    Sedimentation and Type I X-ray Bursts at Low Accretion Rates

    Neutron stars, with their strong surface gravity, have interestingly short timescales for the sedimentation of heavy elements. Motivated by observations of Type I X-ray bursts from sources with extremely low persistent accretion luminosities, L_X < 10^{36}\usp\ergspersecond (\simeq 0.01\ensuremath{L_{\mathrm{Edd}}}), we study how sedimentation affects the distribution of isotopes and the ignition of H and He in the envelope of an accreting neutron star. For local mass accretion rates \mdot \lesssim 10^{-2}\medd (for which the ignition of H is unstable), where \medd = 8.8\times 10^{4}\nsp\gpscps, the helium and CNO elements sediment out of the accreted fuel before reaching a temperature where H would ignite. Using one-zone calculations of the thermonuclear burning, we find a range of accretion rates for which the unstable H ignition does not trigger unstable He burning. This range depends on the emergent flux from reactions in the deep neutron star crust; for F = 0.1\nsp\MeV(\dot{m}/\mb), the range is 3\times 10^{-3}\medd\lesssim\mdot\lesssim 10^{-2}\medd. We speculate that sources accreting in this range will build up a massive He layer that later produces an energetic and long X-ray burst. At mass accretion rates lower than this range, we find that the H flash leads to a strong mixed H/He flash. Surprisingly, even at accretion rates \mdot \gtrsim 0.1\medd, although the H and He do not completely segregate, the H abundance at the base of the accumulated layer is still reduced. While following the evolution of the X-ray burst is beyond the scope of this introductory paper, we note that the reduced proton-to-seed ratio favors the production of \iso{12}{C}--an important ingredient for subsequent superbursts.Comment: 15 pages, 14 figures, submitted to ApJ, revised versio

    Fast X-ray Transients and Their Connection to Gamma-Ray Bursts

    Fast X-ray transients (FXTs) with timescales from seconds to hours have been seen by numerous space instruments. We have assembled archival data from Ariel-5, HEAO-1 (A-1 and A-2), WATCH, ROSAT, and Einstein to produce a global fluence-frequency relationship for these events. Fitting the log N-log S distribution over several orders of magnitude to simple power law we find a slope of -1.0. The sources of FXTs are undoubtedly heterogeneous, the -1 power law is an approximate result of the summation of these multiple sources. Two major contributions come from gamma-ray bursts and stellar flares. Extrapolating from the BATSE catalog of GRBs, we find that the fraction of X-ray flashes that can be the X-ray counterparts of gamma-ray bursts is a function of fluence. Certainly most FXTs are not counterparts of standard gamma-ray bursts. The fraction of FXTs from non-GRB sources, such as magnetic stars, is greatest for the faintest FXTs. Our understanding of the FXT phenomenon remains limited and would greatly benefit from a large, homogeneous data set, which requires a wide-field, sensitive instrument.Comment: 36 pages, 8 figure

    Measurement of migration of a humeral head resurfacing prosthesis using radiostereometry without implant marking: An experimental study

    Today, the shoulder joint is the third most commonly replaced joint after the hip and knee joints and the incidence is increasing. In Sweden, 1863 primary Shoulder Arthroplasties and 195 revisions were performed in 2017. The most common diagnoses are Osteoarthritis and irreparable tears of the rotator cuff, with or without arthropathy, often referred to as cuff tear arthropathy. Different Shoulder Arthroplasty (SA) concepts include anatomical total shoulder arthroplasty (TSA), hemiarthroplasty (HSA) and reversed shoulder arthroplasty, but also humeral head resurfacing (HHR) and stemless arthroplasties. All concepts offer pain relief, improvement of function and in quality of life for the different diagnoses. Unfortunately, there are sometimes complications after SA. They involve periprosthetic joint infection, humeral and glenoid fractures, stress shielding, loosening of the glenoid and humeral component but also glenoid erosion and cuff rupture. Some of these complications are most common within 1 year after operation, some after several years, both may lead to a revision. This, together with the fact that new designs of implants and methods of fixation of SA continues to develop, stresses the importance of continuous monitoring of implant survival and follow-up. The overall aim of this thesis was to describe clinical examples of different methods to assess the outcome after Shoulder Arthroplasty. The most common methods are clinical examination, radiographic assessment, Patient Reported Outcome Measure (PROM), National Joint registries, where revisions are an important outcome, but also Clinical Trials. All of these methods are used in one or more of the 4 papers in this thesis and shows the complexity of the topic and the practical work. In paper I we used Radio Stereometric Analysis (RSA) in an experimental set-up and concluded that marker-free RSA can be used for a humeral head resurfacing arthroplasty. In paper II we used data from the Swedish Shoulder Arthroplasty Registry (SSAR) with PROM and revisions to conclude that age is the only factor that affects revision when comparing HSA and HHR. Paper III is a long-time follow-up of a Randomized controlled study where we used radiological assessment, PROM and revisions. The conclusion was that both TSA and HSA develop severe radiological changes 10 year after primary operation. Paper IV is a prospective RSA cohort study where we also evaluated PROM and revisions. The conclusion is that HHR seems to obtain a secure fixation in the humerus, after an initial migration. But also that the prostheses shows continuous glenoid wear. The main conclusion of this thesis is that patient’s operated with SA needs continuous monitoring and several methods may be used to evaluate the outcome

