    Limitations in Predicting the Space Radiation Health Risk for Exploration Astronauts

    Despite years of research, understanding of the space radiation environment and the risk it poses to long-duration astronauts remains limited. There is a disparity between research results and observed empirical effects seen in human astronaut crews, likely due to the numerous factors that limit terrestrial simulation of the complex space environment and extrapolation of human clinical consequences from varied animal models. Given the intended future of human spaceflight, with efforts now to rapidly expand capabilities for human missions to the moon and Mars, there is a pressing need to improve upon the understanding of the space radiation risk, predict likely clinical outcomes of interplanetary radiation exposure, and develop appropriate and effective mitigation strategies for future missions. To achieve this goal, the space radiation and aerospace community must recognize the historical limitations of radiation research and how such limitations could be addressed in future research endeavors. We have sought to highlight the numerous factors that limit understanding of the risk of space radiation for human crews and to identify ways in which these limitations could be addressed for improved understanding and appropriate risk posture regarding future human spaceflight.Comment: Accepted for publication by Nature Microgravity (2018

    The LSI and MBTI as Predictors of Learning Style

    The benefits of incorporating learning style theories in the educational process are well-documented. The problem facing educators is the choice of assessment tools that provide useful and significant information. The purpose of this study was to compare Kolb’s Learning Style Inventory-1985 (LSI-85) and the Myers- Briggs Type Indicator-Form G (MBIT-G) to identify existence, strength and direction of correlations. Data were collected from 132 nursing and physical therapy students. Results show some correlations between the two instruments. However, the strength of the correlations is weak and not in predicted directions. Overall, the MBTI-G appears superior to the LSI-85 for assessing learning styles in the classroom

    Developing and validating a theoretical measure of modifiable influences on hormonal therapy medication taking behaviour in women with breast cancer

    Objective: Taking adjuvant hormonal therapy for 5-10 years is recommended to prevent breast cancer recurrence in those with estrogen positive disease. Despite proven clinical efficacy many women do not take their hormonal therapy as prescribed. This study reports the development and initial validation of a questionnaire measuring the behavioural determinants of hormonal therapy medication taking behaviour (MTB) based on the Theoretical Domains Framework (TDF).  Design: Women with Stage I-III breast cancer (N=223) completed the questionnaire based on the TDF. The TDF is an integrative framework consisting of 14 domains of behaviour change determinants to inform intervention design.  Main outcome measures: Items were developed from previous research, in-depth patient interviews and consultation with health professionals. Confirmatory factor analysis (CFA) was undertaken to generate the model of best fit.  Results: The final questionnaire consisted of 8 domains and CFA produced a reasonable fit (χ2(810)=942, p < 0.001; RMSEA = 0.03 ; CFI = 0.93 and WRMR=0.91) as well as internal consistency (r=0.16 to 0.64). There were adequate levels of discriminant validity for the majority of the domains.  Conclusions: A TDF based measure of the behavioural determinants of MTB was developed. Further research is needed to confirm the reliability and validity of this measure

    Primary Corrosion Processes for Polymer-Embedded Free-Standing or Substrate-Supported Silicon Microwire Arrays in Aqueous Alkaline Electrolytes

    Solar fuel devices have shown promise as a sustainable source of chemical fuels. However, long-term stability of light absorbing materials remains a substantial barrier to practical devices. Herein, multiple corrosion pathways in 1 M KOH(aq) have been defined for TiOâ‚‚-protected Si microwire arrays in a polymer membrane either attached to a substrate or free-standing. Top-down corrosion was observed in both morphologies through defects in the TiOâ‚‚ coating. For the substrate-based samples, bottom-up corrosion was observed through the substrate and up the adjacent wires. In the free-standing samples, uniform bottom-up corrosion was observed through the membrane with all wire material corroded within 10 days of immersion in the dark in 1 M KOH(aq)

    Nail abnormalities identified in an ageing study of 30 inbred mouse strains.

    In a large-scale ageing study, 30 inbred mouse strains were systematically screened for histologic evidence of lesions in all organ systems. Ten strains were diagnosed with similar nail abnormalities. The highest frequency was noted in NON/ShiLtJ mice. Lesions identified fell into two main categories: acute to chronic penetration of the third phalangeal bone through the hyponychium with associated inflammation and bone remodelling or metaplasia of the nail matrix and nail bed associated with severe orthokeratotic hyperkeratosis replacing the nail plate. Penetration of the distal phalanx through the hyponychium appeared to be the initiating feature resulting in nail abnormalities. The accompanying acute to subacute inflammatory response was associated with osteolysis of the distal phalanx. Evaluation of young NON/ShiLtJ mice revealed that these lesions were not often found, or affected only one digit. The only other nail unit abnormality identified was sporadic subungual epidermoid inclusion cysts which closely resembled similar lesions in human patients. These abnormalities, being age-related developments, may have contributed to weight loss due to impacts upon feeding and should be a consideration for future research due to the potential to interact with other experimental factors in ageing studies using the affected strains of mice.American Hair Research Society Mentorship Grants (to SL and AM) Ellison Medical Foundation (to JPS) National Institutes of Health (AG025707, for the Shock Aging Center).     <p>Abstract</p> <p>Background</p> <p>As part of a National Institute of Allergy and Infectious Diseases funded collaborative project, we have performed over 150 microarray experiments measuring the response of C57/BL6 mouse bone marrow macrophages to toll-like receptor stimuli. These microarray expression profiles are available freely from our project web site <url>http://www.innateImmunity-systemsbiology.org</url>. Here, we report the development of a database of computationally predicted transcription factor binding sites and related genomic features for a set of over 2000 murine immune genes of interest. Our database, which includes microarray co-expression clusters and a host of web-based query, analysis and visualization facilities, is available freely via the internet. It provides a broad resource to the research community, and a stepping stone towards the delineation of the network of transcriptional regulatory interactions underlying the integrated response of macrophages to pathogens.</p> <p>Description</p> <p>We constructed a database indexed on genes and annotations of the immediate surrounding genomic regions. To facilitate both gene-specific and systems biology oriented research, our database provides the means to analyze individual genes or an entire genomic locus. Although our focus to-date has been on mammalian toll-like receptor signaling pathways, our database structure is not limited to this subject, and is intended to be broadly applicable to immunology. By focusing on selected immune-active genes, we were able to perform computationally intensive expression and sequence analyses that would currently be prohibitive if applied to the entire genome. Using six complementary computational algorithms and methodologies, we identified transcription factor binding sites based on the Position Weight Matrices available in TRANSFAC. For one example transcription factor (ATF3) for which experimental data is available, over 50% of our predicted binding sites coincide with genome-wide chromatin immnuopreciptation (ChIP-chip) results. Our database can be interrogated via a web interface. Genomic annotations and binding site predictions can be automatically viewed with a customized version of the Argo genome browser.</p> <p>Conclusion</p> <p>We present the Innate Immune Database (IIDB) as a community resource for immunologists interested in gene regulatory systems underlying innate responses to pathogens.
