    v. ill. 23 cm.QuarterlyThe morphology of Eleotris acanthopoma collected from Moorea in French Polynesia is described. This is the first record of this species from French Polynesia, greatly expanding the known range, which was previously only considered to extend from southern Japan to New Caledonia. Nucleotide sequences of the mitochondrial ND5 gene of several Eleotris species and related genera indicate that E. acanthopoma from Moorea belongs to the same lineage as E. acanthopoma from Japan and the Philippines. Despite being separated by a distance of approximately 10,000 km, two of the specimens from Moorea and one from the Philippines had identical nucleotide sequences. Results of this study indicate that extensive dispersal occurs during the pelagic larval stage of this species

    Modeling and Flight Data Analysis of Spacecraft Dynamics with a Large Solar Array Paddle

    The Advanced Land Observing Satellite (ALOS) was launched on January 24 2006 and has been operated successfully since then. This satellite has the attitude dynamics characterized by three large flexible structures, four large moving components, and stringent attitude/pointing stability requirements. In particular, it has one of the largest solar array paddles. Presented in this paper are flight data analyses and modeling of spacecraft attitude motion induced by the large solar array paddle. On orbit attitude dynamics was first characterized and summarized. These characteristic motions associated with the solar array paddle were identified and assessed. These motions are thermally induced motion, the pitch excitation by the paddle drive, and the role excitation. The thermally induced motion and the pitch excitation by the paddle drive were modeled and simulated to verify the mechanics of the motions. The control law updates implemented to mitigate the attitude vibrations are also reported

    Ischemia Alters the Expression of Connexins in the Aged Human Brain

    Although the function of astrocytic gap junctions under ischemia is still under debate, increased expression of connexin 43 (Cx43) has been observed in ischemic brain lesions, suggesting that astrocytic gap junctions could provide neuronal protection against ischemic insult. Moreover, different connexin subtypes may play different roles in pathological conditions. We used immunohistochemical analysis to investigate alterations in the expression of connexin subtypes in human stroke brains. Seven samples, sectioned after brain embolic stroke, were used for the analysis. Data, evaluated semiquantitatively by computer-assisted densitometry, was compared between the intact hemisphere and ischemic lesions. The results showed that the coexpression of Cx32 and Cx45 with neuronal markers was significantly increased in ischemic lesions. Cx43 expression was significantly increased in the colocalization with astrocytes and relatively increased in the colocalization with neuronal marker in ischemic lesions. Therefore, Cx32, Cx43, and Cx45 may respond differently to ischemic insult in terms of neuroprotection

    First record of Larsonella pumilus (Teleostei: Gobiidae) from Japan, with phylogenetic placement of the genus Larsonella

    During a survey of deep-sea fauna, using a Remotely Operated Vehicle, a single specimen (21.6 mm in standard length) of Larsonella pumilus (Larson & Hoese, 1980) was collected at a depth of 214 m off the coast of Okinawa Island, Japan. It represents the first record of this species from Japan. The collection site was far deeper than previous reports for this species. This suggests that the main habitat of L. pumilus is deeper than previously recognized and it may explain the paucity of records of this species. As the previously available morphological description of L. pumilus was based on only a single specimen (holotype), this new specimen is described herein. Its morphology corresponds closely to the original description of the holotype, except that faint melanophores are arranged radially around the eyes and scattered on the trunk and the fins. Mitochondrial genome sequences of L. pumilus and 19 related species demonstrate close relationships between L. pumilus and the genus Priolepis. These data also indicate that the genus Priolepis is not monophyletic.journal articl

    Suzaku Detection of Extended/Diffuse Hard X-Ray Emission from the Galactic Center

    Five on-plane regions within +/- 0.8deg of the Galactic center were observed with the Hard X-ray Detector (HXD) and the X-ray Imaging Spectrometer (XIS) onboard Suzaku. From all regions, significant hard X-ray emission was detected with HXD-PIN up to 40 keV, in addition to the extended plasma emission which is dominant in the XIS band. The hard X-ray signals are inferred to come primarily from a spatially extended source, rather than from a small number of bright discrete objects. Contributions to the HXD data from catalogued X-ray sources, typically brighter than 1 mCrab, were estimated and removed using information from Suzaku and other satellites. Even after this removal, the hard X-ray signals remained significant, exhibiting a typical 12--40 keV surface brightness of 4E-10 erg cm-2 s-1 deg-2 and power-law-like spectra with a photon index of 1.8. Combined fittings to the XIS and HXD-PIN spectra confirm that a separate hard tail component is superposed onto the hot thermal emission, confirming a previous report based on the XIS data. Over the 5--40 keV band, the hard tail is spectrally approximated by a power law of photon index ~2, but better by those with somewhat convex shapes. Possible origins of the extended hard X-ray emission are discussed.Comment: 13 pages, 18 figure

    Production of TRPM4 knockout cell line using rat cardiomyocyte H9c2

    The method presented in this article are related to the research article entitled as "Role of the TRPM4 channel in mitochondrial function, calcium release, and ROS generation in oxidative stress" [1]. TRPM4, a non-selective monovalent cation channel, is not only involved in the generation of the action potential in cardiomyocytes, but also thought to be a key molecule in the development of the ischemia-reperfusion injury of the brain and the heart [2-5]. However, existing pharmacological inhibitors for the TRPM4 channel have problems of non-specificity [6]. This article describes methods used for targeted genomic deletion in the rat cardiomyocyte H9c2 using the CRISPR-Cas9 genome editing system in order to suppress TRPM4 protein expression. Confocal microscopy, flow cytometry, Sanger sequencing, and western blotting are performed to confirm vector transfection and the subsequent knockout of the TRPM4 protein. These data provide information on the comprehensive analyses for knocking out the rat TRPM4 channel using CRISPR/Cas9. The analyses include confocal microscopy, flow cytometry, Sanger sequencing, and western blotting. This dataset will benefit biological and medical researchers studying the function of TRPM4-expressing cells including neurons, cardiomyocytes, and vascular endothelial cells. It is also useful to study the involvement of the TRPM4 channel in pathological processes such as cardiac arrhythmia and ischemia-reperfusion injury.
