    Impact on permeability due to axial stress disturbances for cretaceous sandy shale

    Seismic waves generated from earthquakes and artificial surface vibration might alter the water level in the wells and oil or gas production. These transient stress disturbances prospectively caused the permeability change due to new pathway occurring or existing pathway being cleared. The permeability change might encourage enhancing gas recovery, inducing small earthquakes preventing future large earthquakes, and de-routing underground water flow for various purposes. The prospective permeability increase by axial stress disturbances of Cretaceous sandy shale may effectively expand the capacity of methane gas recovery of Kushiro Coal Mine. The paper observes the permeability change of intact or triaxially fractured Kushiro Cretaceous sandy shale by axial stress disturbances.     Spontaneous Cardiac Hypertrophy in a Crl:CD(SD) Rat

    Cardiac hypertrophy was observed in a 9-week-old Crl:CD(SD) rat that died unexpectedly. The animal was allocated to the control group of a toxicity study, and no abnormalities in its general conditions, body weight or food intake were observed. Necropsy revealed an increase in heart weight. Gross examination indicated cardiac enlargement with thickening of the right and left ventricular walls. Histopathological examination revealed hypertrophy of the cardiomyocytes in the right and left ventricular walls and the interventricular septum. Electron microscopic examination indicated bizarre nuclei and accumulation of an increased number of various sizes of mitochondria in the perinuclear region of the hypertrophied myocytes. Hypertrophied myocytes connected by intensely folded intercalated disks were also observed. Based on these findings, the animal was diagnosed with cardiac hypertrophy. This is the first case report of cardiac hypertrophy in this strain

    A Case Report of a Malignant Fibrous Histiocytoma in a T-cell Receptor β Chain and p53 Double-knockout Mouse

    A subcutaneous tumor was found in the right abdomen of a 16-week-old male TCRβ and p53 double-knockout mouse. The tumor had indistinct borders with the surrounding tissue. The cut surface after formalin fixation was pale yellowish white, partially dark red and partly white. Histologically, the tumor was composed of three distinct regions. The first region showed pleomorphic cells arranged in sheets. The second region showed spindle cells arranged in interlacing fascicles. The final region contained a mixture of the above mentioned two types of cells. Furthermore, a small amount of collagen fibers, round cells, multinucleated giant cells, and cells with eosinophilic granules were observed between these tumor cells. Immunohistochemical examination and electron microscopy identified that the pleomorphic cells and spindle cells were histiocytes and fibroblasts, respectively, and that the round cells were undifferentiated mesenchymal cells. Based on these findings, the tumor was diagnosed as a malignant fibrous histiocytoma

    Discovery of anti-inflammatory physiological peptides that promote tissue repair by reinforcing epithelial barrier formation

    上皮バリアを形成するペプチドJIPの発見 --JIPは上皮組織修復に貢献する--. 京都大学プレスリリース. 2021-11-18.Epithelial barriers that prevent dehydration and pathogen invasion are established by tight junctions (TJs), and their disruption leads to various inflammatory diseases and tissue destruction. However, a therapeutic strategy to overcome TJ disruption in diseases has not been established because of the lack of clinically applicable TJ-inducing molecules. Here, we found TJ-inducing peptides (JIPs) in mice and humans that corresponded to 35 to 42 residue peptides of the C terminus of alpha 1-antitrypsin (A1AT), an acute-phase anti-inflammatory protein. JIPs were inserted into the plasma membrane of epithelial cells, which promoted TJ formation by directly activating the heterotrimeric G protein G13. In a mouse intestinal epithelial injury model established by dextran sodium sulfate, mouse or human JIP administration restored TJ integrity and strongly prevented colitis. Our study has revealed TJ-inducing anti-inflammatory physiological peptides that play a critical role in tissue repair and proposes a previously unidentified therapeutic strategy for TJ-disrupted diseases

    視聴覚メディアの作成とその効果的な利用方法について : 「学習技術」の実践を通して

    大学生に必要なスタディ・スキルズを身につける授業として,平成13年度より「学習技術」が開講された。専門家が存在せず,しかもチームティーチングの形式で運用されるこの科目において,全クラスで同様の授業内容を実施することは容易ではない。その問題点を解決するための手段が,視聴覚メディアの作成であった。一斉授業で視聴覚メディアを利用することにより,クラス格差は小さいものになると期待される。本稿では,それらのメディアの作成過程を述べ,授業にどのように利用できるのかを考察する。In the 2001 academic year, Kansai University of International Studies started the new program "Learning Strategies (Gakushu Gijutsu)" for freshmen. This program is provided in a number of classes that proceed in parallel. Each class is directed by an instructor who is not necessarily expert in the discipline. This makes it difficult to ensure a uniform quality of instruction. To help the instructors manage classes effectively, and to maintain the required level of educational quality, we decided to develop original audio-visual materials as part of the "Learning Strategies" program. In this article, we report on the process of creating the materials and present some suggestions as to their effective use in class

    The whole blood transcriptional regulation landscape in 465 COVID-19 infected samples from Japan COVID-19 Task Force

    「コロナ制圧タスクフォース」COVID-19患者由来の血液細胞における遺伝子発現の網羅的解析 --重症度に応じた遺伝子発現の変化には、ヒトゲノム配列の個人差が影響する--. 京都大学プレスリリース. 2022-08-23.Coronavirus disease 2019 (COVID-19) is a recently-emerged infectious disease that has caused millions of deaths, where comprehensive understanding of disease mechanisms is still unestablished. In particular, studies of gene expression dynamics and regulation landscape in COVID-19 infected individuals are limited. Here, we report on a thorough analysis of whole blood RNA-seq data from 465 genotyped samples from the Japan COVID-19 Task Force, including 359 severe and 106 non-severe COVID-19 cases. We discover 1169 putative causal expression quantitative trait loci (eQTLs) including 34 possible colocalizations with biobank fine-mapping results of hematopoietic traits in a Japanese population, 1549 putative causal splice QTLs (sQTLs; e.g. two independent sQTLs at TOR1AIP1), as well as biologically interpretable trans-eQTL examples (e.g., REST and STING1), all fine-mapped at single variant resolution. We perform differential gene expression analysis to elucidate 198 genes with increased expression in severe COVID-19 cases and enriched for innate immune-related functions. Finally, we evaluate the limited but non-zero effect of COVID-19 phenotype on eQTL discovery, and highlight the presence of COVID-19 severity-interaction eQTLs (ieQTLs; e.g., CLEC4C and MYBL2). Our study provides a comprehensive catalog of whole blood regulatory variants in Japanese, as well as a reference for transcriptional landscapes in response to COVID-19 infection
