Identification of New, Translatable ProtectomiRs against Myocardial Ischemia/Reperfusion Injury and Oxidative Stress: The Role of MMP/Biglycan Signaling Pathways
Introduction: Ischemic conditionings (ICon) were intensively investigated and several protective signaling pathways were identified. Previously, we have shown the role of matrix metalloproteinases (MMP) in myocardial ischemia/reperfusion injury (MIRI) and the cardioprotective role of biglycan (BGN), a small leucine-rich proteoglycan in vitro. Here, we hypothesized that cardiac MMP and BGN signaling are involved in the protective effects of ICon. Methods: A reverse target-microRNA prediction was performed by using the miRNAtarget™ 2.0 software to identify human microRNAs with a possible regulatory effect on MMP and BGN, such as on related genes. To validate the identified 1289 miRNAs in the predicted network, we compared them to two cardioprotective miRNA omics datasets derived from pig and rat models of MIRI in the presence of ICons. Results: Among the experimentally measured miRNAs, we found 100% sequence identity to human predicted regulatory miRNAs in the case of 37 porcine and 24 rat miRNAs. Upon further analysis, 42 miRNAs were identified as MIRI-associated miRNAs, from which 24 miRNAs were counter-regulated due to ICons. Conclusions: Our findings highlight 24 miRNAs that potentially regulate cardioprotective therapeutic targets associated with MMPs and BGN in a highly translatable porcine model of acute myocardial infarction
A nehézségi erőtér regionális szerkezetének kutatása új típusú földi és szatellita mérések bevonásával = Regional gravity field research by using new kinds of terrestrial and satellite measurements
A vizsgálatainkat a magyarországi geoidmeghatározás, a geoidfelület interpoláció, továbbá a GOCE gradiométeres műhold jövőbeni méréseinek felhasználása főbb területein végeztük. Elvégeztük a kombinált geoidmegoldáshoz felhasználható adatok gyűjtését és rendszerezését, adatbázisba vitelét és az adatok vizsgálatát. Megfelelő számítási eljárásokat dolgoztunk ki az adatok integrált felhasználása céljából, különös tekintettel a GOCE méréseire. A projekt fő eredményeként egy új, HGTUB2007 jelű geoidmegoldást készítettünk el Magyarország területére geopotenciál modell, gravimetriai, GPS/szintezési, függővonal elhajlási, Eötvös-inga és DTM adatok kombinálásával. A végső megoldásban gyakorlati okokból nagy súllyal szerepeltek az Országos GPS Hálózat (OGPSH) 95 szintezett pontjának mérései. A számított geoidmegoldás az OGPSH kiválasztott pontjaira az elvégzett összehasonlítások szerint 2 cm-es szórással és 6 cm-es maximális eltéréssel illeszkedik. Az illeszkedés azért fontos, mert a geoid magyarországi felületdarabja pontos ismeretének kulcsszerepe van a gyakorlati életben a GPS-szel történő magasságmeghatározásban. Ez a meghatározás világviszonylatban is egyedülálló, mivel ilyen jellegű az adatokat, és különösen az Eötvös-inga mérési adatokat most első alkalommal sikerült az egész ország területére egy egységes feldolgozási folyamat keretében összekapcsolni a még pontosabb regionális erőtér modellezés céljából. | Our research was carried out in the main fields of gravity field determination for Hungary, interpolation of the geoid surface and application of gradiometric data of the future GOCE satellite mission. First the task of gravity data collection, data management and testing was done with the intention to carry out a new combination geoid solution. Suitable methods were developed for integrated data processing especially considering GOCE measurements. The main output of the project is a new Hungarian geoid solution HGTUB2007, achieved by combining geopotential, gravimetric, GPS/levelling, vertical deflection, torsion balance and DTM data. For practical reasons th 95 GPS/levelling data of the Hungarian GPS Network (OGPSH) have been assigned large weights. Our tests showed a fit of 2 cm in terms of standard deviation and 6 cm maximum deviation for selected OGPSH points. This is important in practice, since accurate knowledge of the geoid surface is the key to GPS heighting. This determination is unique in several respects, since by now no geoid solution has been achieved, especially for which surface gravity gradients have successfully been combined with other gravity field quantities for accurate regional gravity field determination
A citokeratin-18 sejthalálmarker vizsgálata sikeres cardiopulmonalis resuscitatión átesett betegpopulációban = The prognostic value of cytokeratin-18 cell death marker in cardiac arrest survivors
Absztrakt:
Bevezetés: Citokeratin-18 (CK-18) az újraélesztés kapcsán
kialakuló ischaemiás-reperfúziós károsodás kiváltotta teljes sejthalál során
kerül a véráramba. Kaszpázok által hasított formája specifikus az apoptózis
folyamatára. A markerek számos kórképben prognosztikus értékűnek bizonyultak.
