    Chiroptical signal enhancement in quasi-null-polarization-detection geometry: Intrinsic limitations

    Despite its unique capability of distinguishing molecular handedness, chiroptical spectroscopy suffers from the weak-signal problem, which has restricted more extensive applications. The quasi-null-polarization-detection (QNPD) method has been shown to be useful for enhancing the chiroptical signal. Here, the underlying enhancement mechanism in the QNPD method combined with a heterodyne detection scheme is elucidated. It is experimentally demonstrated that the optical rotatory dispersion signal can be amplified by a factor of similar to 400, which is the maximum enhancement effect achievable with our femtosecond laser setup. The upper limit of the QNPD enhancement effect of chiroptical measurements could, in practice, be limited by imperfection of the polarizer and finite detection sensitivity. However, we show that there exists an intrinsic limit in the enhancement with the QNPD method due to the weak but finite contribution from the homodyne chiroptical signal. This is experimentally verified by measuring the optical rotation of linearly polarized light with the QNPD scheme. We further provide discussions on the connection between this intrinsic limitation in the QNPD scheme for enhanced detection of weak chiroptical signals and those in optical enantioselectivity and Raman optical activity with a structured chiral field.     Isolation and characterization of a new mucoid-free Klebsiella pneumoniae strain for 2,3-butanediol production

    The secretion of mucoid substances by Klebsiella pneumoniae, a natural 2,3-butanediol (2,3-BD) hyper-producer, hinders its application in large-scale fermentation because of pathogenicity, fermentation instability, and downstream difficulty. In this study, 14 K. pneumoniae strains were isolated from a waste water treatment plant and their 2,3-BD production efficiencies were assessed with the strain K. pneumoniae DSM2026. Among various strains isolated, K. pneumoniae GSC010 and GSC112 produced relatively large amounts of 2,3-BD compared to other isolates; and their 2,3-BD production was consistent with DSM2026. Meanwhile, mucoidic characteristics of GSC010 were more or less similar to DSM2026, which was observed by scanning electron microscope (SEM) as a characteristic intercalated thread anchored on the surface of the cells. However, no polysaccharide materials were found in a non-mucoid cell, GSC112. Fed-batch culture of GSC112 with continuous glucose feeding resulted in the production of 2,3-BD at 52.4 g/l with 2,3-BD yield and overall productivity of 0.27 g/g glucose and 0.52 g/l/h, respectively. These results strongly suggest that the newly isolated mucoid-free K. pneumoniae GSC112 has potential for industrial production of 2,3-BD.Keywords: 2,3-Butanediol, Klebsiella pneumoniae, isolation, capsular polysaccharides, scanning electron microscop

    Guideline Development for the Evaluation of Visual Impairment in Korea

    This guideline is developed to provide criteria for evaluating permanent impairment of the visual system as it affects an individual`s ability to perform activities of daily living. This new assessment system is based on the 5th and 6th edition of American Medical Association and McBride impairment assessment system but revised on the consideration of Korean culture and simple application. This evaluation of impairment is based on an assessment of visual acuity and visual field. Especially it weighs binocular vision and binocular visual fields and the binocular vision and binocular visual fields provide 50% of weight and the right and left eye each contribute 25%. A further adjustment of the impairment rating is included at the final step of this evaluation. Functional deficits for individual adjustments include diplopia, problem of accommodation, abnormality of eyelids, tearing, cosmetic problems from cornea opacity, glare, aphakia, and dark-adaptation. The adjustment can be added to impairment rating up to 15%.     Complete Binocular Blindness as the First Manifestation of HIV-Related Cryptococcal Meningitis

    Ocular complications of HIV-related cryptococcal meningitis are reasonably common, but complete binocular blindness as the first manifestation of HIV is extremely rare. A 58-year-old man presented with binocular blindness. He experienced blurred vision for 3 days before the blindness. Mild pleocytosis was present in the cerebrospinal fluid, from which Cryptococcus neoformans was cultured. Serology revealed positivity for HIV antibody. He was treated with antifungal and antiretroviral therapy. This case indicates that HIV-related cryptococcal meningitis should be taken into consideration when determining the cause of unexpected sudden binocular blindness

    Pattern of Hepatitis A Incidence According to Area Characteristics Using National Health Insurance Data

    Objectives: Over the past several years, the incidence of hepatitis A infection has been increasing rapidly in the young-adult population in Korea. We examined the effects of area-level socioeconomic status and environmental hygiene on the incidence of hepatitis A. Methods: This study is based on the registered national population of Korea and the national health insurance data from 2004 to 2008. A total of 73 459 individuals were confirmed to have had hepatitis A. The standardized incidences of hepatitis A in 232 districts adjusted for sex and age of people were calculated for each year, and the rate ratios of the incidence rates were estimated according to area-level socioeconomic status and environmental hygiene using multiple Poisson regression models. Results: The incidence rates of hepatitis A infection were 15.6 (per 100 000) in 2004, 19.0 (per 100 000) in 2005, 27.2 (per 100 000) in 2006, 25.1 (per 100 000) in 2007, and 61.7 (per 100 000) in 2008. The analysis of the area-level effects showed that residential areas of the less deprived than other regions, areas with higher levels of education, and heavily populated areas were significantly associated with increased risk. Conclusions: There is a very strong possibility that both area-level socioeconomic status and environmental hygiene play a role in increasing the risk of hepatitis A infection in Korea. Therefore, to reduce hepatitis A infection, we need a nationwide strategy that considers these area-level characteristics

