Potential of adenovirus-mediated REIC/Dkk-3 gene therapy for use in the treatment of pancreatic cancer
Background and AimThe reduced expression in immortalized cells REIC/the dickkopf 3 (Dkk-3) gene, tumor suppressor gene, is downregulated in various malignant tumors. In a prostate cancer study, an adenovirus vector carrying the REIC/Dkk-3 gene (Ad-REIC) induces apoptosis. In the current study, we examined the effects of REIC/Dkk-3 gene therapy in pancreatic cancer.
MethodsREIC/Dkk-3 expression was assessed by immunoblotting and immunohistochemistry in the pancreatic cancer cell lines (ASPC1, MIAPaCa2, Panc1, BxPC3, SUIT-2, KLM1, and T3M4) and pancreatic cancer tissues. The Ad-REIC agent was used to investigate the apoptotic effect in vitro and antitumor effects in vivo. We also assessed the therapeutic effects of Ad-REIC therapy with gemcitabine.
ResultsThe REIC/Dkk-3 expression was lost in the pancreatic cancer cell lines and decreased in pancreatic cancer tissues. Ad-REIC induced apoptosis and inhibited cell growth in the ASPC1 and MIAPaCa2 lines in vitro, and Ad-REIC inhibited tumor growth in the mouse xenograft model using ASPC1 cells. The antitumor effect was further enhanced in combination with gemcitabine. This synergistic effect may be caused by the suppression of autophagy via the enhancement of mammalian target of rapamycin signaling.
ConclusionsAd-REIC induces apoptosis and inhibits tumor growth in pancreatic cancer cell lines. REIC/Dkk-3 gene therapy is an attractive therapeutic tool for pancreatic cancer
PoGOLite - A High Sensitivity Balloon-Borne Soft Gamma-ray Polarimeter
We describe a new balloon-borne instrument (PoGOLite) capable of detecting
10% polarisation from 200mCrab point-like sources between 25 and 80keV in one 6
hour flight. Polarisation measurements in the soft gamma-ray band are expected
to provide a powerful probe into high-energy emission mechanisms as well as the
distribution of magnetic fields, radiation fields and interstellar matter. At
present, only exploratory polarisation measurements have been carried out in
the soft gamma-ray band. Reduction of the large background produced by
cosmic-ray particles has been the biggest challenge. PoGOLite uses Compton
scattering and photo-absorption in an array of 217 well-type phoswich detector
cells made of plastic and BGO scintillators surrounded by a BGO anticoincidence
shield and a thick polyethylene neutron shield. The narrow FOV (1.25msr)
obtained with well-type phoswich detector technology and the use of thick
background shields enhance the detected S/N ratio. Event selections based on
recorded phototube waveforms and Compton kinematics reduce the background to
that expected for a 40-100mCrab source between 25 and 50keV. A 6 hour
observation on the Crab will differentiate between the Polar Cap/Slot Gap,
Outer Gap, and Caustic models with greater than 5 sigma; and also cleanly
identify the Compton reflection component in the Cygnus X-1 hard state. The
first flight is planned for 2010 and long-duration flights from Sweden to

Abstract Background and Aims
The Golgi apparatus of goblet cells in the mouse descending colon : three-dimensional visualization using a confocal laser scanning microscope
The three-dimensional structure of the Golgi apparatus was studied in goblet cells in lectin-stained sections of the mouse descending colon by using a confocal laser scanning microscope. In the lower part of the crypt, the Golgi apparatus formed a dome-or globe-like structure in the supranuclear region. The wall of the dome or the globe had some holes, one of which usually faced toward the nucleus and others toward the apical cytoplasm. Mucous granules seemed to be initially released into the interior of the dome and transported toward the apical cytoplasm through the holes. In the upper part of the crypt, on the other hand, the Golgi apparatus formed a cup- or funnel-like structure with a larger opening toward the cell apex and a smaller opening toward the nucleus. A large mass ofmucous granules occupied the inside of the cup to the apical cytoplasm. It is thought that the accumulation of mucous granules enlarges holes at the ceiling of the dome to form a large opening, which makes the configuration of the Golgi apparatus cup-shaped
