Estrogen-Dependent Expression and Subcellular Localization of the Tight Junction Protein Claudin-4 in HEC-1A Endometrial Cancer Cells
Endometrial cancer is the most common female reproductive cancer in the United States and is associated with deregulated tight junction protein expression. Given the highly estrogen-responsive nature of this tissue, we investigated the effects of estrogen and its agonist, 4-OH TAM, on the expression and subcellular localization of the tight junction protein claudin-4 (CLDN-4), in HEC-1A endometrial cancer cells. In untreated HEC-1A cells, we observed dramatic overexpression of claudin-4 protein. In addition, differential detergent extraction analysis indicated that claudin-4 was localized primarily in the membrane but also found in the cytosolic, nuclear and cytoskeletal fractions. Upon exposure of HEC-1A to estradiol (E2), we observed a biphasic effect both on the overall expression of claudin-4 protein and on its cytosolic and cytoskeletal presence as demonstrated by immunoblot analysis. Immunofluorescence analysis also revealed a biphasic effect of E2 on claudin-4 expression. In contrast, we observed no changes in expression levels nor in the subcellular distribution patterns of claudin-4 in HEC-1A cells treated with different concentrations of 4-OH TAM. The intracellular presence of CLDN-4 coupled with the biphasic effects of E2 on CLDN-4 expression in the cytoskeleton suggest that this protein may be involved in cell signaling to and from TJs

On-Wafer Measurement of a Silicon-Based CMOS VCO at 324 GHz
The world s first silicon-based complementary metal oxide/semiconductor (CMOS) integrated-circuit voltage-controlled oscillator (VCO) operating in a frequency range around 324 GHz has been built and tested. Concomitantly, equipment for measuring the performance of this oscillator has been built and tested. These accomplishments are intermediate steps in a continuing effort to develop low-power-consumption, low-phase-noise, electronically tunable signal generators as local oscillators for heterodyne receivers in submillimeter-wavelength (frequency > 300 GHz) scientific instruments and imaging systems. Submillimeter-wavelength imaging systems are of special interest for military and law-enforcement use because they could, potentially, be used to detect weapons hidden behind clothing and other opaque dielectric materials. In comparison with prior submillimeter- wavelength signal generators, CMOS VCOs offer significant potential advantages, including great reductions in power consumption, mass, size, and complexity. In addition, there is potential for on-chip integration of CMOS VCOs with other CMOS integrated circuitry, including phase-lock loops, analog- to-digital converters, and advanced microprocessors
A Spectroscopic Study of Mass Outflows in the Interacting Binary RY Scuti
The massive interacting binary RY Scuti is an important representative of an
active mass-transferring system that is changing before our eyes and which may
be an example of the formation of a Wolf-Rayet star through tidal stripping.
Utilizing new and previously published spectra, we present examples of how a
number of illustrative absorption and emission features vary during the binary
orbit. We identify spectral features associated with each component, calculate
a new, double-lined spectroscopic binary orbit, and find masses of 7.1 +/- 1.2
M_sun for the bright supergiant and 30.0 +/- 2.1 M_sun for the hidden massive
companion. Through tomographic reconstruction of the component spectra from the
composite spectra, we confirm the O9.7 Ibpe spectral class of the bright
supergiant and discover a B0.5 I spectrum associated with the hidden massive
companion; however, we suggest that the latter is actually the spectrum of the
photosphere of the accretion torus immediately surrounding the massive
companion. We describe the complex nature of the mass loss flows from the
system in the context of recent hydrodynamical models for beta Lyr, leading us
to conclude RY Scuti has matter leaving the system in two ways: 1) a bipolar
outflow from winds generated by the hidden massive companion, and 2) mass from
the bright O9.7 Ibpe supergiant flowing from the region near the L2 point to
fill out a large, dense circumbinary disk. This circumbinary disk (radius ~ 1

Waveguide Transition for Submillimeter-Wave MMICs
An integrated waveguide-to-MMIC (monolithic microwave integrated circuit) chip operating in the 300-GHz range is designed to operate well on high-permittivity semiconductor substrates typical for an MMIC amplifier, and allows a wider MMIC substrate to be used, enabling integration with larger MMICs (power amplifiers). The waveguide-to- CBCPW (conductor-backed coplanar waveguide) transition topology is based on an integrated dipole placed in the E-plane of the waveguide module. It demonstrates low loss and good impedance matching. Measurement and simulation demonstrate that the loss of the transition and waveguide loss is less than 1-dB over a 340-to-380-GHz bandwidth. A transition is inserted along the propagation direction of the waveguide. This transition uses a planar dipole aligned with the maximum E-field of the TE10 waveguide mode as an inter face between the waveguide and the MMIC. Mode conversion between the coplanar striplines (CPS) that feed the dipole and the CBCPW transmission line is accomplished using a simple air-bridge structure. The bottom side ground plane is truncated at the same reference as the top-side ground plane, leaving the end of the MMIC suspended in air
