    Prototype laser-diode-pumped solid state laser transmitters

    Monolithic, diode-pumped Nd:YAG ring lasers can provide diffraction-limited, single-frequency, narrow-linewidth, tunable output which is adequate for use as a local oscillator in a coherent communication system. A laser was built which had a linewidth of about 2 kHz, a power of 5 milliwatts, and which was tunable over a range of 30 MHz in a few microseconds. This laser was phase-locked to a second, similar laser. This demonstrates that the powerful technique of heterodyne detection is possible with a diode-pumped laser used as the local oscillator. Laser diode pumping of monolithic Nd:YAG rings can lead to output powers of hundreds of milliwatts from a single laser. A laser was built with a single-mode output of 310 mW. Several lasers can be chained together to sum their power, while maintaining diffraction-limited, single frequency operation. This technique was demonstrated with two lasers, with a total output of 340 mW, and is expected to be practical for up to about ten lasers. Thus with lasers of 310 mW, output of up to 3 W is possible. The chaining technique, if properly engineered, results in redundancy. The technique of resonant external modulation and doubling is designed to efficiently convert the continuous wave, infrared output of our lasers into low duty-cycle pulsed green output. This technique was verified through both computer modeling and experimentation. Further work would be necessary to develop a deliverable system using this technique

    Molecular characterization of freshwater snails in the genus Bulinus: a role for barcodes?

    <p>Abstract</p> <p>Background</p> <p>Reliable and consistent methods are required for the identification and classification of freshwater snails belonging to the genus <it>Bulinus </it>(Gastropoda, Planorbidae) which act as intermediate hosts for schistosomes of both medical and veterinary importance. The current project worked towards two main objectives, the development of a cost effective, simple screening method for the routine identification of <it>Bulinus </it>isolates and the use of resultant sequencing data to produce a model of relationships within the group.</p> <p>Results</p> <p>Phylogenetic analysis of the DNA sequence for a large section (1009 bp) of the mitochondrial gene cytochrome oxidase subunit 1 (<it>cox1</it>) for isolates of <it>Bulinus </it>demonstrated superior resolution over that employing the second internal transcribed spacer <it>(its2</it>) of the ribosomal gene complex. Removal of transitional substitutions within <it>cox1 </it>because of saturation effects still allowed identification of snails at species group level. Within groups, some species could be identified with ease but there were regions where the high degree of molecular diversity meant that clear identification of species was problematic, this was particularly so within the <it>B. africanus </it>group.</p> <p>Conclusion</p> <p>The sequence diversity within <it>cox1 </it>is such that a barcoding approach may offer the best method for characterization of populations and species within the genus from different geographical locations. The study has confirmed the definition of some accepted species within the species groups but additionally has revealed some unrecognized isolates which underlines the need to use molecular markers in addition to more traditional methods of identification. A barcoding approach based on part of the <it>cox1 </it>gene as defined by the Folmer primers is proposed.</p

    The human mutator gene homolog MSH2 and its association with hereditary nonpolyposis colon cancer

    We have identified a human homolog of the bacterial MutS and S. cerevisiae MSH proteins, called hMSH2. Expression of hMSH2 in E. coli causes a dominant mutator phenotype, suggesting that hMSH2, like other divergent MutS homologs, interferes with the normal bacterial mismatch repair pathway. hMSH2 maps to human chromosome 2p22-21 near a locus implicated in hereditary nonpolyposis colon cancer (HNPCC). A T to C transition mutation has been detected in the -6 position of a splice acceptor site in sporadic colon tumors and in affected individuals of two small HNPCC kindreds. These data and reports indicating that S. cerevisiae msh2 mutations cause an instability of dinucleotide repeats like those associated with HNPCC suggest that hMSH2 is the HNPCC gene

    Spin and interaction effects on charge distribution and currents in one-dimensional conductors and rings within the Hartree-Fock approximation

    Using the self--consistent Hartree-Fock approximation for electrons with spin at zero temperature, we study the effect of the electronic interactions on the charge distribution in a one-dimensional continuous ring containing a single δ\delta scatterer. We reestablish that the interaction suppresses the decay of the Friedel oscillations. Based on this result, we show that in an infinite one dimensional conductor containing a weak scatterer, the current is totally suppressed because of a gap opened at the Fermi energy. In a canonical ensemble of continuous rings containing many scatterers, the interactions enhance the average and the typical persistent current.Comment: 5 pages, 4 figure

