    How does the chromatin fiber deal with topological constraints?

    In the nuclei of eukaryotic cells, DNA is packaged through several levels of compaction in an orderly retrievable way that enables the correct regulation of gene expression. The functional dynamics of this assembly involves the unwinding of the so-called 30 nm chromatin fiber and accordingly imposes strong topological constraints. We present a general method for computing both the twist and the writhe of any winding pattern. An explicit derivation is implemented for the chromatin fiber which provides the linking number of DNA in eukaryotic chromosomes. We show that there exists one and only one unwinding path which satisfies both topological and mechanical constraints that DNA has to deal with during condensation/decondensation processes.Comment: Presented in Nature "News and views in brief" Vol. 429 (13 May 2004).     Selective vulnerability of neurons to acute toxicity after proteasome inhibitor treatment: Implications for oxidative stress and insolubility of newly synthesized proteins

    Maintaining protein homeostasis is vital to cell viability, with numerous studies demonstrating a role for proteasome inhibition occurring during the aging of a variety of tissues and, presumably, contributing to the disruption of cellular homeostasis during aging. In this study we sought to elucidate the differences between neurons and astrocytes in regard to basal levels of protein synthesis, proteasome-mediated protein degradation, and sensitivity to cytotoxicity after proteasome inhibitor treatment. In these studies we demonstrate that neurons have an increased vulnerability, compared to astrocyte cultures, to proteasome-inhibitor-induced cytotoxicity. No significant difference was observed between these two cell types in regard to the basal rates of protein synthesis, or basal rates of protein degradation, in the pool of short-lived proteins. After proteasome inhibitor treatment neuronal crude lysates were observed to undergo greater increases in the levels of ubiquitinated and oxidized proteins and selectively exhibited increased levels of newly synthesized proteins accumulating within the insoluble protein pool, compared to astrocytes. Together, these data suggest a role for increased oxidized proteins and sequestration of newly synthesized proteins in the insoluble protein pool, as potential mediators of the selective neurotoxicity after proteasome inhibitor treatment. The implications for neurons exhibiting increased sensitivity to acute proteasome inhibitor exposure, and the corresponding changes in protein homeostasis observed after proteasome inhibition, are discussed in the context of both aging and age-related disorders of the nervous system.Fil: Dasuri, Kalavathi.     Amino acid analog toxicity in primary rat neuronal and astrocyte cultures: Implications for protein misfolding and TDP-43 regulation

    Amino acid analogs promote translational errors that result in aberrant protein synthesis, and have been used to understand the effects of protein misfolding in a variety of physiological and pathological settings. TDP-43 is a protein that is linked to protein aggregation and toxicity in a variety of neurodegenerative diseases. In this study we exposed primary rat neurons and astrocyte cultures to established amino acid analogs (Canavanine and Azetidine-2-carboxylic acid), and observed both cell types undergo a dose-dependent increase in toxicity, with neurons exhibiting a greater degree of toxicity as compared to astrocytes. Neurons and astrocytes exhibited similar increases in ubiquitinated and oxidized protein following analog treatment. Analog treatment increased Heat shock protein (Hsp) levels in both neurons and astrocytes. In neurons, and to a lesser extent astrocytes, the levels of TDP-43 increased in response to analog treatment. Taken together, these data indicate that neurons exhibit preferential toxicity and alterations in TDP-43, in response to increased protein misfolding, as compared to astrocytes.Fil: Dasuri, Kalavathi.     First-order interference of nonclassical light emitted spontaneously at different times

    We study first-order interference in spontaneous parametric down-conversion generated by two pump pulses that do not overlap in time. The observed modulation in the angular distribution of the signal detector counting rate can only be explained in terms of a quantum mechanical description based on biphoton states.     Zum Zusammenhang zwischen Rational-Emotiver Theorie und Attributionstheorie: Irrationale Gedanken als Determinanten depressogener Ursachenzuschreibungen und maladaptiver Emotionen

    Die Rational-Emotive Theorie (RET) nach Ellis sowie die Attributionstheorien betrachten Kognitionen als notwendige und hinreichende Bedingungen bestimmter Emotionen. Zudem beinhalten beide Theoriekomplexe spezifische Darstellungen von Kognitionen, die der psychischen Gesundheit abträglich sind: Diese sind in der RET durch irrationale Gedanken („ich muss unbedingt...“) in der Attributionstheorie insbesondere durch den depressogenen Attributionsstil gekennzeichnet. Auf der Grundlage dieser theoretischen Gemeinsamkeiten überprüft eine experimentelle Fragebogenstudie mittels unterschiedlicher Szenarien in permutierten Darbietungen, inwiefern irrational („ich muss unbedingt...“) und rational („ich möchte gerne...“) denkenden Stimuluspersonen unterschiedliche Emotionen, Kausalattributionen und Verhaltensweisen zugeschrieben werden. Es zeigt sich, dass adaptive Emotionen bei rational denkenden und maladaptive Emotionen bei irrational denkenden Personen vermutet werden. Hinsichtlich der Dimensionen Stabilität, Lokation und Globalität ergeben sich für irrational im Unterschied zu den rational Denkenden signifikante und konsistente depressogene Attributionsmuster. Zudem wird bei den irrationalen Denkern tendenziell eine stärkere wahrgenommene Kontrollierbarkeit vermutet. Bezüglich zukünftiger Verhaltensweisen werden überwiegend bei rational denkenden Personen produktive Verhaltensresultate vermutet. Zusätzlich erhobene Einschätzungen bestätigen eine höhere Funktionalität adaptiver Emotionen

    Quantum interference by two temporally distinguishable pulses

    We report a two-photon interference effect, in which the entangled photon pairs are generated from two laser pulses well-separated in time. In a single pump pulse case, interference effects did not occur in our experimental scheme. However, by introducing a second pump pulse delayed in time, quantum interference was then observed. The visibility of the interference fringes shows dependence on the delay time between two laser pulses. The results are explained in terms of indistinguishability of biphoton amplitudes which originated from two temporally separated laser pulses.Comment: two-column, 4pages, submitted to PRA, minor change

    ウィスコンシン大学マディソン校で推進している南極無人気象観測計画(Antarctic Automatic Weather Station(AWS)program)の,2012-2013年のフィールド調査および結果の概要を報告する.今期はAWS 観測網の歴史上,特異なシーズンであった.ロス島地域が温暖であったことは氷上滑走路の利用に影響を及ぼし,いくつかの設営面での制約に直面した. 柔軟な計画により,限られた条件下でAWS サービルを最大化し,自動観測ネットワークへの要求に対応する最善の手段をとることができた.This report reviews 2012-2013 field season activities of the University of Wisconsin-Madison's Antarctic Automatic Weather Station (AWS) program, summarizes the science that these sites are supporting, and outlines the factors that impact the number of AWS sites serviced in any given field season. The 2012-2013 austral summer season was unusual in the AWS network history. Challenges encountered include, but are not limited to, warmer than normal conditions in the Ross Island area impacting airfield operations, changes to logistical procedures, and competition for shared resources. A flexible work plan provides the best means for taking on these challenges while maximizing AWS servicing efforts under restricted conditions and meeting the need for routine servicing that maintaining an autonomous observing network demands
