    Temporal variation in the genetic composition of an endangered marsupial reflects reintroduction history

    The loss of genetic variation and genetic divergence from source populations are common problems for reintroductions that use captive animals or a small number of founders to establish a new population. This study evaluated the genetic changes occurring in a captive and a reintroduced population of the dibbler (Parantechinus apicalis) that were established from multiple source populations over a twelve-year period, using 21 microsatellite loci. While the levels of genetic variation within the captive and reintroduced populations were relatively stable, and did not differ significantly from the source populations, their effective population size reduced 10–16-fold over the duration of this study. Evidence of some loss of genetic variation in the reintroduced population coincided with genetic bottlenecks that occurred after the population had become established. Detectable changes in the genetic composition of both captive and reintroduced populations were associated with the origins of the individuals introduced to the population. We show that interbreeding between individuals from different source populations lowered the genetic relatedness among the offspring, but this was short-lived. Our study highlights the importance of sourcing founders from multiple locations in conservation breeding programs to avoid inbreeding and maximize allelic diversity. The manipulation of genetic composition in a captive or reintroduced population is possible with careful management of the origins and timings of founder releases

    Phytoplankton and microbial plankton of the Northeast Atlantic Shelf

    The Northeast Atlantic Shelf region includes the sites from all coastal waters of Ireland, the Irish Sea, and western Scottish and Norwegian Sea waters. The region was defined by WGPME to include locations on the northern margin of Europe that were outside the North Sea/English Channel influence. The character of sites in the region are shallow, coastal-water sites ranging from sheltered bays on the south coast of Ireland and fjordic sea lochs of Scotland to fully exposed locations on the west coasts of Ireland and Scotland. Bathymetry of the region ranges from shallow embayments to regions of shallow, exposed continental-shelf waters. The topography of the shelf drops rapidly to 80–100 m within 20 km of the coast, where it extends to the shelf edge as a relatively flat plateau. Time-series of phytoplankton data from the Atlantic Shelf exhibit a typical seasonal pattern of temperate waters, with considerable geographical and temporal variation. The well-mixed winter conditions lead to a region-wide strong spring bloom observed at all sites. The ensuing decrease in nutrient levels lead to a variable summer period characterized by stratified conditions in coastal areas and periodic blooms of mixed or occasionally monospecific diatom and dinoflagellate composition. The growth period tails off in autumn, when a secondary bloom may occur in response to increased mixing and breakdown of the summer thermocline. The seasonal cycle returns to a quiescent winter phase, with generally mixed conditions, light limitation, and increased nutrients return. Seasonal stabilization and destabilization of the water column in this region accounts for most of the natural variation in both phytoplankton species composition and biomass

    Oxalate-Induced Damage to Renal Tubular Cells

    Our own studies and those of others have shown that the incidence of calcium oxalate stones and plaques is markedly increased by nephrotoxins. The possible role of oxalate as a nephrotoxin has not been fully appreciated. However, recent studies in experimental animals and in cultured cells support this possibility. The results of these studies led us to hypothesize that hyperoxaluria promotes stone formation in several ways: by providing a substrate for the formation of the most common form of renal stones, calcium oxalate stones, and by inducing damage to renal epithelial cells. Damaged cells in turn would produce an environment favorable for crystal retention and provide membranous debris that promotes crystal nucleation, aggregation and adherence. The present report summarizes evidence for oxalate nephrotoxicity and discusses the potential importance of oxalate toxicity in the pathogenesis of stone disease

    Population genetic structure associated with a landscape barrier in the Western Grasswren (Amytornis textilis textilis)

    Dispersal patterns can dictate genetic population structure and, ultimately, population resilience, through maintaining gene flow and genetic diversity. However, geographical landforms, such as peninsulas, can impact dispersal patterns and thus be a barrier to gene flow. Here, we use 13 375 genome-wide single-nucleotide polymorphisms (SNPs) to evaluate genetic population structure and infer dispersal patterns of the Western Grasswren Amytornis textilis textilis (WGW, n = 140) in the Shark Bay region of Western Australia. We found high levels of genetic divergence between subpopulations on the mainland (Hamelin) and narrow peninsula (Peron). In addition, we found evidence of further genetic sub-structuring within the Hamelin subpopulation, with individuals collected from the western and eastern regions of a conservation reserve forming separate genetic clusters. Spatial autocorrelation analysis within each subpopulation revealed significant local-scale genetic structure up to 35 km at Hamelin and 20 km at Peron. In addition, there was evidence of male philopatry in both subpopulations. Our results suggest a narrow strip of land may be acting as a geographical barrier in the WGW, limiting dispersal between a peninsula and mainland subpopulation. In addition, heterogeneous habitat within Hamelin may be restricting dispersal at the local scale. Furthermore, there is evidence to suggest that the limited gene flow is asymmetrical, with directional dispersal occurring from the bounded peninsula subpopulation to the mainland. This study highlights the genetic structure existing within and between some of the few remaining WGW subpopulations, and shows a need to place equal importance on conservation efforts to maintain them in the future