    Oral intake of Lactobacillus rhamnosus M21 enhances the survival rate of mice lethally infected with influenza virus

    BackgroundInfluenza viruses cause acute respiratory disease. Because of the high genetic variability of viruses, effective vaccines and antiviral agents are limited. Considering the fact that the site of influenza virus entry is the mucosa of the upper respiratory tract, probiotics that can enhance mucosal immunity as well as systemic immunity could be an important source of treatment against influenza infection.MethodsMice were fed with Lactobacillus rhamnosus M21 or skim milk and were challenged with influenza virus. The resulting survival rate, lung inflammation, and changes in the cytokine and secretory immunoglobulin A (sIgA) levels were examined.ResultsBecause of infection (influenza virus), all the mice in the control group and 60% of the mice in the L. rhamnosus M21 group died; however, the remaining 40% of the mice fed with L. rhamnosus M21 survived the infection. Pneumonia was severe in the control group but moderate in the group treated with L. rhamnosus M21. Although there were no significant changes in the proinflammatory cytokines in the lung lysates of mice collected from both groups, levels of interferon-γ and interleukin-2, which are representative cytokines of type I helper T cells, were significantly increased in the L. rhamnosus M21-treated group. An increase in sIgA as well as the diminution of inflammatory cells in bronchoalveolar lavage fluid was also observed in the L. rhamnosus M21-treated group.ConclusionThese results demonstrate that orally administered L. rhamnosus M21 activates humoral as well as cellular immune responses, conferring increased resistance to the host against influenza virus infection

    High frequencies of Y-chromosome haplogroup O2b-SRY465 lineages in Korea: a genetic perspective on the peopling of Korea

    <p>Abstract</p> <p>Background</p> <p>Koreans are generally considered a Northeast Asian group, thought to be related to Altaic-language-speaking populations. However, recent findings have indicated that the peopling of Korea might have been more complex, involving dual origins from both southern and northern parts of East Asia. To understand the male lineage history of Korea, more data from informative genetic markers from Korea and its surrounding regions are necessary. In this study, 25 Y-chromosome single nucleotide polymorphism markers and 17 Y-chromosome short tandem repeat (Y-STR) loci were genotyped in 1,108 males from several populations in East Asia.</p> <p>Results</p> <p>In general, we found East Asian populations to be characterized by male haplogroup homogeneity, showing major Y-chromosomal expansions of haplogroup O-M175 lineages. Interestingly, a high frequency (31.4%) of haplogroup O2b-SRY465 (and its sublineage) is characteristic of male Koreans, whereas the haplogroup distribution elsewhere in East Asian populations is patchy. The ages of the haplogroup O2b-SRY465 lineages (~9,900 years) and the pattern of variation within the lineages suggested an ancient origin in a nearby part of northeastern Asia, followed by an expansion in the vicinity of the Korean Peninsula. In addition, the coalescence time (~4,400 years) for the age of haplogroup O2b1-47z, and its Y-STR diversity, suggest that this lineage probably originated in Korea. Further studies with sufficiently large sample sizes to cover the vast East Asian region and using genomewide genotyping should provide further insights.</p> <p>Conclusions</p> <p>These findings are consistent with linguistic, archaeological and historical evidence, which suggest that the direct ancestors of Koreans were proto-Koreans who inhabited the northeastern region of China and the Korean Peninsula during the Neolithic (8,000-1,000 BC) and Bronze (1,500-400 BC) Ages.</p

    Safety and Effectiveness of the Novel Catheter 3.0 System for Diagnostic Cerebral Angiography: A Pilot Study

    Purpose The purpose of this study was to evaluate the safety and effectiveness of a new angiographic system (Catheter 3.0 system) using a 5 French (Fr), large-bore angiography catheter, a 0.032-inch stiff guidewire, and a continuous flushing system in diagnostic cerebral angiography. Materials and Methods This retrospective study included 30 consecutive patients who underwent transfemoral cerebral angiography using the Catheter 3.0 system from October 2019 to March 2020. As the control group, we included 30 consecutive patients examined before the Catheter 3.0 system was introduced. Procedural outcomes, including technical success, procedure time, dose metrics, procedure-related complications, and image quality were reviewed and analyzed. Results All transfemoral cerebral angiographies were performed for a diagnosis of unruptured intracranial aneurysms. The Catheter 3.0 system showed a significantly shorter fluoroscopy time (6.2 vs. 9.7 minutes, P=0.008) and lower fluoroscopy dose (387.2 vs. 614.4, P=0.002) compared with the conventional 4-Fr catheter system. The Catheter 3.0 system also showed better results in terms of procedural time (21.0 vs. 22.5 minutes, P=0.072) and technical success rate (98.1% vs. 94.0%, P=0.078), although a statistical significance was not reached. The complication rate and qualitative assessment of the digital subtraction angiography (DSA) image quality were similar between the two groups. Conclusion The Catheter 3.0 system using a 5 Fr catheter with a large inner diameter was convenient, effective, and safe compared with the conventional system in diagnostic cerebrovascular angiography
