    Physiological and Biochemical Characterization of Three Nucleoside Diphosphate Kinase Isozymes from Rice (Oryza sativaL.)

    Nucleoside diphosphate kinase (NDPK) is a ubiquitous enzyme that catalyzes the transfer of the γ-phosphoryl group from a nucleoside triphosphate to a nucleoside diphosphate. In this study, we examined the subcellular localization, tissue-specific gene expression, and enzymatic characteristics of three rice NDPK isozymes (OsNDPK1-OsNDPK3). Sequence comparison of the three OsNDPKs suggested differential subcellular localization. Transient expression of green fluorescence protein-fused proteins in onion cells indicated that OsNDPK2 and OsNDPK3 are localized to plastid and mitochondria respectively, while OsNDPK1 is localized to the cytosol. Expression analysis indicated that all the OsNDPKs are expressed in the leaf, leaf sheath, and immature seeds, except for OsNDPK1, in the leaf sheath. Recombinant OsNDPK2 and OsNDPK3 showed lower optimum pH and higher stability under acidic pH than OsNDPK1. In ATP formation, all the OsNDPKs displayed lower K(m) values for the second substrate, ADP, than for the first substrate, NTP, and showed lowest and highest K(m) values for GTP and CTP respectively

    Case Control Study in Diagnosis of Pancreatic Cancer: Using multiple-logistic regression

    We determined 13 markers of pancreatic cancer which are used presently and tried quantitative combination assay of pancreatic cancer. Case-control study using the logistic regression showed the possibility of pancreatic cancer quantitatively. When the level of CA19-9 is more than Mean + 2S.D. (40 u/ml), the possibility of pancreatic cancer is 15 times higher than when the level is less than Mean + S.D .. In the case that the high-level of CA19-9 and α1-AT which are more than Mean + S.D., the possibility of pancreatic cancer is 30 times higher than the case that the level of CA19-9 and α1-AT are less than Mean + S.D. when both levels of CA19-9 and α1-AT are more than Mean + S.D .

    The specifications of washstand for wheelchair were re-examined to help materialize more smooth and comfortableness. The face washing activities in sitting positions were recorded with 3-D motion capture system.The experiments were run for different heights, sizes, and shapes of washstands, and also for different distances between the washstand and the seat. The captured motions were analyzed to identify inconveniences and difficulties during the activities.As the results, the optimum difference of height between the top of washstand and the seat is in the range of 250-300mm. The optimum distance from the front edge of washstand to greater trochanter is around 250mm. It is important to form the front of shape of washstand in order not to hamper the movement of upper extremity. Based on the results, the current height of standard washstand for wheelchair (750mm) should be lowered

    A preliminary proposal of policy for dairy herd health management from the viewpoints of culling in the Hiroshima University Farm

    近年日本の乳牛の平均除籍産次数が3.4産程度に低下しており,対策が必要な現状にある.本研究の目的は,本学農場の乳牛除籍の傾向を把握して,対策について考察することである.2000~2015年度の本農場の記録をもとに解析した.本農場では常時20~25頭程度の搾乳牛が飼養され,2011年3月までは1日2回のパーラー搾乳,それ以降は24時間自動搾乳が行われた.期間内に83頭の乳牛が除籍になり,うち67頭が肉畜として出荷され,16頭が農場内で死亡した.除籍頭数割合は産次とともに増加し,6産時が最も多かった.除籍率は,初産時が最も低く,徐々に増加して4産,5産時に40%を超え,6産時に急激に増加して77.8%となった.除籍理由として,繁殖障害,乳房炎等の疾病が推察された.一般的な繁殖成績改善と疾病予防を進めるとともに,6産以降は繁殖と疾病の履歴に基づき,飼養継続の判断基準確立が必要と考えられた.In Japan, mean value for parity of dairy cows at culling has decreased to 3.4; measures to improve this are required. The aim of the present study is to understand the trends of culling of dairy cows at the Hiroshima University Farm. For that purpose, the farm’s records for dairy cow culling during the period of 2000–2015 were analyzed. At the farm, 20–27 dairy cows are regularly being milked. In March 2011, the milking system at the farm was changed from a milking parlor system to an automatic milking system. During the 16 years analyzed, 67 cows sold for meat and 16 cows that died at the farm totaled to 83 culled cows. The proportion of culled cows for each parity to total culled cows increased with increasing parity, and the number of cows culled at their 6th parity was the highest. The proportion of culled cows to cows that delivered at each parity gradually increased to more than 40% at the 5th parity, and drastically increased to more than 77.8% at the 6th parity. The reasons for culling were reproductive difficulty and diseases, such as mastitis. Therefore, general measures for improving reproductive performance and disease prevention should be taken for the cows at the 4th or lower parity. In addition, for dairy cows at the 5th or higher parity, a diagnostic decision on whether they are artificially inseminated for further delivery or not might be required depending on the individual cow’s record of reproduction and diseases

