    Programmatic feasibility of dried blood spots for the virological follow-up of patients on antiretroviral treatment in Nord Kivu, Democratic Republic of the Congo

    Background:As part of its policy to shift monitoring of antiretroviral therapy (ART) to primary health care (PHC) workers, the Ministry of Health of the Democratic Republic of Congo (DRC) tested the feasibility of using dried blood spots (DBS) for viral load (VL) quantification and genotypic drug resistance testing in off-site high-throughput laboratories.Methods:DBS samples from adults on ART were collected in 13 decentralized PHC facilities in the Nord-Kivu province and shipped during program quarterly supervision to a reference laboratory 2000 km away, where VL was quantified with a commercial assay (m2000rt, Abbott). A second DBS was sent to a World Health Organization (WHO)-accredited laboratory for repeat VL quantification on a subset of samples with a generic assay (Biocentric) and genotypic drug resistance testing when VL >1000 copies per milliliter.Findings:Constraints arose because of an interruption in national laboratory funding rather than to technical or logistic problems. All samples were assessed by both VL assays to allow ART adjustment. Median DBS turnaround time was 37 days (interquartile range: 9-59). Assays performed unequally with DBS, impacting clinical decisions, quality assurance, and overall cost-effectiveness. Based on m2000rt or generic assay, 31.3% of patients were on virological failure (VF) and 14.8% presented resistance mutations versus 50.3% and 15.4%, respectively.Conclusion:This study confirms that current technologies involving DBS make virological monitoring of ART possible at PHC level, including in challenging environments, provided organizational issues are addressed. Adequate core funding of HIV laboratories and adapted choice of VL assays require urgent attention to control resistance to ART as coverage expands

    Population dynamics and genetic connectivity in recent chimpanzee history

    Knowledge on the population history of endangered species is critical for conservation, but whole-genome data on chimpanzees (<Pan troglodytes) is geographically sparse. Here, we produced the first non-invasive geolocalized catalog of genomic diversity by capturing chromosome 21 from 828 non-invasive samples collected at 48 sampling sites across Africa. The four recognized subspecies show clear genetic differentiation correlating with known barriers, while previously undescribed genetic exchange suggests that these have been permeable on a local scale. We obtained a detailed reconstruction of population stratification and fine-scale patterns of isolation, migration, and connectivity, including a comprehensive picture of admixture with bonobos (Pan paniscus). Unlike humans, chimpanzees did not experience extended episodes of long-distance migrations, which might have limited cultural transmission. Finally, based on local rare variation, we implement a fine-grained geolocalization approach demonstrating improved precision in determining the origin of confiscated chimpanzees

    Co2+ substituted Mg–Cu–Zn ferrite: Evaluation of structural, magnetic, and electromagnetic properties

    Abstract We report the synthesis of Co2+ substituted Mg–Cu–Zn ferrite via citrate gel combustion process and thereby its structural, transport, and magnetic properties for the use in electromagnetic energy absorption application. The polycrystalline ferrite system is investigated by interplay of stoichiometric composition with Mg0.25–x Co x Cu0.25Zn0.5Fe2O4 (0 â©œ x â©œ 0.25). Structural investigations using X-ray diffraction (XRD) and selected area electron diffraction (SAED) reveal the formation of spinel structure with linear growth of lattice constant due to Co2+ substitution. The microstructural analysis (TEM and SEM) depicts the dense microstructure with the average grain size of 0.42–1.25 ÎŒm. The elemental analysis (EDS) confirms the elemental composition of the as-prepared ferrite with respect to the initial concentrations of the synthetic composition used. The observed variations in initial permeability (ÎŒ i) and magnetic moment (n B) are explained based on deviation in saturation magnetization (M s), anisotropy constant (K 1), density values, and exchange interaction. The temperature dependence of DC resistivity confirms the semiconducting behavior of the as-prepared ferrite material, with an increase in the DC resistivity by an incorporation of cobalt. Furthermore, the effects of adding Co2+ on the Curie temperature, frequency dependent dielectric properties of the ferrite material are also discussed

    Evaluating methods for detecting and monitoring pangolin (Pholidata: Manidae) populations

    The behaviours, ecologies and morphologies of pangolins make them challenging to survey and monitor, and non-targeted wildlife surveys have not produced robust status assessments, especially where population densities are low because of overexploitation. To inform the development of feasible survey and monitoring techniques for pangolins, we conducted a systematic review of all traceable efforts used to survey and monitor pangolins to date: 87 articles were included in the review. Pitfalls of current approaches are discussed and recommendations made on suitable methods. Recommendations include the use of mark-recapture for burrow-dwelling species, community interviews, sign-based surveys in arid and open habitats, detection dog teams, and targeted camera-trapping. Occupancy sampling using camera-traps could be used to monitor some pangolin populations, particularly ground-dwelling species, but the rarity of all species makes it uncertain whether this would provide enough data for monitoring; combinations of methods used within an occupancy sampling framework are likely to be the most effective. There will be many circumstances where direct monitoring of a population at a site, to a level that will generate precise data, is not financially viable nor the best use of conservation resources. In many sites, particularly in Asia, pangolins are too rare as a result of overexploitation, and/or occur in inaccessible areas where significant resources will be needed to implement a targeted monitoring programme. Under such circumstances, the use of proxy variables, including status of other hunting-sensitive species that are easier to record, in combination with enforcement or patrol data and/or community interviews, is likely to be the most cost-effective method for assessing the impact of conservation interventions on pangolin status. The publication of incidental observations and survey ‘by-catch’ would significantly improve understanding of pangolin status and ecology, and therefore how best to identify, conserve and monitor priority populations

