    New Method for Obtaining a Culture of Peritoneal Mast Cells

    Актуальным является разработка методики получения культуры животных клеток, которую впоследствии можно будет использовать в качестве экспериментальной модели для проведения как фундаментальных исследований, так и доклинических испытаний лекарственных средств. Таковой является и разработка методики выделения линии перитонеальных тучных клеток из перитонеальной жидкости и ее культивирования в специализированных условиях. Полученная культура является подходящей экспериментальной моделью для проведения фундаментальных исследований тучных клеток – изучения их свойств и их взаимодействия с различными биологически активными веществами и другими клетками в со-культуре in vitro, и для проведения доклинических исследований специализированных лекарственных препаратов на ней.Currently, the development of a method for obtaining a culture of animal cells, which can later be used as an experimental model for conducting both basic research and preclinical trials of drugs, is urgent. The development of a technique for isolating a line of peritoneal mast cells from the peritoneal fluid and its cultivation in specialized conditions is also urgent.     Regulation of Tcf7l1 DNA Binding and Protein Stability as Principal Mechanisms of Wnt/β-Catenin Signaling

    SummaryWnt/β-catenin signal transduction requires direct binding of β-catenin to Tcf/Lef proteins, an event that is classically associated with stimulating transcription by recruiting coactivators. This molecular cascade plays critical roles throughout embryonic development and normal postnatal life by affecting stem cell characteristics and tumor formation. Here, we show that this pathway utilizes a fundamentally different mechanism to regulate Tcf7l1 (formerly named Tcf3) activity. β-catenin inactivates Tcf7l1 without a switch to a coactivator complex by removing it from DNA, which leads to Tcf7l1 protein degradation. Mouse genetic experiments demonstrate that Tcf7l1 inactivation is the only required effect of the Tcf7l1-β-catenin interaction. Given the expression of Tcf7l1 in pluripotent embryonic and adult stem cells, as well as in poorly differentiated breast cancer, these findings provide mechanistic insights into the regulation of pluripotency and the role of Wnt/β-catenin in breast cancer

    Comparison of UNet, ENet, and BoxENet for Segmentation of Mast Cells in Scans of Histological Slices

    Deep neural networks show high accuracy in the problem of semantic and instance segmentation of biomedical data. However, this approach is computationally expensive. The computational cost may be reduced with network simplification after training or choosing the proper architecture, which provides segmentation with less accuracy but does it much faster. In the present study, we analyzed the accuracy and performance of UNet and ENet architectures for the problem of semantic image segmentation. In addition, we investigated the ENet architecture by replacing of some convolution layers with box-convolution layers. The analysis performed on the original dataset consisted of histology slices with mast cells. These cells provide a region for segmentation with different types of borders, which vary from clearly visible to ragged. ENet was less accurate than UNet by only about 1-2%, but ENet performance was 8-15 times faster than UNet one. © 2019 IEEE

    Spin chemistry investigation of peculiarities of photoinduced electron transfer in donor-acceptor linked system

    Photoinduced intramolecular electron transfer in linked systems, (R,S)- and (S,S)-naproxen-N-methylpyrrolidine dyads, has been studied by means of spin chemistry methods [magnetic field effect and chemically induced dynamic nuclear polarization (CIDNP)]. The relative yield of the triplet state of the dyads in different magnetic field has been measured, and dependences of the high-field CIDNP of the N-methylpyrrolidine fragment on solvent polarity have been investigated. However, both (S,S)- and (R,S)-enantiomers demonstrate almost identical CIDNP effects for the entire range of polarity.