    he purpose of this study was to evaluate the difference in T2-elongated spots (T2ES) between self-referred and third party-referred subjects.The brain MRI studies of 814 healthy adults were assessed. The subjects were categorized into two groups. Group A included 312 self-referred subjects ranging in age from 49 to 65 years (mean age, 56.5 years). Group B included 502 third party-referred subjects same ranging in age (mean age, 54.3 years). All subjects were asked to complete an interview sheet dealing with current and past diseases. To compare the two groups, an ‘Age-related Grading System\u27 was created.Grade 4 was defined as including patients who had 10 to 14 more T2ESs than their age minus 49; 20.027771275620f Group B and 13.51111400240f Group A (P<0.05) were classified as Grade 4. Diabetes mellitus was present in 15.016010062550f Group A and 9.615734071165f Group B (P<0.05). Hyperlipidemia was present in 18.015710062563f Group A and 9.015035020146f Group B (P<0.01).Although diabetes mellitus and hyperlipidemia were more common in Group A, these diseases were considered to be well controlled. It would appear that the patients in Group A were more health conscious than those in Group B

    Effect of Saxagliptin on Endothelial Function in Patients with Type 2 Diabetes : A Prospective Multicenter Study

    The dipeptidyl peptidase-4 inhibitor saxagliptin is a widely used antihyperglycemic agent in patients with type 2 diabetes. The purpose of this study was to evaluate the effects of saxagliptin on endothelial function in patients with type 2 diabetes. This was a prospective, multicenter, interventional study. A total of 34 patients with type 2 diabetes were enrolled at four university hospitals in Japan. Treatment of patients was initially started with saxagliptin at a dose of 5 mg daily. Assessment of endothelial function assessed by flow-mediated vasodilation (FMD) and measurement of stromal cell-derived factor-1α (SDF-1α) were conducted at baseline and at 3 months after treatment with saxagliptin. A total of 31 patients with type 2 diabetes were included in the analysis. Saxagliptin significantly increased FMD from 3.1 ± 3.1% to 4.2 ± 2.4% (P = 0.032) and significantly decreased total cholesterol from 190 ± 24 mg/dL to 181 ± 25 mg/dL (P = 0.002), glucose from 160 ± 53 mg/dL to 133 ± 25 mg/dL (P < 0.001), HbA1c from 7.5 ± 0.6% to 7.0 ± 0.6% (P < 0.001), urine albumin-to-creatinine ratio from 63.8 ± 134.2 mg/g to 40.9 ± 83.0 mg/g (P = 0.043), and total SDF-1α from 2108 ± 243 pg/mL to 1284 ± 345 pg/mL (P < 0.001). These findings suggest that saxagliptin is effective for improving endothelial function

    Diagnostic Criteria of FMD and NID

    Background - Diagnostic criteria of flow-mediated vasodilation (FMD), an index of endothelial function, and nitroglycerin-induced vasodilation (NID), an index of vascular smooth muscle function, of the brachial artery have not been established. The purpose of this study was to propose diagnostic criteria of FMD and NID for normal endothelial function and normal vascular smooth muscle function. Methods and Results - We investigated the cutoff values of FMD and NID in subjects with (risk group) and those without cardiovascular risk factors or cardiovascular diseases (no-risk group) in 7277 Japanese subjects (mean age 51.4±10.8 years) from the Flow-Mediated Dilation Japan study and the Flow-Mediated Dilatation Japan Registry study for analysis of the cutoff value of FMD and in 1764 Japanese subjects (62.2±16.1 years) from the registry of Hiroshima University Hospital for analysis of the cutoff value of NID. Receiver-operator characteristic curve analysis of FMD to discriminate subjects in the no-risk group from patients in the risk group showed that the optimal cutoff value of FMD to diagnose subjects in the no-risk group was 7.1%. Receiver-operator characteristic curve analysis of NID to discriminate subjects in the no-risk group from patients in the risk group showed that the optimal cutoff value of NID to diagnose subjects in the no-risk group was 15.6%. Conclusions - We propose that the cutoff value for normal endothelial function assessed by FMD of the brachial artery is 7.1% and that the cutoff value for normal vascular smooth muscle function assessed by NID of the brachial artery is 15.6% in Japanese subjects