    Increasingly, libraries are recognizing the importance of providing access to the research output of their universities. In a June 10, 2019, news release from the provosts of the Big Ten Academic Alliance (BTAA) titled “Sustaining Values and Scholarship” (available at https://tinyurl.com/yyu94aa9), they state, “The Big Ten Academic Alliance will continue its advocacy for a sustainable and open ecosystem of publication. . . . Collectively, our institutions’ more than 50,000 faculty are supported by over $10 billion (2017) in research funding, and our institutions have similarly invested significantly in our capacity to further our missions to advance knowledge. Together, we produce roughly 15% of the research publications in the United States.” This commitment to open information is reflected in the mission of the Media and Repository Support (MARS) unit of the UNL Libraries. We support the creation and preservation of the scholarly accomplishments of the University of Nebraska–Lincoln by providing an infrastructure for disseminating information and scholarship through Digital Commons (articles, reports, journals, books, and more), Luna (multimedia projects), and Rosetta (data) as well as by providing equipment that students and faculty can check out to capture video and digital images. Through our efforts, the intellectual contributions of UNL are provided, when possible, as open access to disseminate information to as wide a community as possible. We provide advice and consult with researchers and students on scholarly communication issues surrounding open education resources, copyright, options for rights management, publication, and preservation of information in coordination with other faculty and staff in the Libraries. We maintain close contact with staff from Archives & Special Collections to ensure that nondigital information is not overlooked in preservation plans. Faculty within the unit have developed close relationships with the UNL Office of Research, faculty across our campus, and colleagues at the other University of Nebraska campuses (through the University of Nebraska Consortium of Libraries, UNCL)

    On representation of the t-J model via spin-charge variables

    We show that the t-J Hamiltonian is not in general reduced to H(S,f), where S and f stand for independent ([S,f]=0) SU(2) (spin) generators and spinless fermionic (hole) field, respectively. The proof is based upon an identification of the Hubbard operators with the generators of the su(2|1) superalgebra in the degenerate fundamental representation and ensuing SU(2|1) path integral representation of the partition function.Comment: 15 pages, latex, no figure

    Biomphalaria glabrata transcriptome: cDNA microarray profiling identifies resistant- and susceptible-specific gene expression in haemocytes from snail strains exposed to Schistosoma mansoni

    BACKGROUND: Biomphalaria glabrata is an intermediate snail host for Schistosoma mansoni, one of the important schistosomes infecting man. B. glabrata/S. mansoni provides a useful model system for investigating the intimate interactions between host and parasite. Examining differential gene expression between S. mansoni-exposed schistosome-resistant and susceptible snail lines will identify genes and pathways that may be involved in snail defences. RESULTS: We have developed a 2053 element cDNA microarray for B. glabrata containing clones from ORESTES (Open Reading frame ESTs) libraries, suppression subtractive hybridization (SSH) libraries and clones identified in previous expression studies. Snail haemocyte RNA, extracted from parasite-challenged resistant and susceptible snails, 2 to 24 h post-exposure to S. mansoni, was hybridized to the custom made cDNA microarray and 98 differentially expressed genes or gene clusters were identified, 94 resistant-associated and 4 susceptible-associated. Quantitative PCR analysis verified the cDNA microarray results for representative transcripts. Differentially expressed genes were annotated and clustered using gene ontology (GO) terminology and Kyoto Encyclopaedia of Genes and Genomes (KEGG) pathway analysis. 61% of the identified differentially expressed genes have no known function including the 4 susceptible strain-specific transcripts. Resistant strain-specific expression of genes implicated in innate immunity of invertebrates was identified, including hydrolytic enzymes such as cathepsin L, a cysteine proteinase involved in lysis of phagocytosed particles; metabolic enzymes such as ornithine decarboxylase, the rate-limiting enzyme in the production of polyamines, important in inflammation and infection processes, as well as scavenging damaging free radicals produced during production of reactive oxygen species; stress response genes such as HSP70; proteins involved in signalling, such as importin 7 and copine 1, cytoplasmic intermediate filament (IF) protein and transcription enzymes such as elongation factor 1α and EF-2. CONCLUSION: Production of the first cDNA microarray for profiling gene expression in B. glabrata provides a foundation for expanding our understanding of pathways and genes involved in the snail internal defence system (IDS). We demonstrate resistant strain-specific expression of genes potentially associated with the snail IDS, ranging from signalling and inflammation responses through to lysis of proteinacous products (encapsulated sporocysts or phagocytosed parasite components) and processing/degradation of these targeted products by ubiquitination

    Addressing the Ethical, Legal, and Social Issues Raised by Voting by Persons with Dementia

    This article addresses an emerging policy problem in the United States participation in the electoral process by citizens with dementia. At present, health care professionals, family caregivers, and long-term care staff lack adequate guidance to decide whether individuals with dementia should be precluded from or assisted in casting a ballot. Voting by persons with dementia raises a series of important questions about the autonomy of individuals with dementia, the integrity of the electoral process, and the prevention of fraud. Three subsidiary issues warrant special attention: development of a method to assess capacity to vote; identification of appropriate kinds of assistance to enable persons with cognitive impairment to vote; and formulation of uniform and workable policies for voting in long-term care settings. In some instances, extrapolation from existing policies and research permits reasonable recommendations to guide policy and practice. However, in other instances, additional research is necessary
