    Detection of amblyopia utilizing generated retinal reflexes

    Investigation confirmed that GRR images can be consistently obtained and that these images contain information required to detect the optical inequality of one eye compared to the fellow eye. Digital analyses, electro-optical analyses, and trained observers were used to evaluate the GRR images. Two and three dimensional plots were made from the digital analyses results. These plotted data greatly enhanced the GRR image content, and it was possible for nontrained observers to correctly identify normal vs abnormal ocular status by viewing the plots. Based upon the criteria of detecting equality or inequality of ocular status of a person's eyes, the trained observer correctly identified the ocular status of 90% of the 232 persons who participated in this program

    A genetic assessment of parentage in the blackspot sergeant damselfish, Abudefduf sordidus (Pisces: Pomacentridae)

    Microsatellite markers were used to investigate the reproductive behavior of the damselfish Abudefduf sordidus at Johnston Atoll, Central Pacific Ocean. Genetic results indicated that ten males maintained guardianship over their nest territories for up to nine nest cycles during a 3.5 month period. Genotypes of 1025 offspring sampled from 68 nests (composed of 129 clutches) were consistent with 95% of the offspring being sired by the guardian male. Offspring lacking paternal alleles at two or more loci were found in 19 clutches, indicating that reproductive parasitism and subsequent alloparental care occurred. Reconstructed maternal genotypes allowed the identification of a minimum of 74 different females that spawned with these ten territorial males. Males were polygynous, mating with multiple females within and between cycles. Genetic data from nests, which consisted of up to four clutches during a reproductive cycle, indicated that each clutch usually had only one maternal contributor and that different clutches each had different dams. Females displayed sequential polyandry spawning with one male within a cycle but switched males in subsequent spawning cycles. These results highlight new findings regarding male parasitic spawning, polygyny, and sequential polyandry in a marine fish with exclusive male paternal care.Published versio

    Spectral Energy Distributions for Disk and Halo M--Dwarfs

    We have obtained infrared (1 to 2.5 micron) spectroscopy for 42 halo and disk dwarfs with spectral type M1 to M6.5. These data are compared to synthetic spectra generated by the latest model atmospheres of Allard & Hauschildt. Photospheric parameters metallicity, effective temperature and radius are determined for the sample. We find good agreement between observation and theory except for known problems due to incomplete molecular data for metal hydrides and water. The metal-poor M subdwarfs are well matched by the models as oxide opacity sources are less important in this case. The derived effective temperatures for the sample range from 3600K to 2600K; at these temperatures grain formation and extinction are not significant in the photosphere. The derived metallicities range from solar to one-tenth solar.     In vitro and in vivo studies of the trypanocidal properties of WRR-483 against Trypanosoma cruzi.

    BackgroundCruzain, the major cysteine protease of Trypanosoma cruzi, is an essential enzyme for the parasite life cycle and has been validated as a viable target to treat Chagas' disease. As a proof-of-concept, K11777, a potent inhibitor of cruzain, was found to effectively eliminate T. cruzi infection and is currently a clinical candidate for treatment of Chagas' disease.Methodology/principal findingsWRR-483, an analog of K11777, was synthesized and evaluated as an inhibitor of cruzain and against T. cruzi proliferation in cell culture. This compound demonstrates good potency against cruzain with sensitivity to pH conditions and high efficacy in the cell culture assay. Furthermore, WRR-483 also eradicates parasite infection in a mouse model of acute Chagas' disease. To determine the atomic-level details of the inhibitor interacting with cruzain, a 1.5 A crystal structure of the protease in complex with WRR-483 was solved. The structure illustrates that WRR-483 binds covalently to the active site cysteine of the protease in a similar manner as other vinyl sulfone-based inhibitors. Details of the critical interactions within the specificity binding pocket are also reported.ConclusionsWe demonstrate that WRR-483 is an effective cysteine protease inhibitor with trypanocidal activity in cell culture and animal model with comparable efficacy to K11777. Crystallographic evidence confirms that the mode of action is by targeting the active site of cruzain. Taken together, these results suggest that WRR-483 has potential to be developed as a treatment for Chagas' disease

    Astrophysical limitations to the identification of dark matter: indirect neutrino signals vis-a-vis direct detection recoil rates

    A convincing identification of dark matter (DM) particles can probably be achieved only through a combined analysis of different detections strategies, which provides an effective way of removing degeneracies in the parameter space of DM models. In practice, however, this program is made complicated by the fact that different strategies depend on different physical quantities, or on the same quantities but in a different way, making the treatment of systematic errors rather tricky. We discuss here the uncertainties on the recoil rate in direct detection experiments and on the muon rate induced by neutrinos from dark matter annihilations in the Sun, and we show that, contrarily to the local DM density or overall cross section scale, irreducible astrophysical uncertainties affect the two rates in a different fashion, therefore limiting our ability to reconstruct the parameters of the dark matter particle. By varying within their respective errors astrophysical parameters such as the escape velocity and the velocity dispersion of dark matter particles, we show that the uncertainty on the relative strength of the neutrino and direct-detection signal is as large as a factor of two for typical values of the parameters, but can be even larger in some circumstances.Comment: 12 pages, 3 figures.     It is shown using three series of Rayleigh number simulations of varying aspect ratio AR and Prandtl number Pr that the normalized dissipation at the wall, while significantly greater than 1, approaches a constant dependent upon AR and Pr. It is also found that the peak velocity, not the mean square velocity, obeys the experimental scaling of Ra^{0.5}. The scaling of the mean square velocity is closer to Ra^{0.46}, which is shown to be consistent with experimental measurements and the numerical results for the scaling of Nu and the temperature if there are strong correlations between the velocity and temperature.Comment: 5 pages, 3 figures, new version 13 Mar, 200
