    Experimental and mathematical modelling of grain material velocity

    The present article represents the next stage in the cycle of work on the research of vibrating motion of grain material. This paper contains the experimental results of the operation of a vibrating conveying machine. The investigation was carried out in three stages. The purpose of the first full-factor experiment with four varying factors was to identify factors having the least impact on the time of movement of grain material. As a result, it was found that the angle of vibration direction that has an impact on the amplitude has almost no effect on the velocity of the grain material. In the second phase of experiment carried out with three varying factors at fixed angle of vibration direction, the equation of mathematical model of time of passage of the control area has been obtained. Moreover, on the basis of previous studies, the angle of vibration direction was fixed at the position which has the greatest influence on the vibration amplitude. The third stage of the experiment was to evaluate applicability of the obtained model equation to describe movement of other grain crops with ellipsoidal grain shape. Verification of the obtained model for adequacy using Fisher's criterion showed that with 95% probability the obtained equation can be used for estimation of movement of grain material having ellipsoidal shape on vibrating surface

    Development of a mathematical model of the process of mixing liquid feed in an experimental setup and optimization of design parameters

    The role of agricultural ecology in current conditions is important in the modern world, as it offers new methods and approaches to the implementation of production processes that should reduce the harmful effects on the environment. The paper outlines the prerequisites for the development of universal mechanization tools for the process of mixing dry and liquid components. The essence of the method is to study the motion of a particle with different design parameters, as well as the physical properties of the medium. The mathematical model of particle motion is built on the basis of theoretical mechanics and hydraulics. The calculation scheme of the impeller is performed from several iterations with a step-by-step approach to the simulated mixing process. In this case, differential equations of motion of dry material particles are applied. Based on the solutions of these equations, the particle motion zone within the impeller structure is determined. At the next stage, the calculation of velocities at different angular speeds and viscosity of the liquid medium is carried out. Thus, there is a consistent approach to the description of the process of mixing a particle with a liquid. According to the proposed algorithm, a program has been developed using MathCad V15 that provides the possibility of graphical and numerical control of the calculation results with a change in parameters. It is possible to change the structural and physical parameters of the environment. Examples of calculation and graphical interpretation of particle motion in the working chamber are presented. According to their results, the optimal modes of operation of the setup for the preparation of liquid feed are proposed

    Antibody expressing pea seeds as fodder for prevention of gastrointestinal parasitic infections in chickens

    <p>Abstract</p> <p>Background</p> <p>Coccidiosis caused by protozoans of genus <it>Eimeria </it>is a chicken parasitic disease of great economical importance. Conventional disease control strategies depend on vaccination and prophylactic use of anticoccidial drugs. Alternative solution to prevent and treat coccidiosis could be provided by passive immunization using orally delivered neutralizing antibodies. We investigated the possibility to mitigate the parasitic infection by feeding poultry with antibody expressing transgenic crop seeds.</p> <p>Results</p> <p>Using the phage display antibody library, we generated a panel of anti-<it>Eimeria </it>scFv antibody fragments with high sporozoite-neutralizing activity. These antibodies were expressed either transiently in agrobacteria-infiltrated tobacco leaves or stably in seeds of transgenic pea plants. Comparison of the scFv antibodies purified either from tobacco leaves or from the pea seeds demonstrated no difference in their antigen-binding activity and molecular form compositions. Force-feeding experiments demonstrated that oral delivery of flour prepared from the transgenic pea seeds had higher parasite neutralizing activity <it>in vivo </it>than the purified antibody fragments isolated from tobacco. The pea seed content was found to protect antibodies against degradation by gastrointestinal proteases (>100-fold gain in stability). <it>Ad libitum </it>feeding of chickens demonstrated that the transgenic seeds were well consumed and not shunned. Furthermore, feeding poultry with shred prepared from the antibody expressing pea seeds led to significant mitigation of infection caused both by high and low challenge doses of <it>Eimeria </it>oocysts.</p> <p>Conclusion</p> <p>The results suggest that our strategy offers a general approach to control parasitic infections in production animals using cost-effective antibody expression in crop seeds affordable for the animal health market.</p

    A bispecific diabody directed against prostate-specific membrane antigen and CD3 induces T-cell mediated lysis of prostate cancer cells

    BACKGROUND: Although cancer of the prostate is one of the most commonly diagnosed cancers in men, no curative treatment currently exists after its progression beyond resectable boundaries. Therefore, new agents for targeted treatment strategies are needed. Cross-linking of tumor antigens with T-cell associated antigens by bispecific monoclonal antibodies have been shown to increase antigen-specific cytotoxicity in T-cells. Since the prostate-specific membrane antigen (PSMA) represents an excellent tumor target, immunotherapy with bispecific diabodies could be a promising novel treatment option for prostate cancer. METHODS: A heterodimeric diabody specific for human PSMA and the T-cell antigen CD3 was constructed from the DNA of anti-CD3 and anti-PSMA single chain Fv fragments (scFv). It was expressed in E. coli using a vector containing a bicistronic operon for co-secretion of the hybrid scFv V<sub>H</sub>CD3-V<sub>L</sub>PSMA and V<sub>H</sub>PSMA-V<sub>L</sub>CD3. The resulting PSMAxCD3 diabody was purified from the periplasmic extract by immobilized metal affinity chromatography (IMAC). The binding properties were tested on PSMA-expressing prostate cancer cells and PSMA-negative cell lines as well as on Jurkat cells by flow cytometry. For in vitro functional analysis, a cell viability test (WST) was used. For in vivo evaluation the diabody was applied together with human peripheral blood lymphocytes (PBL) in a C4-2 xenograft-SCID mouse model. RESULTS: By Blue Native gel electrophoresis, it could be shown that the PSMAxCD3 diabody is mainly a tetramer. Specific binding both to CD3-expressing Jurkat cells and PSMA-expressing C4-2 cells was shown by flow cytometry. In vitro, the diabody proved to be a potent agent for retargeting PBL to lyze C4-2 prostate cancer cells. Treatment of SCID mice inoculated with C4-2 tumor xenografts with the diabody and PBL efficiently inhibited tumor growth. CONCLUSIONS: The PSMAxCD3 diabody bears the potential for facilitating immunotherapy of prostate cancer and for the elimination of minimal residual disease

    Development and application of recombinant antibody-based immunoassays to tetraconazole residue analysis in fruit juices

    Tetraconazole is currently used as a fungicide in fruit and vegetables. The aim of this work was the development of immunochemical techniques based on recombinant antibodies for the screening of tetraconazole residues in fruit juices. Recombinant antibodies were produced from a hybridoma cell line secreting a monoclonal antibody specific for tetraconazole and from lymphocytes of mice hyperimmunised with tetraconazole haptens conjugated to bovine serum albumin. From these antibodies, enzyme-linked immunosorbent assays in the conjugate-coated format were developed, which were able to detect tetraconazole standards down to 1 ng/mL. From recovery studies with spiked samples, these immunoassays determined tetraconazole in orange and apple juices with acceptable reproducibility (coefficients of variation below 25%) and recoveries (ranging from 78% to 145%) for a screening technique. The analytical performance of RAb-based immunoassays was fairly similar to that of the MAb-based immunoassays. Due to their simplicity and high sample throughput, the developed recombinant-based immunoassays can be valuable analytical tools for the screening of tetraconazole residues in fruit juices at regulatory levels.This work was funded by Ministerio de EducaciOn y Ciencia (MEC, Spain, Project AGL2002-03266).
