    Nöroblastoma Hücre Hattında Uzun Süreli Darbeli Elektromanyetik Alan Maruziyetinin Apoptoz Üzerine Etkileri

    Darbeli Elektromanyetik Alan (PEMF) düşük frekanslı elektromanyetik alan olup son yıllarda klinik araştırmalarda tedavi amaçlı uygulanmasına yönelik çalışmalar hız kazanmıştır. Farklı frekans, yoğunluk, dalga boyu ve sürelerde kematerapötik ilaçlarla birlikte uygulanan PEMF maruziyetinin beyin kanseri hücreleri dahil çeşitli kanser hücrelerinde apoptoz üzerine etkilerini değerlendiren çalışmalardan farklı olarak çalışmada sabit frekans ve yoğunlukta (50 Hz, 1 mT) uzun süre (48 saat) PEMF maruziyetinin SK-N-SH insan nöroblastoma hücresinde apoptoz mekanizmasına olası etkileri farklı tekniklerle araştırılmıştır. Hücreler kontrol grubu, PEMF maruziyetinin olmadığı SK-N-SH hücre grubu, ve PEMF’ye 48 saat maruz bırakılan SK-N-SH hücre grubu olmak üzere üçe ayrılmıştır. Hücre canlılığı, apoptoz tayini, kaspaz-8 mRNA düzeyi ve kaspaz-8 protein ekspresyonu sırasıyla alamar mavisi, akış sitometri, qRT-PCR ve Western-Blot teknikleriyle belirlenmiştir. Uzun süreli PEMF maruziyetinin insan nöroblastoma hücresinde hücre canlılığını belirgin şekilde azaltıp hücreleri daha fazla erken apoptoza uğratarak hücreleri apoptoza sürüklediği ve bu mekanizmanın kaspaz-8 mRNA düzeyinde ve protein ekspresyon seviyesinde artışla ilişkili olabileceği gösterilmiştir

    HPLC-UV analysis of phenolic compounds and biological activities of Padina pavonica and Zanardinia typus marine macroalgae species

    The marine macroalgae species are recognized as the food of the future with excellent bioactive properties. This study aimed to investigate phenolic compounds; cytotoxic, antibacterial, and antioxidant activities; total phenolic (TPC) and flavonoid (TFC) contents of the methanol, ethanol, and water extracts of Padina pavonica (PP) and Zanardinia typus (ZT). To the best of our knowledge, this is the first report in which ZT was evaluated in terms of phenolic content, antioxidant, antibacterial, and cytotoxic activities. The HPLC analysis allowed the identification of five phenolic compounds containing rutin (0.31 ± 0.04 – 3.00 ± 0.21 ppm) in all extracts and trans-p-coumaric acid (0.15 ± 0.02 – 3.95 ± 0.02 ppm) in MPP, EPP, MZT, EZT, WZT as the major compounds. TPC and TFC of the extracts were calculated as 11.78 ± 0.75 – 76.78 ± 0.54 µg GAEs/mg extract and 6.78 ± 0.17 – 29.50 ± 2.23 µg QEs/mg extract, respectively. The highest cytotoxicity was observed in EZT (CC50:132.3 ± 22.4 µg/mL) against MCF-7 and MZT (CC50: 91.4 ± 20.9 µg/mL) against MIA PaCa-2. Among the studied extracts, EPP showed the best antibacterial activity against all test pathogens. Also, EPP indicated superior antibacterial activity against Plesiomonas shigelloides (MIC: 1.25 mg/mL) and Staphylococcus aureus (MIC: 1.25 mg/mL). EZT displayed the highest antioxidant activity in DPPH• (IC50: 49.03 ± 0.28 µg/mL), CUPRAC (A0.50: 15.20 ± 0.14 µg/mL), and ABTS•+ (IC50: 18.86 ± 0.74 µg/mL) assays. The results approved that P. pavonica and Z. typus marine macroalgae species could be valued as natural sources of bioactive agents for food and pharmacology applications

    Real life experience of patients with locally advanced gastric and gastroesophageal junction adenocarcinoma treated with neoadjuvant chemotherapy: a Turkish oncology group study

    Neoadjuvant chemotherapy (NACT) in gastroesophageal junction (GEJ) and gastric cancer (GC) was shown to improve survival in recent studies. We aimed to share our real-life experience of patients who received NACT to compare the efficacy and toxicity profile of different chemotherapy regimens in our country. This retrospective multicentre study included locally advanced GC and GEJ cancer patients who received NACT between 2007 and 2021. Relation between CT regimens and pathological evaluation were analysed. A total of 794 patients from 45 oncology centers in Turkey were included. Median age at the time of diagnosis was 60 (range: 18-86). Most frequent NACT regimens used were FLOT (65.4%), DCF (17.4%) and ECF (8.1%), respectively. In the total study group, pathological complete remission (pCR) rate was 7.2%, R0 resection rate 86.4%, and D2 dissection rate was 66.8%. Rate of pCR and near-CR (24%), and R0 resection (84%) were numerically higher in FLOT arm (p > 0.05). Patients who received FLOT had also higher chemotherapy-related toxicity rate compared to patients who received other regimens (p > 0.05). Median follow-up time was 16 months (range: 1-154 months). Estimated median overall survival (OS) was 58.4months (95% CI: 35.2-85.7) and disease-free survival (DFS) was 50.7 months (95% CI: 25.4-75.9). The highest 3-year estimated OS rate was also shown in FLOT arm (68%). We still do not know which NACT regimen is the best choice for daily practice. Clinicians should tailor treatment regimens according to patients' multifactorial status and comorbidities for to obtain best outcomes. Longer follow-up period needs to validate our results

    Incidence and clinical characteristics of acute rheumatic fever in Turkey: Results of a nationwide multicentre study

    © 2021 Paediatrics and Child Health Division (The Royal Australasian College of Physicians)Aim: To evaluate the incidence and clinical features of acute rheumatic fever (ARF) in Turkey, following the revised Jones criteria in 2015. Methods: This multicentre study was designed by the Acquired Heart Diseases Working Group of the Turkish Pediatric Cardiology and Pediatric Cardiac Surgery Association in 2016. The data during the first attack of 1103 ARF patients were collected from the paediatric cardiologists between 1 January 2016 and 31 December 2016. Results: Turkey National Institute of Statistics records of 2016 were used for the determination of ARF incidence with regard to various cities and regions separately. The estimated incidence rate of ARF was 8.84/100 000 in Turkey. The ARF incidence varied considerably among different regions. The highest incidence was found in the Eastern Anatolia Region as 14.4/100 000, and the lowest incidence was found in the Black Sea Region as 3.3/100 000 (P < 0.05). Clinical carditis was the most common finding. The incidence of clinical carditis, subclinical carditis, polyarthritis, aseptic monoarthritis, polyarthralgia and Sydenham's Chorea was 53.5%, 29.1%, 52.8%, 10.3%, 18.6% and 7.9%, respectively. The incidences of clinical carditis, subclinical carditis, polyarthritis and polyarthralgia were found to be significantly different among different regions (P < 0.05). Conclusion: The findings of this nationwide screening of ARF suggest that Turkey should be included in the moderate-risk group