The role of single nucleotide polymorphisms in breast cancer metastasis
Our understanding of many aspects of cancer biology has been advanced through the use of modern genetics. These studies have already shown that germ line polymorphisms play a significant role in disease initiation and response to therapy. However, what is less well studied is the role of germ line polymorphisms in cancer progression. Studies in rodents indicate that differential susceptibility to cancer metastasis can be heritable; thus, the search for the genes that control cancer metastasis is underway. Although some provocative studies suggest potential candidates for metastasis regulating genes, the conclusive identification of a specific inherited genetic variant that alters metastatic potential awaits further studies
HD 98800: A Unique Stellar System of Post-T Tauri Stars
HD 98800 is a system of four stars, and it has a large infrared excess that is thought to be due to a dust disk within the system. In this paper we present new astrometric observations made with Hipparcos, as well as photometry from Hubble Space Telescope WFPC2 images. Combining these observations and reanalyzing previous work allow us to estimate the age and masses of the stars in the system. Uncertainty in these ages and masses results from uncertainty in the temperatures of the stars and any reddening they may have. We find that HD 98800 is most probably about 10 Myr old, although it may be as young as 5 Myr or as old as 20 Myr. The stars in HD 98800 appear to have metallicities that are about solar. An age of 10 Myr means that HD 98800 is a member of the post-T Tauri class of objects, and we argue that the stars in HD 98800 can help us understand why post-T Tauris have been so elusive. HD 98800 may have formed in the Centaurus star-forming region, but it is extraordinary in being so young and yet so far from where it was born
Socioeconomic differences in the benefits of structured physical activity compared with health education on the prevention of major mobility disability in older adults: the LIFE study.
BACKGROUND: Evidence is lacking on whether health-benefiting community-based interventions differ in their effectiveness according to socioeconomic characteristics. We evaluated whether the benefit of a structured physical activity intervention on reducing mobility disability in older adults differs by education or income. METHODS: The Lifestyle Interventions and Independence for Elders (LIFE) study was a multicentre, randomised trial that compared a structured physical activity programme with a health education programme on the incidence of mobility disability among at-risk community-living older adults (aged 70-89â
years; average follow-up of 2.6â
years). Education (†high school (0-12â
years), college (13-17â
years) or postgraduate) and annual household income were self-reported (<25â
000 to 50â
000). The risk of disability (objectively defined as loss of ability to walk 400â
m) was compared between the 2 treatment groups using Cox regression, separately by socioeconomic group. Socioeconomic groupĂintervention interaction terms were tested. RESULTS: The effect of reducing the incidence of mobility disability was larger for those with postgraduate education (0.72, 0.51 to 1.03; N=411) compared with lower education (high school or less (0.93, 0.70 to 1.24; N=536). However, the education groupĂintervention interaction term was not statistically significant (p=0.54). Findings were in the same direction yet less pronounced when household income was used as the socioeconomic indicator. CONCLUSIONS: In the largest and longest running trial of physical activity amongst at-risk older adults, intervention effect sizes were largest among those with higher education or income, yet tests of statistical interactions were non-significant, likely due to inadequate power. On-Wafer S-Parameter Measurements in the 325-508-GHz Band
New circuits have been designed and fabricated with operating frequencies over 325 GHz. In order to measure S-parameters of these circuits, an extensive process of wafer dicing and packaging, and waveguide transition design, fabrication, and packaging would be required. This is a costly and time-consuming process before the circuit can be tested in waveguide. The new probes and calibration procedures will simplify the testing process. New on-wafer probes, and a procedure for their calibration, have been developed that allow fast and inexpensive S-parameter characterization of circuits in the 325 -508 -GHz frequency band. The on-wafer probes transition from rectangular waveguide to coplanar waveguide probe tips with 40- m nominal signal-to-ground pin pitch so as to allow for probing circuits on a wafer. The probes with bias tees have been optimized for minimal insertion loss and maximum return loss when placed on 50-ohm structures to allow for calibration. The calibration process has been developed using the Thru-Reflect-Line Agilent algorithm with JPL determined calibration structures and calibration coefficients for the algorithm. This new test capability is presently unique to JPL. With it, researchers will be able to better develop circuits such as low-noise amplifiers, power amplifiers, multipliers, and mixers for heterodyne receivers in the 325-508-GHz frequency band for remote sensing/spectroscopy
