Fate of neutral-charged gold nanoparticles in the roots of the Hordeum vulgare L. cultivar Karat
Nanoparticles (NPs) have a significant impact on the environment and living organisms. The influence of NPs on plants is intensively studied and most of the data indicate that NPs can penetrate into plants. The studies presented here were performed on the roots of Hordeum vulgare L. seedlings using neutral-charge gold nanoparticles (AuNPs) of different sizes. In contrast to the majority of the published data, the results presented here showed that during the culture period, AuNPs: 1/did not enter the root regardless of their size and concentration, 2/that are applied directly into the cells of a root do not move into neighbouring cells. The results that were obtained indicate that in order to extend our knowledge about the mechanisms of the interactions between NPs and plants, further studies including, among others, on different species and a variety of growth conditions are needed
Effect of Pseudomonas moorei KB4 Cells’ Immobilisation on Their Degradation Potential and Tolerance towards Paracetamol
Pseudomonas moorei KB4 is capable of degrading paracetamol, but high concentrations
of this drug may cause an accumulation of toxic metabolites. It is known that immobilisation can
have a protective effect on bacterial cells; therefore, the toxicity and degradation rate of paracetamol
by the immobilised strain KB4 were assessed. Strain KB4 was immobilised on a plant sponge. A
toxicity assessment was performed by measuring the concentration of ATP using the colony-forming
unit (CFU) method. The kinetic parameters of paracetamol degradation were estimated using the
Hill equation. Toxicity analysis showed a protective effect of the carrier at low concentrations of
paracetamol. Moreover, a pronounced phenomenon of hormesis was observed in the immobilised
systems. The obtained kinetic parameters and the course of the kinetic curves clearly indicate
a decrease in the degradation activity of cells after their immobilisation. There was a delay in
degradation in the systems with free cells without glucose and immobilised cells with glucose.
However, it was demonstrated that the immobilised systems can degrade at least ten succeeding
cycles of 20 mg/L paracetamol degradation. The obtained results indicate that the immobilised strain
may become a useful tool in the process of paracetamol degradation
Changes in oxidative stress parameters in patients with peripheral vascular disease in response to conservative and surgical treatment
Wstęp. Typowym zjawiskiem w niedokrwieniu i reperfuzji tkanek
jest "wybuch tlenowy" związany z aktywacją płytek krwi i neutrofilów, produkcją
wolnych rodników i wywołane nimi uszkodzenie komórek śródbłonka naczyń oraz dalsze
upośledzenie tkanek.
Cel pracy. Badanie przeprowadzono w celu porównania wpływu dwóch rodzajów
terapii: chirurgicznej rewaskularyzacji i terapii zachowawczej (zaprogramowany
trening i pentoksyfilina p.o.) na wybrane parametry powysiłkowego stresu oksydacyjnego
u pacjentów z miażdżycą naczyń obwodowych.
Materiał i metody. Do badań zakwalifikowano pacjentów z przewlekłym niedokrwieniem
kończyn dolnych leczonych zachowawczo (n = 24, II stopień wg klasyfikacji Fontaine'a,
grupa C) lub leczonych chirurgicznie (n = 14, III stopień wg klasyfikacji Fontaine'a,
grupa O). Pacjentów leczonych zachowawczo poddawano przez 12 tygodni regularnym
treningom na bieżni ruchomej i podawano im pentoksyfilinę p.o. w dawce 3 × 400
mg/d. Leczenie chirurgiczne polegało na wszczepianiu zespoleń omijających aortalno-udowych
bądź udowo-podkolanowych. Próbki krwi do badań biochemicznych pobierano przed
rozpoczęciem terapii, po 6 i 12 tygodniu od rozpoczęcia leczenia, przed testem
wysiłkowym i po jego wykonywaniu. Oznaczano parametry stresu oksydacyjnego (osoczowe
stężenia nadtlenków lipidowych wyrażone stężeniem TBARS i podatność LDL na oksydację
ex vivo) oraz parametry charakterystyczne dla mechanizmów antyoksydacyjnych osocza
(całkowita osoczowa zdolność redukcyjna: FRAP oraz stosunek stężeń grup tiolowych
do albuminy: SH/A).
Wyniki. Leczenie chirurgiczne spowodowało znaczną poprawę parametrów klinicznych
wyrażoną zmniejszeniem dolegliwości subiektywnych oraz statystycznie znamiennym
wzrostem indeksu kostka/ramię (ABI, ankle-brachial index), zmniejszeniem pola niedokrwienia
i wydłużeniem dystansu chromania. U chorych leczonych zachowawczo również nastąpiła
poprawa wyrażona wydłużeniem dystansu chromania i zmniejszeniem pola niedokrwienia,
ale bez istotnej poprawy ABI. Leczenie chirurgiczne nie chroniło jednak chorych
przed wzmożonym stresem oksydacyjnym wyrażonym wzrostem TBARS i wzmożoną podatnością
LDL na oksydację.
Natomiast u chorych leczonych zachowawczo obserwowano istotny wzrost wartości
FRAP i SH/A, co świadczy o poprawie potencjału antyoksydacyjnego.
Wniosek. W przeciwieństwie do leczenia zachowawczego leczenie chirurgiczne
u pacjentów z zaawansowaną miażdżycą nie chroni naczyń obwodowych przed stresem
oksydacyjnym. Wyniki badań wskazują na korzystny wpływ wykonywania regularnych
ćwiczeń i stosowania farmakoterapii (antyoksydanty) u chorych z miażdżycą naczyń
obwodowych po zabiegach rewaskularyzacyjnych.Introduction. Reperfusion of ischemic tissue is associated with oxidative
burst. Systemic response e.g. neutrophil and platelet activation, metabolic changes,
free radical and cytokine production resulting in endothelial injury are the typical
features of ischemia/reperfusion.
Materials and methods. Twenty four patients (group C) with chronic leg
ischemia (Fontaine grade II) undergoing conservative (12 weeks of controlled treadmill
training and pentoxifylline administration (400 mg p.o; t.i.d.) and 14 patients
(group O) (Fontaine grade III), who underwent surgical revascularization were
included in the present study. Revascularization included aorto-femoral or femoro-popliteal
bypass implantation. Blood samples for evaluation of oxidative stress: plasma
lipid peroxide level (TBARS) and the ex vivo LDL suscepti bility for oxidation,
as well as for measurement of the plasma antioxidant status (total plasma antioxidative
power - FRAP), and total plasma thiol/albumin ratio were collected before and
after the treadmill exercise workload at the entry, after 6 and after 12 weeks
from initiation of the treatment. The changes in clinical status were expressed
by the treadmill pain-free walking distance and ankle/arm index values.
Results. The surgical revascularization resulted in the significant improvement
of clinical parameters demonstrated by reduction of patient complaints as well
as by increased ankle/arm index values, smaller ischemic field and prolongation
of pain-free and maximal walking time. An increase of the pain-free walking distance
and the decrease of ischaemic field, but any improvement of the ankle/arm index
was observed in the conservative therapy group. However, surgical revascularisation
did not protect patients from increased oxidative stress expressed by the TBARS
concentration and LDL susceptibility to oxidation ex vivo. On the contrary, the
conservative therapy resulted in the increased plasma antioxidant status measured
by the FRAP assay and the increased plasma thiol/albumin ratio.
Conclusion. The surgical revascularization did not protect patients with
advanced peripheral arterial disease from ischemia/reperfusion oxidative stress
what argue for the complementary supplementation of antioxidants during therapy