    Design, Synthesis and Discovery of N,N'-Carbazoyl-aryl-urea Inhibitors of Zika NS5 Methyltransferase and Virus Replication

    The recent outbreaks of Zika virus (ZIKV) infection worldwide make the discovery of novel antivirals against flaviviruses a research priority. This work describes the identification of novel inhibitors of ZIKV through a structure‐based virtual screening approach using the ZIKV NS5‐MTase. A novel series of molecules with a carbazoyl‐aryl‐urea structure has been discovered and a library of analogues has been synthesized. The new compounds inhibit ZIKV MTase with IC50 between 23–48 μM. In addition, carbazoyl‐aryl‐ureas also proved to inhibit ZIKV replication activity at micromolar concentration

    An analogue of the antibiotic teicoplanin prevents flavivirus entry in vitro.

    There is an urgent need for potent inhibitors of dengue virus (DENV) replication for the treatment and/or prophylaxis of infections with this virus. We here report on an aglycon analogue of the antibiotic teicoplanin (code name LCTA-949) that inhibits DENV-induced cytopathic effect (CPE) in a dose-dependent manner. Virus infection was completely inhibited at concentrations that had no adverse effect on the host cells. These findings were corroborated by quantification of viral RNA levels in culture supernatant. Antiviral activity was also observed against other flaviviruses such as the yellow fever virus and the tick-borne encephalitis virus (TBEV). In particular, potent antiviral activity was observed against TBEV. Time-of-drug-addition experiments indicated that LCTA-949 inhibits an early stage in the DENV replication cycle; however, a virucidal effect was excluded. This observation was corroborated by the fact that LCTA-949 lacks activity on DENV subgenomic replicon (that does not encode structural proteins) replication. Using a microsopy-based binding and fusion assay employing DiD-labeled viruses, it was shown that LCTA-949 targets the early stage (binding/entry) of the infection. Moreover, LCTA-949 efficiently inhibits infectivity of DENV particles pre-opsonized with antibodies, thus potentially also inhibiting antibody-dependent enhancement (ADE). In conclusion, LCTA-949 exerts in vitro activity against several flaviviruses and does so (as shown for DENV) by interfering with an early step in the viral replication cycle.Fil: De Burghgraeve, Tine. The thematic collection, which this paper complements, entitled ‘Integrity and Its Counterfeits: Implications for Economy, Business and Management’, makes a contribution towards filling the gap between the abstract concept of integrity and its application into business and economy, with a particular attention on the ambiguous, equivocal and diverse meanings of the concept, the complex and dynamic practicality of integrity, and the grey and dark areas of business out of integrity. This article introduces the background of the research theme and provides exemplary debates and emerging avenues of discussion on this topic

    Tune in to your emotions: a robust personalized affective music player

    The emotional power of music is exploited in a personalized affective music player (AMP) that selects music for mood enhancement. A biosignal approach is used to measure listeners’ personal emotional reactions to their own music as input for affective user models. Regression and kernel density estimation are applied to model the physiological changes the music elicits. Using these models, personalized music selections based on an affective goal state can be made. The AMP was validated in real-world trials over the course of several weeks. Results show that our models can cope with noisy situations and handle large inter-individual differences in the music domain. The AMP augments music listening where its techniques enable automated affect guidance. Our approach provides valuable insights for affective computing and user modeling, for which the AMP is a suitable carrier application
