabolism as an antimalarial chemotherapy strategy may require targeting both polyamine biosynthesis and transport simultaneously

    K2 Variable Catalogue: Variable Stars and Eclipsing Binaries in K2 Campaigns 1 and 0

    We have created a catalogue of variable stars found from a search of the publicly available K2 mission data from Campaigns 1 and 0. This catalogue provides the identifiers of 8395 variable stars, including 199 candidate eclipsing binaries with periods up to 60d and 3871 periodic or quasi-periodic objects, with periods up to 20d for Campaign 1 and 15d for Campaign 0. Lightcurves are extracted and detrended from the available data. These are searched using a combination of algorithmic and human classification, leading to a classifier for each object as an eclipsing binary, sinusoidal periodic, quasi periodic, or aperiodic variable. The source of the variability is not identified, but could arise in the non-eclipsing binary cases from pulsation or stellar activity. Each object is cross-matched against variable star related guest observer proposals to the K2 mission, which specifies the variable type in some cases. The detrended lightcurves are also compared to lightcurves currently publicly available. The resulting catalogue is made available online via the MAST archive at https://archive.stsci.edu/prepds/k2varcat/, and gives the ID, type, period, semi-amplitude and range of the variation seen.     Multimodality imaging in vivo for preclinical assessment of tumor-targeted doxorubicin nanoparticles.

    This study presents a new multimodal imaging approach that includes high-frequency ultrasound, fluorescence intensity, confocal, and spectral imaging to improve the preclinical evaluation of new therapeutics in vivo. Here we use this approach to assess in vivo the therapeutic efficacy of the novel chemotherapy construct, HerDox during and after treatment. HerDox is comprised of doxorubicin non-covalently assembled in a viral-like particle targeted to HER2+ tumor cells, causing tumor cell death at over 10-fold lower dose compared to the untargeted drug, while sparing the heart. Whereas our initial proof-of-principle studies on HerDox used tumor growth/shrinkage rates as a measure of therapeutic efficacy, here we show that multimodal imaging deployed during and after treatment can supplement traditional modes of tumor monitoring to further characterize the particle in tissues of treated mice. Specifically, we show here that tumor cell apoptosis elicited by HerDox can be monitored in vivo during treatment using high frequency ultrasound imaging, while in situ confocal imaging of excised tumors shows that HerDox indeed penetrated tumor tissue and can be detected at the subcellular level, including in the nucleus, via Dox fluorescence. In addition, ratiometric spectral imaging of the same tumor tissue enables quantitative discrimination of HerDox fluorescence from autofluorescence in situ. In contrast to standard approaches of preclinical assessment, this new method provides multiple/complementary information that may shorten the time required for initial evaluation of in vivo efficacy, thus potentially reducing the time and cost for translating new drug molecules into the clinic

    Spontaneous emergence of spatial patterns ina a predator-prey model

    We present studies for an individual based model of three interacting populations whose individuals are mobile in a 2D-lattice. We focus on the pattern formation in the spatial distributions of the populations. Also relevant is the relationship between pattern formation and features of the populations' time series. Our model displays travelling waves solutions, clustering and uniform distributions, all related to the parameters values. We also observed that the regeneration rate, the parameter associated to the primary level of trophic chain, the plants, regulated the presence of predators, as well as the type of spatial configuration.Comment: 17 pages and 15 figure

    Origin of complexity in multicellular organisms

    Through extensive studies of dynamical system modeling cellular growth and reproduction, we find evidence that complexity arises in multicellular organisms naturally through evolution. Without any elaborate control mechanism, these systems can exhibit complex pattern formation with spontaneous cell differentiation. Such systems employ a `cooperative' use of resources and maintain a larger growth speed than simple cell systems, which exist in a homogeneous state and behave 'selfishly'. The relevance of the diversity of chemicals and reaction dynamics to the growth of a multicellular organism is demonstrated.     Current Star Formation in the Ophiuchus and Perseus Molecular Clouds: Constraints and Comparisons from Unbiased Submillimeter and Mid-Infrared Surveys. II

    We present a census of the population of deeply embedded young stellar objects (YSOs) in the Ophiuchus molecular cloud complex based on a combination of Spitzer Space Telescope mid-infrared data from the "Cores to Disks" (c2d) legacy team and JCMT/SCUBA submillimeter maps from the COMPLETE team. We have applied a method developed for identifying embedded protostars in Perseus to these datasets and in this way construct a relatively unbiased sample of 27 candidate embedded protostars with envelopes more massive than our sensitivity limit (about 0.1 M_sun). Embedded YSOs are found in 35% of the SCUBA cores - less than in Perseus (58%). On the other hand the mid-infrared sources in Ophiuchus have less red mid-infrared colors, possibly indicating that they are less embedded. We apply a nearest neighbor surface density algorithm to define the substructure in each of the clouds and calculate characteristic numbers for each subregion - including masses, star formation efficiencies, fraction of embedded sources etc. Generally the main clusters in Ophiuchus and Perseus (L1688, NGC1333 and IC348) are found to have higher star formation efficiencies than small groups such as B1, L1455 and L1448, which on the other hand are completely dominated by deeply embedded protostars. We discuss possible explanations for the differences between the regions in Perseus and Ophiuchus, such as different evolutionary timescales for the YSOs or differences, e.g., in the accretion in the two clouds.Comment: Accepted for publication in ApJ (56 pages, 13 figures; abstract abridged).     A description of an algorithm for a rather general modal grating calculation is presented. Arbitrary profiles, depth, and permittivity are allowed. Gratings built up from sub-gratings are allowed, as are coatings on the sidewalls of lines, and arbitrary complex structure. Conical angles and good conductors are supported