    We sought to determine whether motor and cognitive deficits associated with cassava (food) cyanogenic poisoning were associated with high concentrations of F2-isoprostanes, well-established indicators of oxidative damage. Concentrations of serum F2-isoprostanes were quantified by LC-MS/MS and anchored to measures of motor proficiency and cognitive performance, which were respectively assessed through BOT-2 (Bruininks/Oseretsky Test, 2nd Edition) and KABC-II (Kaufman Assessment Battery for Children, 2nd edition) testing of 40 Congolese children (21 with konzo and 19 presumably healthy controls, overall mean age (SD): 9.3 (3.2) years). Exposure to cyanide was ascertained by concentrations of its main metabolite thiocyanate (SCN) in plasma and urine. Overall, SCN concentrations ranged from 91 to 325 and 172 to 1032 ”mol/l in plasma and urine, respectively. Serum isoprostanes ranged from 0.1 to 0.8 (Isoprostane-III), 0.8 to 8.3 (total Isoprostane-III), 0.1 to 1.5 (Isoprostane-VI), 2.0 to 9.0 (total Isoprostane-VI), or 0.2 to 1.3 ng/ml (8,12-iso-iPF2α-VI isoprostane). Children with konzo poorly performed at the BOT-2 and KABC-II testing relative to presumably healthy children (p<0.01). Within regression models adjusting for age, gender, motor proficiency, and other biochemical variables, 8,12-iso-iPF2α-VI isoprostane was significantly associated with the overall cognitive performance (ÎČ = -32.36 (95% CI: -51.59 to -13.03; P<0.001). This model explained over 85% of variation of the KABC-II score in children with konzo, but was not significant in explaining the motor proficiency impairment. These findings suggest that cognitive deficits and, possibly, brain injury associated with cassava poisoning is mediated in part by oxidative damage in children with konzo. 8,12-iso-iPF2α-VI isoprostane appears to be a good marker of the neuropathogenic mechanisms of konzo and may be used to monitor the impact of interventional trials to prevent the neurotoxic effects of cassava cyanogenic poisoning

    Precursor-free eruption triggered by edifice rupture at Nyiragongo volcano

    International audienceAbstract Classical mechanisms of volcanic eruptions mostly involve pressure buildup and magma ascent towards the surface 1 . Such processes produce geophysical and geochemical signals that may be detected and interpreted as eruption precursors 1–3 . On 22 May 2021, Mount Nyiragongo (Democratic Republic of the Congo), an open-vent volcano with a persistent lava lake perched within its summit crater, shook up this interpretation by producing an approximately six-hour-long flank eruption without apparent precursors, followed—rather than preceded—by lateral magma motion into the crust. Here we show that this reversed sequence was most likely initiated by a rupture of the edifice, producing deadly lava flows and triggering a voluminous 25-km-long dyke intrusion. The dyke propagated southwards at very shallow depth (less than 500 m) underneath the cities of Goma (Democratic Republic of the Congo) and Gisenyi (Rwanda), as well as Lake Kivu. This volcanic crisis raises new questions about the mechanisms controlling such eruptions and the possibility of facing substantially more hazardous events, such as effusions within densely urbanized areas, phreato-magmatism or a limnic eruption from the gas-rich Lake Kivu. It also more generally highlights the challenges faced with open-vent volcanoes for monitoring, early detection and risk management when a significant volume of magma is stored close to the surface

    Development and evaluation of a genome-wide Coffee 8.5K SNP array and its application for high-density genetic mapping and for investigating the origin of Coffea arabica L

    Coffee species such as Coffea canephora P. (Robusta) and C. arabica L. (Arabica) are important cash crops in tropical regions around the world. C. arabica is an allotetraploid (2n = 4x = 44) originating from a hybridization event of the two diploid species C. canephora and C. eugenioides (2n = 2x = 22). Interestingly, these progenitor species harbour a greater level of genetic variability and are an important source of genes to broaden the narrow Arabica genetic base. Here, we describe the development, evaluation and use of a single-nucleotide polymorphism (SNP) array for coffee trees. A total of 8580 unique and informative SNPs were selected from C. canephora and C. arabica sequencing data, with 40% of the SNP located in annotated genes. In particular, this array contains 227 markers associated to 149 genes and traits of agronomic importance. Among these, 7065 SNPs (82.3%) were scorable and evenly distributed over the genome with a mean distance of 54.4 Kb between markers. With this array, we improved the Robusta high-density genetic map by adding 1307 SNP markers, whereas 945 SNPs were found segregating in the Arabica mapping progeny. A panel of C. canephora accessions was successfully discriminated and over 70% of the SNP markers were transferable across the three species. Furthermore, the canephora-derived subgenome of C. arabica was shown to be more closely related to C. canephora accessions from northern Uganda than to other current populations. These validated SNP markers and high-density genetic maps will be useful to molecular genetics and for innovative approaches in coffee breeding
