Türkiye'deki köy tavuklarında i̇nfeksiyöz laringotrakeitis virüsünün moleküler tespiti ve karakterizasyonu
Gallid herpesvirus I (GaHV1) is etiological agent of infectious laryngotracheitis (ILT) and ILT is one of the
important disease that included in respiratory infections of chickens. Few studies have been conducted in Turkey
and there is no data about the existence of ILT in backyards. The purpose of the study was to document the
detection and characterization of GaHV1 in backyard chickens using PCR, RFLP and sequencing. Of the163
tracheal swap samples which were taken from 43 backyard flocks 5 (3.07%) were found to be positive for ILT
infection. Positivity was 4.65 % (2/43) at flock level. The nucleic acid sequences of the ICP4 gene compared with
ILTV sequences in GenBank and the level of identity differed from 96.82 to 100%. When the sequences of
samples compared with TCO strains 99.77-100% homology was observed. The virtual RFLP analysis with the
HgaI restriction enzyme characterized strains as having a pattern similar to the vaccine strain TCO. This is the
first study that presents presence and characterization of GaHV1 in backyards. Large scale studies are needed to
estimate prevalence of ILT in Turkey. Chickens should be monitored and growers should avoid to contacting
vaccinated birds with non-vaccinated chickens, to control ILTV outbreaks.Gallid herpesvirüs I (GaHV1), infeksiyöz laringotrakeitisin (ILT) etiyolojik ajanıdır ve ILT, tavukların solunum yolu
enfeksiyonları arasında yer alan önemli hastalıklardan biridir. Türkiye'de çok az çalışma yapılmış ve köy
tavuklarında ILT'nin varlığına dair veri bulunmamaktadır. Çalışmada, köy tavuklarında PCR, RFLP ve sekans
verileri kullanarak GaHV1'in tespitini ve karakterizasyonu amaçlandı. Toplamda 43 köy tavuğu işletmesinden
alınan 163 trakeal swap örneğinin 5'inde (% 3,07) ILT enfeksiyonu pozitif bulundu. Sürü düzeyinde pozitiflik %
4,65 (2/43) idi. ICP4 geninin nükleik asit dizileri GenBank'taki ILTV dizileri ile karşılaştırıldığında benzerlik %
96.82 ile % 100 arasında değiştiği görüldü. Örneklerin sekans dizileri TCO suşları ile karşılaştırıldığında % 99.77-
100 homoloji gözlemlendi. HgaI enzimi ile yapılan sanal RFLP analizinde, çalışmadaki suşlar TCO aşı suşuna
benzer bir paterne sahip olarak karakterize edildi. Bu araştırma, köy tavuklarında GaHV1'in varlığını ve
karakterizasyonunu ortaya koyan ilk çalışmadır. Türkiye'de ILT prevalansını tahmin etmek için büyük ölçekli
çalışmalara ihtiyaç vardır. ILTV salgınlarını kontrol etmek için tavuklar izlenmeli ve yetiştiriciler aşılanmamış
tavuklarla aşılanmış olanların temas etmesinden kaçınmalıdır
KOMORBİD RİSK FAKTÖRLERİYLE AKUT İSKEMİK İNME HASTALARINDA COVID-19 MORTALİTEYİ ARTIRABİLİR
GİRİŞ VE AMAÇ: COVID-19 hastalarında birçok sistemikkomplikasyonlar olduğu gibi santral ve periferik sinirsistemi tutulumlarıda görülebilmektedir. COVID-19vakalarında yaklaşık %36 oranında nörolojiksemptomlar ortaya çıkmaktadır. COVID-19enfeksiyonunda ortaya çıkan sitokin artışı ile birlikteprotrombotik aktivasyonla yaygın mikrovaskülertrombozların geliştiği bildirilmiştir ve klinik pratikte buhastalarda D-dimer düzeyleri yüksek bulunmaktadır. Buçalışmada amacımız COVID-19 pozitif hastalarda serebrovasküler hastalık görülme sıklığı ve klinikkorelasyonunu değerlendirmektir.YÖNTEM: Erciyes Üniversitesi Tıp fakültesinde COVID19 tanısı ile pandemi servisinde takip edilip taburcu olanve ayaktan takip edilen hastalar çalışmaya alındı.Hastaların demografik bilgileri, akut serebrovaskülerhastalık hikayesi, beyin tomografi ve/veya manyetikrezonans görüntüleri, ek hastalıkları, kullandıklarıilaçlar, kan kolesterol düzeyleri, rutin biyokimyasal kantetkikleri, hemogram, CRP, D-dimer gibi laboratuvarverileri elektronik ortamdaki medikal kayıtlardantarandı. Akut gelişen ve COVID-19 tanısıyla zamansalilişkili serebrovasküler hastalıklar değerlendirmeyealındı. Serebrovasküler olaylar iskemik ve hemorajikolay, geçici iskemik atak ve serebral venöz trombozşeklinde 4 grupta incelendi. Bu hastalarınserebrovasküler hastalığa yönelik risk faktörleri dedeğerlendirilip hangi risk faktörüne sahip hastalardakoronavirüs serebrovasküler hastalık açısından dahafazla risk oluşturduğu da incelendi.BULGULAR: Koronavirüs enfeksiyonu geçirenhastalarda inme sıklığını belirlemek amacıylaretrospektif olarak 1000 hasta tarandı. İskemikserebrovasküler hastalık ve/veya geçici iskemik ataktanısı alan 14 hasta tespit edildi (11 iskemik olay, 3geçici iskemik atak). Akut iskemik inmeli COVID-19 tanılıhastaların yaş