    The Backup Planning Scale (BUPS): A Brief, Self-Reported Measure of a Person's Tendency to Develop, Reserve, and Use Backup Plans

    To mitigate uncertainty in their goal pursuits, people use backup plans, i.e., alternative means that are developed to potentially replace "Plan A." Several studies have demonstrated that backup plans can introduce unexpected costs into goal pursuits that decrease a person's motivation to continue using their "Plan A," and reduce their chances for achieving their goal. These existing studies used time-intensive experimental and/or observational approaches to assess the effects of backup planning. The present research examines the newly-developed Backup Planning Scale (BUPS) for its measurement invariance, reliability, validity, and other psychometric characteristics across three independent samples with more than 1,500 participants. Consistent with prior theorizing, we found support for a nine-item, three factor structure for the BUPS, indexing latent factors for a person's tendency to develop, reserve, and replace with (or use) backup plans. Furthermore, a novel "IRTree" based statistical technique provided evidence for the validity of the measure, as participants' responses to the BUPS were associated with their actual developing, reserving, and replacing backup planning behaviors in a logic task. We conclude that the freely-available BUPS is a simple, brief, reliable, and valid self-reported instrument for assessing backup planning behaviors across adulthood

    Deinococcus glutaminyl-tRNA synthetase is a chimer between proteins from an ancient and the modern pathways of aminoacyl-tRNA formation

    Glutaminyl-tRNA synthetase from Deinococcus radiodurans possesses a C-terminal extension of 215 residues appending the anticodon-binding domain. This domain constitutes a paralog of the Yqey protein present in various organisms and part of it is present in the C-terminal end of the GatB subunit of GatCAB, a partner of the indirect pathway of Gln-tRNA(Gln) formation. To analyze the peculiarities of the structure–function relationship of this GlnRS related to the Yqey domain, a structure of the protein was solved from crystals diffracting at 2.3 Å and a docking model of the synthetase complexed to tRNA(Gln) constructed. The comparison of the modeled complex with the structure of the E. coli complex reveals that all residues of E. coli GlnRS contacting tRNA(Gln) are conserved in D. radiodurans GlnRS, leaving the functional role of the Yqey domain puzzling. Kinetic investigations and tRNA-binding experiments of full length and Yqey-truncated GlnRSs reveal that the Yqey domain is involved in tRNA(Gln) recognition. They demonstrate that Yqey plays the role of an affinity-enhancer of GlnRS for tRNA(Gln) acting only in cis. However, the presence of Yqey in free state in organisms lacking GlnRS, suggests that this domain may exert additional cellular functions

    Hierarchy of adhesion forces in patterns of photoreactive surface layers

    Precise control of surface properties including electrical characteristics, wettability, and friction is a prerequisite for manufacturing modern organic electronic devices. The successful combination of bottom up approaches for aligning and orienting the molecules and top down techniques to structure the substrate on the nano and micrometer scale allows the cost efficient fabrication and integration of future organic light emitting diodes and organic thin film transistors. One possibility for the top down patterning of a surface is to utilize different surface free energies or wetting properties of a functional group. Here, we used friction force microscopy (FFM) to reveal chemical patterns inscribed by a photolithographic process into a photosensitive surface layer. FFM allowed the simultaneous visualization of at least three different chemical surface terminations.     Recently forming stalagmites from the Baradla Cave and their suitability assessment for climate–proxy relationships

    Determination of the long-term behavior of cave systems and their response to changing environmental conditions is essential for further paleoclimate analyses of cave-hosted carbonate deposits. For this purpose, four actively forming stalagmites were collected in the Baradla Cave where a three-year monitoring campaign was also conducted. Based on textural characteristics and radiocarbon analyses, the stalagmites are composed of annual laminae, whose counting was used to establish age–depth relationships. Fast and slowly growing stalagmites have different stable carbon and oxygen isotope compositions as well as trace element contents that could be attributed to differences in drip water migration pathways. The stable isotope compositions were compared with meteorological data of the last ∼100 years indicating that carbon isotope compositions of the stalagmites may reflect changes in precipitation amount, while oxygen isotope compositions are more related to temperature variations. The combined textural–geochemical–meteorological interpretation lead us to select the isotope record that can best reflect variations in environmental conditions and can be used for further evaluation of the climate–proxy relationships