Tanulmányunkban elsőként vizsgáltuk prognosztikus értéküket reanimált
betegpopulációban. Módszer: 40, sikeresen újraélesztett
betegnél határoztuk meg a sejthalálmarkerek szintjét 6 órán belül, 24 és 72 óra
múlva. Ezeket összevetettük a 30 napos túléléssel, a neurológiai kimenetellel, a
szervfunkciós károsodást jellemző laboratóriumi, fizikális és terápiás
jellemzőkkel, valamint a reanimáció körülményeivel. Eredmények:
A reanimált betegek CK-18-plazmakoncentrációja a szakirodalomban leírt
egészséges, posztoperatív és szeptikus populáció értékeinek a többszöröse volt
(3842 vs. 242; 559; 1644 ng/l); a hasított és intakt CK-18 aránya alacsonyabb
volt (0,14 vs. 0,58; 0,22; 0,24), ami jelentős sejtkárosodásra és a nekrózis
dominanciájára utal. A markerek szintje azonban nem mutatott összefüggést a
túléléssel, a neurológiai statusszal és a reanimáció körülményeivel sem.
Veseelégtelenség esetén a CK-18 szintjének csökkenése elmaradt. Szignifikáns
negatív korrelációt figyeltünk meg a 6 órás hemoglobin- és CK-18-szint között (r
= –0,400, p<0,01), a 30 napos túlélésnek mégis az alacsonyabb
hemoglobinértékek kedveztek. Következtetés: Várakozásunkkal
ellentétben a vizsgált markerek nem bírtak prognosztikus értékkel újraélesztett
betegpopulációban. A kimenetelt valószínűleg nem a teljes sejtkárosodás, hanem
egy kisebb, a fenti markerekkel szenzitíven nem vizsgálható kritikus szerepű
sejtpopuláció károsodása, valamint a beteg tartalékkapacitásai befolyásolják.

Abstract:
Introduction: Cytokeratin-18 (CK-18) is releasing into the blood
during systemic cell death due to ischemia-reperfusion injury after cardiac
arrest. Its caspase-cleaved form is specific to apoptosis. Previous
investigations proved their prognostic value in different conditions. We firstly
investigated the prognostic value of these markers after cardiac arrest.
Method: Plasma samples of 40 resuscitated patients were
collected 6, 24, and 72 hours after successful resuscitation to determine the
marker concentrations. We investigated the association of the markers with the
30-day mortality, neurological outcome, circumstances of the cardiac arrest,
laboratory and physical parameters. Results: Resuscitated
patients had highly elevated CK-18 levels (3842 vs. 242; 559; 1644 ng/L) and
decreased caspase-cleaved CK-18/CK-18 ratio (0.14 vs. 0.58; 0.22; 0.24) compared
to healthy subjects, septic and postoperative patients suggesting severe grade
of cell death, mainly necrosis. Neither the marker concentrations nor their
kinetics showed difference between survivors and non-survivors. They did not
show association with the length of the resuscitation, the initial rhythm or the
neurological outcome either. CK-18 decreased in patients with good renal
function in contrast to patients with renal failure. Significant negative
correlation was observed between the 6-hour cytokeratin-18 and hemoglobin
concentrations (r = –0.400, p<0.01), while the 30-day survival was associated
with lower hemoglobin levels. Conclusion: Surprisingly the
biomarkers did not show prognostic value among resuscitated population. The
outcome is probably not determined by the complete cell damage, but the loss of
a small group of cells with critical role and the reserve capacity of the

Chronic treatment with rofecoxib but not ischemic preconditioning of the myocardium ameliorates early intestinal damage following cardiac ischemia/reperfusion injury in rats
There is some recent evidence that cardiac ischemia/reperfusion (I/R) injury induces intestinal damage within days, which contributes to adverse cardiovascular outcomes after myocardial infarction. However, it is not clear whether remote gut injury has any detectable early signs, and whether different interventions aiming to reduce cardiac damage are also effective at protecting the intestine. Previously, we found that chronic treatment with rofecoxib, a selective inhibitor of cyclooxygenase-2 (COX-2), limited myocardial infarct size to a comparable extent as cardiac ischemic preconditioning (IPC) in rats subjected to 30-min coronary artery occlusion and 120-min reperfusion. In the present study, we aimed to analyse the early intestinal alterations caused by cardiac I/R injury, with or without the above-mentioned infart size-limiting interventions. We found that cardiac I/R injury induced histological changes in the small intestine within 2 h, which were accompanied by elevated tissue level of COX-2 and showed positive correlation with the activity of matrix metalloproteinase-2 (MMP-2), but not of MMP-9 in the plasma. All these changes were prevented by rofecoxib treatment. By contrast, cardiac IPC failed to reduce intestinal injury and plasma MMP-2 activity, although it prevented the transient reduction in jejunal blood flow in response to cardiac I/R. Our results demonstrate for the first time that rapid development of intestinal damage follows cardiac I/R, and that two similarly effective infarct size-limiting interventions, rofecoxib treatment and cardiac IPC, have different impacts on cardiac I/R-induced gut injury. Furthermore, intestinal damage correlates with plasma MMP-2 activity, which may be a biomarker for its early diagnosis