ortalaması 67 idi. Hastaların 8’i erkek, 6’sıkadın idi. COVID-19 tanılı akut iskemik inmeli 14 hastadaCOVID’e bağlı en sık görülen semptomlar ateş vesolunum güçlüğü olarak belirlendi. Hastaların ortalamabaşvuru glukoz değerleri 151 mg/dl, lenfosit değerleri1.2 birim, D-Dimer seviyeleri 1 hastada 14540 gibiyüksek bir değerde iken ortalama değeri 2445 olaraktespit edildi. CRP ortalama değerleri 90 idi. Başvuruortalama lökosit değeri 6200 idi. Tüm hastaların beyintomografisi ve diffüzyon MR görüntülemeleri incelendi.Enfarkt lokalizasyonuna baktığımızda, 4 hastadaposterior sistem enfarktı, 8 hastada ise anterior sistemenfarktı ve 2 hastada multipl enfarkt alanları izlendi.Başvuru NIHSS skoru 1 hastada 11 iken ortalama değer2.2 olarak belirlendi. Taburculuk ortalama NIHSS 1.1olarak tespit edildi. İnme ve akciğer tutulumu olan 4hasta vefat ederken, taburcu olanların ortalama modifiyerankin skoru 2 olarak bulundu. İnme risk faktörleriincelendiğinde, 4 hastada hipertansiyon, 3 hastadadiyabetes mellitus, 3 hastada hipertansiyon ve diyabetbirlikteliği, 2 hastada hipertansiyon ve geçirilmişiskemik serebrovasküler hastalık, 2 hastada kalpyetmezliği, 4 hastada atrial fibrilasyon tespit edildi.Hastaların yarısında çoklu inme risk faktörü olduğugörüldü.TARTIŞMA VE SONUÇ: Çalışmamızda COVID-19 pozitifhasta grubunda inme oranını %1.4 olarak bulduk.Pandeminin başından beri yapılan birçok çalışmalar,COVID-19 olan hastalarda inme insidansının %0,9 ile%2,7 arasında değiştiğini göstermiştir. Koronavirüsünher zaman solunum yolu ile sınırlı olmayıp; nörolojikbelirtilerle de karşımıza gelebilmektedir. COVID-19hastalığı ile birlikte akut iskemik inmesi olan hastalarda,inmesi olmayanlara göre karaciğer, renal ve solunumsistemi gibi multisistem tutulum daha fazlagörülmektedir. Bizim çalışmamızda da vefat edenhastaların solunum sıkıntısı ve akciğer tutulumu ön plandaydı. Özellikle akciğer tutulumu ön planda olan veakut iskemik inme geçiren hastalarda mortalitenin dahayüksek seyredeceğini öngörerek bu hastalarda yakıntakibin daha dikkatli yapılması gerektiği literatürleuyumlu olarak bizim çalışmamızın sonucuyla dasöylenebilir.</p
Production, characterization and therapeutic efficacy of egg yolk antibodies specific to Nosema ceranae.
Nosema disease, caused by Nosema ceranae, one of the single-celled fungal microsporidian parasites, is one of the most important and common diseases of adult honey bees. Since fumagillin, which has been used for decades in the control of Nosema disease in honey bees (Apis mellifera), poses a toxic threat and its efficacy against N. ceranae is uncertain, there is an urgent need to develop alternative prophylactic and curative strategies for the treatment of this disease. The main aim of this study was to investigate the therapeutic potential of specific egg yolk immunoglobulins (IgY) on Nosema disease. For this purpose, the presence of N. ceranae was determined by microscopic and PCR methods in honey bees collected from Nosema suspicious colonies by conducting a field survey. Layered Ataks chickens, divided into four groups each containing 20 animals, were vaccinated with live and inactivated vaccines prepared from field isolates of N. ceranae. Eggs were collected weekly for 10 weeks following the last vaccination. IgY extraction was performed using the PEG precipitation method from egg yolks collected from each group, and the purity of the antibodies was determined by SDS-PAGE and Western Blot. The presence of N. ceranae-specific IgYs was investigated by Western Blot and indirect ELISA methods. It was determined that specific IgYs showed high therapeutic efficacy on Nosema disease in naturally infected bee colonies. In addition, honey bees collected from infected colonies were brought to the laboratory and placed in cages with 30 bees each, and the effectiveness of IgYs was investigated under controlled conditions. It was detected that specific IgY reduced the Nosema spore load and the number of infected bees significantly in both the field and experimental study groups treated for seven days. It was concluded that chicken IgYs, an innovative and eco-friendly method, had a significant potential for use as an alternative to antifungal drugs

The data were presented as mean ± standard deviationa-d. The difference between the groups within the same week, expressed with different letter, was statistically significant (p<0.05). One-way ANOVA was used in the comparison of the groups and Post Hoc Tukey test was employed in the detection of the groups causing the difference.</p
