    On the toughness of thermoplastic polymer nanocomposites as assessed by the essential work of fracture (EWF) approach

    The essential work of fracture (EWF) approach is widely used to determine the plane stress fracture toughness of highly ductile polymers and related systems. To shed light on how the toughness is affected by nanofillers EWF-suited model polymers, viz. amorphous copolyester and polypropylene block copolymer were modified by multiwall carbon nanotube (MWCNT), graphene (GR), boehmite alumina (BA), and organoclay (MMT) in 1 wt% each. EWF tests were performed on deeply double-edge notched tensile-loaded specimens under quasistatic loading conditions. Data reduction occurred by energy partitioning between yielding and necking/tearing. The EWF prerequisites were not met with the nanocomposites containing MWCNT and GR by contrast to those with MMT and BA. Accordingly, the toughness of nanocomposites with homogeneously dispersed and low aspect ratio fillers may be properly determined using the EWF. Results indicated that incorporation of nanofillers may result in an adverse effect between the specific essential and non-essential EWF parameters

    Knockdown of LRP/LR Induces Apoptosis in breast and oesophageal cancer cells.

    Cancer is a global burden due to high incidence and mortality rates and is ranked the second most diagnosed disease amongst non-communicable diseases in South Africa. A high expression level of the 37kDa/67kDa laminin receptor (LRP/LR) is one characteristic of cancer cells. This receptor is implicated in the pathogenesis of cancer cells by supporting tumor angiogenesis, metastasis and especially for this study, the evasion of apoptosis. In the current study, the role of LRP/LR on cellular viability of breast MCF-7, MDA-MB 231 and WHCO1 oesophageal cancer cells was investigated. Western blot analysis revealed that total LRP expression levels of MCF-7, MDA-MB 231 and WHCO1 were significantly downregulated by targeting LRP mRNA using siRNA-LAMR1. This knockdown of LRP/LR resulted in a significant decrease of viability in the breast and oesophageal cancer cells as determined by an MTT assay. Transfection of MDA-MB 231 cells with esiRNA-RPSA directed against a different region of the LRP mRNA had similar effects on LRP/LR expression and cell viability compared to siRNA-LAMR1, excluding an off-target effect of siRNALAMR1. This reduction in cellular viability is as a consequence of apoptosis induction as indicated by the exposure of the phosphatidylserine protein on the surface of breast MCF-7, MDA-MB 231 and oesophageal WHCO1 cancer cells, respectively, detected by an Annexin-V/FITC assay as well as nuclear morphological changes observed post-staining with Hoechst. These observations indicate that LRP/LR is crucial for the maintenance of cellular viability of breast and oesophageal cancer cells and recommend siRNA technology targeting LRP expression as a possible novel alternative technique for breast and oesophageal cancer treatment.This work was supported by the National Research Foundation, the Republic of South Africa, the Republic of South Africa.NCS201

    Investigating hair zinc concentrations in children with and without atopic dermatitis

    Background. Atopic dermatitis (AD) is a common chronic inflammatory skin condition that disproportionately affects children and is associated with reduced quality of life. Zinc deficiency may contribute to the pathogenesis of AD because zinc plays a role in epidermal barrier integrity and the immune system. Systematic review evidence suggests that low zinc is associated with AD, but limitations of included studies support further investigation.Objectives. To investigate hair zinc concentrations in children with AD v. healthy controls in a low- to middle-income country setting.Methods. One hundred and five children aged 1 - 12 yea­rs participated in a frequency-matched for age case-control study. The outcome variable, AD, was confirmed by a clinician and corroborated using the UK Working Party criteria. The primary predictor, long-term average zinc concentration, was determined by measuring hair zinc using inductively coupled mass spectrometry. Baseline demographic characteristics, anthropometry and measures of socioeconomic status were included in our logistic regression analysis. Subgroup analysis was performed where interaction terms suggested effect modification.Results. Using data from the overall sample, population median hair zinc was not significantly different between children with AD and healthy controls. However, subgroup analysis suggested a clinically and statistically significant difference in median zinc between children with AD (175.35 µg/g) and healthy controls (206.4 µg/g) in the older age group (5 - 12 years) (p=0.01). In this age group, multivariable logistic regression analysis also found significantly decreased hair zinc concentrations in AD (odds ratio 0.83; 95% confidence interval 0.66 - 0.96; p=0.046).Conclusions. The inverse association between zinc status and AD in children aged 5 - 12 years in our setting is consistent with the international literature. The clinical importance of decreased zinc levels in AD is not yet known. Further investigation into relevant underlying mechanisms seems warranted given the global reach of AD, its effect on quality of life, and the low cost of potential zinc-based interventions.

