    Dunce mutants of Drosophila melanogaster: mutants defective in the cyclic AMP phosphodiesterase enzyme system

    The cyclic AMP and cyclic GMP phosphodiesterase activities present in flies of six mutant strains of the dunce gene and in the parent wild- type strains are characterized. All of the mutants exhibit aberrant cyclic AMP metabolism. The mutant strains dunceM14, dunceM11, and dunceML appear to be amorphic, because they completely lack the cAMP-specific phosphodiesterase normally present in adult flies. These strains exhibit extremely high levels of cAMP. The mutant strains dunce1, dunce2, and dunceCK are hypomorphic and exhibit reduced levels of the cAMP-specific phosphodiesterase. These strains exhibit less marked increases in cAMP content compared with the three amorphic strains. The dunce2 strain possesses a residual enzyme activity that exhibits anomalous kinetics compared with those of the normal enzyme. The possibility that the dunce locus is the structural gene for the cAMP-specific phosphodiesterase is discussed

    Cells rely on the coordinated regulation of lipid phosphoinositides and Rab GTPases to define membrane compartment fates along distinct trafficking routes. The family of disease-related myotubularin (MTM) phosphoinositide phosphatases includes catalytically inactive members, or pseudophosphatases, with poorly understood functions. We found that Drosophila MTM pseudophosphatase Sbf coordinates both phosphatidylinositol 3-phosphate (PI(3)P) turnover and Rab21 GTPase activation in an endosomal pathway that controls macrophage remodeling. Sbf dynamically interacts with class II phosphatidylinositol 3-kinase and stably recruits Mtm to promote turnover of a PI(3)P subpool essential for endosomal trafficking. Sbf also functions as a guanine nucleotide exchange factor that promotes Rab21 GTPase activation associated with PI(3)P endosomes. Of importance, Sbf, Mtm, and Rab21 function together, along with Rab11-mediated endosomal trafficking, to control macrophage protrusion formation. This identifies Sbf as a critical coordinator of PI(3)P and Rab21 regulation, which specifies an endosomal pathway and cortical control

    K band Spectroscopy of Ultraluminous Infrared Galaxies: The 2 Jy Sample

    We present near-infrared spectroscopy for a complete sample of 33 ultraluminous infrared galaxies at a resolution of R\approx 1000. Most of the wavelength range from 1.80-2.20 microns in the rest frame is covered, including the Pa-alpha and Br-gamma hydrogen recombination lines, and the molecular hydrogen vibration-rotation 1-0 S(1) and S(3) lines. Other species, such as He I, [Fe II], and [Si VI] appear in the spectra as well, in addition to a number of weaker molecular hydrogen lines. Nuclear extractions for each of the individual galaxies are presented here, along with spectra of secondary nuclei, where available. The Pa-alpha emission is seen to be highly concentrated on the nuclei, typically with very little emision extending beyond a radius of 1 kpc. Signatures of active nuclei are rare in the present sample, occurring in only two of the 33 galaxies. It is found that visual extinctions to the nuclei via the Pa-alpha/Br-gamma line ratio in excess of 10 magnitudes are relatively common among ULIRGs, and that visual extinctions greater than 25 mag are necessary to conceal a QSO emitting half the total bolometric luminosity. The vibration-rotation lines of molecular hydrogen appear to be predominantly thermal in origin, with effective temperatures generally around 2200 K. The relative nuclear velocities between double nucleus ULIRGs are investigated, through which it is inferred that the maximum deprojected velocity difference is about 200 km/s.     Midwives' competence : is it affected by working in a rural location?