    A wide array of new N-acyl thiazinanethiones are employed in a number of direct and enantioselective carbon-carbon bond forming reactions catalyzed by nickel(II) complexes. The electrophilic species are mostly prepared in situ from ortho esters, methyl ethers, acetals, and ketals, which makes the overall process highly efficient and experimentally straightforward. Theoretical calculations indicate that the reactions pro-ceed through an open transition state in a SN1-like mechanism. The utility of this novel procedure has been demonstrated by the asymmetric preparation of syntheti-cally useful intermediates and the total synthesis of pe-peromin D

    Perspectives of patients and healthcare professionals on mHealth for asthma self-management

    Copyright ©ERS 2017. Mobile healthcare (mHealth) has the potential to revolutionise the self-management of long-term medical conditions such as asthma. A user-centred design is integral if mHealth is to be embraced by patients and healthcare professionals. The aim of this study was to determine the perspectives of individuals with asthma and healthcare professionals on the use of mHealth for asthma self-management. We used a sequential exploratory mixed methods design; focus groups informed the development of questionnaires, which were disseminated to individuals with asthma and healthcare professionals. Focus group participants (18 asthma patients and five healthcare professionals) identified 12 potential uses of mHealth. Questionnaire results showed that individuals with asthma (n=186) most frequently requested an mHealth system to monitor asthma over time (72%) and to collect data to present to healthcare teams (70%). In contrast, healthcare professionals (n=63) most frequently selected a system alerting patients to deteriorating asthma control (86%) and advising them when to seek medical attention (87%). Individuals with asthma were less likely than healthcare professionals (p < 0.001) to believe that assessing medication adherence and inhaler technique could improve asthma control. Our data provide strong support for mHealth for asthma self-management, but highlight fundamental differences between the perspectives of patients and healthcare professionals

    Allele-Selective Suppression of Mutant Huntingtin in Primary Human Blood Cells

    Post-transcriptional gene silencing is a promising therapy for the monogenic, autosomal dominant, Huntington\u27s disease (HD). However, wild-type huntingtin (HTT) has important cellular functions, so the ideal strategy would selectively lower mutant HTT while sparing wild-type. HD patients were genotyped for heterozygosity at three SNP sites, before phasing each SNP allele to wild-type or mutant HTT. Primary ex vivo myeloid cells were isolated from heterozygous patients and transfected with SNP-targeted siRNA, using glucan particles taken up by phagocytosis. Highly selective mRNA knockdown was achieved when targeting each allele of rs362331 in exon 50 of the HTT transcript; this selectivity was also present on protein studies. However, similar selectivity was not observed when targeting rs362273 or rs362307. Furthermore, HD myeloid cells are hyper-reactive compared to control. Allele-selective suppression of either wild-type or mutant HTT produced a significant, equivalent reduction in the cytokine response of HD myeloid cells to LPS, suggesting that wild-type HTT has a novel immune function. We demonstrate a sequential therapeutic process comprising genotyping and mutant HTT-linkage of SNPs, followed by personalised allele-selective suppression in a small patient cohort. We further show that allele-selectivity in ex vivo patient cells is highly SNP-dependent, with implications for clinical trial target selection

    A Model for Solving the Optimal Water Allocation Problem in River Basins with Network Flow Programming When Introducing Non-Linearities

    [EN] The allocation of water resources between different users is a traditional problem in many river basins. The objective is to obtain the optimal resource distribution and the associated circulating flows through the system. Network flow programming is a common technique for solving this problem. This optimisation procedure has been used many times for developing applications for concrete water systems, as well as for developing complete decision support systems. As long as many aspects of a river basin are not purely linear, the study of non-linearities will also be of great importance in water resources systems optimisation. This paper presents a generalised model for solving the optimal allocation of water resources in schemes where the objectives are minimising the demand deficits, complying with the required flows in the river and storing water in reservoirs. Evaporation from reservoirs and returns from demands are considered, and an iterative methodology is followed to solve these two non-network constraints. The model was applied to the Duero River basin (Spain). Three different network flow algorithms (Out-of-Kilter, RELAX-IVand NETFLO) were used to solve the allocation problem. Certain convergence issues were detected during the iterative process.
