    Vector-Based Approach for the Detection of Initial Dips Using Functional Near-Infrared Spectroscopy

    Functional near-infrared spectroscopy (fNIRS) is a non-invasive method for the detection of local brain activity using changes in the local levels of oxyhemoglobin (oxyHb) and deoxyhemoglobin (deoxyHb). Simultaneous measurement of the levels of oxyHb and deoxyHb is an advantage of fNIRS over other modalities. This review provides a historical description of the physiological problems involved in the accurate identification of local brain activity using fNIRS. The need for improved spatial and temporal identification of local brain activity is described in terms of the physiological challenges of task selection and placement of probes. In addition, this review discusses challenges with data analysis based on a single index, advantages of the simultaneous analysis of multiple indicators, and recently established composite indicators. The vector-based approach provides quantitative imaging of the phase and intensity contrast for oxygen exchange responses in a time series and may detect initial dips related to neuronal activity in the skull. The vector plane model consists of orthogonal vectors of oxyHb and deoxyHb. Initial dips are hemodynamic reactions of oxyHb and deoxyHb induced by increased oxygen consumption in the early tasks of approximately 2–3 seconds. The new analytical concept of fNIRS, able to effectively detect initial dips, may extend further the clinical and social applications of fNIRS

    Burrow Plasticity in the Deep-Sea Isopod Bathynomus doederleini (Crustacea: Isopoda: Cirolanidae)

    We investigated whether the deep-sea isopod Bathynomus doederleini has the capacity to change burrow length in response to changes in environmental conditions. We observed burrowing behavior in individuals that were placed on substrates with either simple (ST) or complex (CT) surface topographies. Individuals in the ST group (N = 10) constructed seven burrows. The mean ratio of the burrow length to body length was 1.8. The individuals in the CT group ( N = 10) constructed eight burrows with a mean ratio of burrow length to body length of 2.5. Thus the burrows were significantly longer in the CT group. In addition, the isopods in the CT group often incorporated a chamber in the mid-section of the burrow.     Relationship Between Brain Activity and Real-Road Driving Behavior: A Vector-Based Whole-Brain Functional Near-Infrared Spectroscopy Study

    Automobile driving requires multiple brain functions. However, the brain regions related to driving behavior are unknown. Therefore, we measured activity of the frontal, parietal and occipital lobes during driving using functional near-infrared spectroscopy (fNIRS). Cortical activation patterns were examined in relation to driving behaviors, such as steering motion, accelerator pedal motion, and speed control. Six healthy adults participated in the experiment. Cerebral oxygen exchange (COE) was calculated based on the oxyhemoglobin and deoxyhemoglobin concentrations measured by fNIRS. The COE and driving behavior data were collected every 1 m and averaged for all subjects. Functional NIRS data for all 98 channels were extracted using principal component analysis. Similarity between extracted components and driving behaviors were confirmed by |cosine similarity|\u3e0.3. Among the factors with confirmed similarity, we identified brain regions with high principal component loading (|PCL|\u3e0.4). Among the 16 COE factors extracted, COE factor 1 and factor 5 exhibited similarity with steering motion (cosine similarity: factor 1, -0.538; factor 5, 0.551). The PCLs of COE factor 1 and factor 5 were high in the frontal lobe (Brodmann areas [BAs] 9, 8, and 4/3) (PCL\u3e0.8). COE factor 6 exhibited a similarity with accelerator pedal motion (cosine similarity: 0.369), and the PCL of COE factor 6 was highest in the parietal lobe (BA7) (PCL= -0.62). Speed control did not exhibit similarity with any COE factor. These findings will contribute to the selection of brain measurement areas when fNIRS is used for vehicle driving assessment

    Genomic traces of Japanese malting barley breeding in two modern high-quality cultivars, ‘Sukai Golden’ and ‘Sachiho Golden’

    Two modern high-quality Japanese malting barley cultivars, ‘Sukai Golden’ and ‘Sachiho Golden’, were subjected to RNA-sequencing of transcripts extracted from 20-day-old immature seeds. Despite their close relation, 2,419 Sukai Golden-specific and 3,058 Sachiho Golden-specific SNPs were detected in comparison to the genome sequences of two reference cultivars: ‘Morex’ and ‘Haruna Nijo’. Two single nucleotide polymorphism (SNP) clusters respectively showing the incorporation of (1) the barley yellow mosaic virus (BaYMV) resistance gene rym5 from six-row non-malting Chinese landrace Mokusekko 3 on the long arm of 3H, and (2) the anthocyanin-less ant2 gene from a two-row Dutch cultivar on the long arm of 2H were detected specifically in ‘Sukai Golden’. Using 221 recombinant inbred lines of a cross between ‘Ishukushirazu’ and ‘Nishinochikara’, another BaYMV resistance rym3 gene derived from six-row non-malting Japanese cultivar ‘Haganemugi’ was mapped to a 0.4-cM interval on the proximal region of 5H. Haplotype analysis of progenitor accessions of the two modern malting cultivars revealed that rym3 of ‘Haganemugi’ was independently introduced into ‘Sukai Golden’ and ‘Sachiho Golden’. Residual chromosome 5H segments of ‘Haganemugi’ surrounding rym3 were larger in ‘Sukai Golden’. Available results suggest possibilities for malting quality improvement by minimizing residual segments surrounding rym3

    Deficiency of the basic helix‐loop‐helix transcription factor DEC1 prevents obesity induced by a high‐fat diet in mice

    Obesity is a major public health problem in developed countries resulting from increased food intake and decreased energy consumption and usually associated with abnormal lipid metabolism. Here, we show that DEC1, a basic helix‐loop‐helix transcription factor, plays an important role in the regulation of lipid consumption in mouse brown adipose tissue (BAT), which is the major site of thermogenesis. Homozygous Dec1 deletion attenuated high‐fat‐diet‐induced obesity, adipocyte hypertrophy, fat volume and hepatic steatosis. Furthermore, DEC1 deficiency increased body temperature during daytime and enhanced the expression of uncoupler protein 1, a key factor of thermogenesis, and various lipolysis‐related genes in interscapular BAT. In vitro experiments suggested that DEC1 suppresses the expression of various lipolysis‐related genes induced by the heterodimer of peroxisome proliferator‐activated receptor Îł and retinoid X receptor α (RXRα) through direct binding to RXRα. These observations suggest that enhanced lipolysis in BAT caused by DEC1 deficiency leads to an increase in lipid consumption, thereby decreasing lipid accumulation in adipose tissues and the liver.     Rituximab-combination chemotherapy achieves a 10th cycle of remission for Burkitt's lymphoma.

    A 14-year-old girl with multiple intra-abdominal tumors was diagnosed with stage III Burkitt's lymphoma. She achieved complete remission after multi-drug chemotherapy, but she relapsed after six courses. Autologous peripheral blood stem cells (PBSC) or allogeneic PBSC harvested from an HLA-identical sibling were insufficient, and her family did not agree to bone marrow collection from the sibling. Although the patient relapsed nine times (the relapses involved intra-abdominal organs or bone) during the following 4 years 7 months, treatment with rituximab monotherapy or in combination with ifosphamide, carboplastin, and etoposide, or local irradiation (33.8-40.0 Gy) to treat the bone metastases, proved effective, resulting in complete or partial remission. At the time of writing, the patient was in a 10th cycle of remission lasting 1 year 6 months and had not required transplantation. Thus, a chemotherapy regimen including rituximab might be effective for Burkitt's lymphoma in patients experiencing multiple relapse