    Fusiform vateritic inclusions observed in European eel (Anguilla anguilla L.) sagittae

    Microscopic inclusions have been observed in 7 out of 106 European eel (Anguilla anguilla L.) sagittae using polarizing microscope and scanning electron microscope meanwhile the annual increments were studied to characterize the age structure of the population living in Lake Balaton. The presence of vaterite, a rare calcium carbonate polymorph was observed in these inclusions using Raman spectroscopy. Vateritic sagittae in wild fish are usually considered as symptom of physiological stress. The observed fusiform inclusions represent a new morphological type of vaterite inclusions in eel otolith. Two alternatives are hypothesized to explain their formation: 1) metabolic disorder, such as erroneous protein synthesis; 2) introduction of an alien protein into the eel’s inner ear. The origin and physiological significance of this new morphological type of vateritic inclusions is still an open question. Same as whether it can be found in other species or specific only to eel otoliths

    Regional Differences in Dietary Habits of Adult Croatian Population

    The aim of this study was to investigate the dietary habits of the adult Croatian population according to geographical regions and gender. Date was obtained from the Croatian Adult Health Survey questionnaire, carried out in six regions of Republic of Croatia (Eastern, Northern, Central, City of Zagreb, Mountainous and Coastal), with a total of 9,070 responses. Results have shown that the highest prevalence of unhealthy dietary habits was recorded in Eastern and Central regions, while Coastal region and city of Zagreb had significantly lower prevalence of unhealthy dietary habits (P<0.05). Prevalence of unhealthy dietary habits was higher in men in all regions. Women had healthier dietary habits, what was strongly expressed in Coastal and Mountainous region. This study has shown that the unhealthy dietary habits were practiced by approximately one quarter of the adult population, regardless on the region in which they resided. Regional differences in dietary habits should be taken into account in any new public health studies and interventions

    Enhanced Activity of Meprin-α, a Pro-Migratory and Pro-Angiogenic Protease, in Colorectal Cancer

    Meprin-α is a metalloprotease overexpressed in cancer cells, leading to the accumulation of this protease in a subset of colorectal tumors. The impact of increased meprin-α levels on tumor progression is not known. We investigated the effect of this protease on cell migration and angiogenesis in vitro and studied the expression of meprin-α mRNA, protein and proteolytic activity in primary tumors at progressive stages and in liver metastases of patients with colorectal cancer, as well as inhibitory activity towards meprin-α in sera of cancer patient as compared to healthy controls. We found that the hepatocyte growth factor (HGF)- induced migratory response of meprin-transfected epithelial cells was increased compared to wild-type cells in the presence of plasminogen, and that the angiogenic response in organ-cultured rat aortic explants was enhanced in the presence of exogenous human meprin-α. In patients, meprin-α mRNA was expressed in colonic adenomas, primary tumors UICC (International Union Against Cancer) stage I, II, III and IV, as well as in liver metastases. In contrast, the corresponding protein accumulated only in primary tumors and liver metastases, but not in adenomas. However, liver metastases lacked meprin-α activity despite increased expression of the corresponding protein, which correlated with inefficient zymogen activation. Sera from cancer patients exhibited reduced meprin-α inhibition compared to healthy controls. In conclusion, meprin-α activity is regulated differently in primary tumors and metastases, leading to high proteolytic activity in primary tumors and low activity in liver metastases. By virtue of its pro-migratory and pro-angiogenic activity, meprin-α may promote tumor progression in colorectal cancer