    Anti-LRP/LR specific antibody IgG1-iS18 impedes adhesion and invasion of liver cancer cells.

    Two key events, namely adhesion and invasion, are pivotal to the occurrence of metastasis. Importantly, the 37 kDa/67 kDa laminin receptor (LRP/LR) has been implicated in enhancing these two events thus facilitating cancer progression. In the current study, the role of LRP/LR in the adhesion and invasion of liver cancer (HUH-7) and leukaemia (K562) cells was investigated. Flow cytometry revealed that the HUH-7 cells displayed significantly higher cell surface LRP/LR levels compared to the poorly-invasive breast cancer (MCF-7) control cells, whilst the K562 cells displayed significantly lower cell surface LRP/LR levels in comparison to the MCF-7 control cells. However, Western blotting and densitometric analysis revealed that all three tumorigenic cell lines did not differ significantly with regards to total LRP/LR levels. Furthermore, treatment of liver cancer cells with anti-LRP/LR specific antibody IgG1-iS18 (0.2 mg/ml) significantly reduced the adhesive potential of cells to laminin-1 and the invasive potential of cells through the ECM-like Matrigel, whilst leukaemia cells showed no significant differences in both instances. Additionally, Pearson's correlation coefficients suggested direct proportionality between cell surface LRP/LR levels and the adhesive and invasive potential of liver cancer and leukaemia cells. These findings suggest the potential use of anti-LRP/LR specific antibody IgG1-iS18 as an alternative therapeutic tool for metastatic liver cancer through impediment of the LRP/LR- laminin-1 interaction.National Research Foundation.NCS201

    Countrywide roll-out of Xpert(®) MTB/RIF in Swaziland: the first three years of implementation.

    SETTING: All 19 public health laboratories in Swaziland that had Xpert(®) MTB/RIF machines installed as part of a countrywide roll-out between June 2011 and June 2014. OBJECTIVE: To evaluate the utilisation and functionality of Xpert from 2011 to mid-2014. DESIGN: Descriptive study of Xpert implementation using routinely collected data. RESULTS: Of 48 829 Xpert tests conducted, 93% were successful: 14% detected Mycobacterium tuberculosis and 12% showed rifampicin resistance. The most common cause of unsuccessful tests was an 'Error' result (62%). Similar findings were obtained in government-supported and partner-supported laboratories. Annual utilisation of Xpert improved from 51% of maximum capacity in 2011 and 2012 to 74% in 2013 and 2014. A monitoring and supervision exercise of all Xpert testing sites in 2014 showed a generally good performance, with over 50% of laboratories achieving a ⩾80% score on most components. However, poor scores were obtained with equipment use and maintenance (6% achieving a score of ⩾80%), internal audit (19% achieving a score of ⩾80%) and process control (25% achieving a score of ⩾80%). CONCLUSION: Countrywide roll-out of Xpert in Swaziland has been successful, although operational issues have been identified and need to be resolved

    Gamma ray production cross sections in proton induced reactions on natural Mg, Si and Fe targets over the proton energy range 30 up to 66 MeV

    Gamma-ray excitation functions have been measured for 30, 42, 54 and 66 MeV proton beams accelerated onto C + O (Mylar), Mg, Si, and Fe targets of astrophysical interest at the separate-sector cyclotron of iThemba LABS in Somerset West (Cape Town, South Africa). A large solid angle, high energy resolution detection system of the Eurogam type was used to record Gamma-ray energy spectra. Derived preliminary results of Gamma-ray line production cross sections for the Mg, Si and Fe target nuclei are reported and discussed. The current cross section data for known, intense Gamma-ray lines from these nuclei consistently extend to higher proton energies previous experimental data measured up to Ep ~ 25 MeV at the Orsay and Washington tandem accelerators. Data for new Gamma-ray lines observed for the first time in this work are also reported.Comment: 11 pages, 6 figures.