    Introduction: Rising health care costs and the need to consolidate expertise in tertiary services have led to the centralisation of services. In the UK, the result has been that many rural maternity units have become midwife-led. A key consideration is that midwives have the skills to competently and confidently provide maternity services in rural areas, which may be geographically isolated and where the midwife may only see a small number of pregnant women each year. Our objective was to compare the views of midwives in rural and urban settings, regarding their competence and confidence with respect to ‘competencies’ identified as being those which all professionals should have in order to provide effective and safe care for low-risk women. Method: This was a comparative questionnaire survey involving a stratified sample of remote and rural maternity units and an ad hoc comparison group of three urban maternity units in Scotland. Questionnaires were sent to 82 midwives working in remote and rural areas and 107 midwives working in urban hospitals with midwife-led units. Results: The response rate from midwives in rural settings was considerably higher (85%) than from midwives in the urban areas (60%). Although the proportion of midwives who reported that they were competent was broadly similar in the two groups, there were some significant differences regarding specific competencies. Midwives in the rural group were more likely to report competence for breech delivery (p = 0.001), while more urban midwives reported competence in skills such as intravenous fluid replacement (p <0.001) and initial and discharge examination of the newborn (p <0.001). Both groups reported facing barriers to continuing professional development; however, more of the rural group had attended an educational event within the last month (p <0.001). Lack of time was a greater barrier for urban midwives (p = 0.02), whereas distance to training was greater for rural midwives (p = 0.009). Lack of motivation or interest was significantly higher in urban units (p = 0.006). Conclusion: It is often assumed that midwives in rural areas where there are fewer deliveries, will be less competent and confident in their practice.     Fabrication and transport critical currents of multifilamentary MgB2/Fe wires and tapes

    Multifilamentary MgB2/Fe wires and tapes with high transport critical current densities have been fabricated using a straightforward powder-in-tube (PIT) process. After annealing, we measured transport jc values up to 1.1 * 105 A/cm2 at 4.2 K and in a field of 2 T in a MgB2/Fe square wire with 7 filaments fabricated by two-axial rolling, and up to 5 * 104 A/cm2 at 4.2 K in 1 T in a MgB2/Fe tape with 7 filaments. For higher currents these multifilamentary wires and tapes quenched due to insufficient thermal stability of filaments. Both the processing routes and deformation methods were found to be important factors for fabricating multifilamentary MgB2 wires and tapes with high transport jc values.Comment: 13 pages, 7 figure

    Multi‐scale heterogeneity in vegetation and soil carbon in exurban residential land of southeastern Michigan, USA

    Exurban residential land (one housing unit per 0.2–16.2 ha) is growing in importance as a human‐dominated land use. Carbon storage in the soils and vegetation of exurban land is poorly known, as are the effects on C storage of choices made by developers and residents. We studied C storage in exurban yards in southeastern Michigan, USA, across a range of parcel sizes and different types of neighborhoods. We divided each residential parcel into ecological zones (EZ) characterized by vegetation, soil, and human behavior such as mowing, irrigation, and raking. We found a heterogeneous mixture of trees and shrubs, turfgrasses, mulched gardens, old‐field vegetation, and impervious surfaces. The most extensive zone type was turfgrass with sparse woody vegetation (mean 26% of parcel area), followed by dense woody vegetation (mean 21% of parcel area). Areas of turfgrass with sparse woody vegetation had trees in larger size classes (> 50 cm dbh) than did areas of dense woody vegetation. Using aerial photointerpretation, we scaled up C storage to neighborhoods. Varying C storage by neighborhood type resulted from differences in impervious area (8–26% of parcel area) and area of dense woody vegetation (11–28%). Averaged and multiplied across areas in differing neighborhood types, exurban residential land contained 5240 ± 865 g C/m2 in vegetation, highly sensitive to large trees, and 13 800 ± 1290 g C/m2 in soils (based on a combined sampling and modeling approach). These contents are greater than for agricultural land in the region, but lower than for mature forest stands. Compared with mature forests, exurban land contained more shrubs and less downed woody debris and it had similar tree size‐class distributions up to 40 cm dbh but far fewer trees in larger size classes. If the trees continue to grow, exurban residential land could sequester additional C for decades. Patterns and processes of C storage in exurban residential land were driven by land management practices that affect soil and vegetation, reflecting the choices of designers, developers, and residents.
