    Antibacterial Effect of Bovine Lactoferrin Against Udder Pathogens

    The antibacterial effect of lactoferrin (Lf) was tested on isolates of Escherichia coli (E. coli), Staphylococcus aureus (S. aureus), and coagulase-negative staphylococci (CNS) as well as on Pseudomonas aeruginosa (P. aeruginosa) and Klebsiella pneumoniae (K. pneumoniae), originally isolated from bovine mastitis. Concentrations of Lf used were 0.67 mg/ml, 1.67 mg/ml, and 2.67 mg/ml. Growth of udder pathogens was monitored by turbidometry either in broth culture or in whey prepared from normal milk. We focused on 3 different growth variables: lag time, slope, and maximum absorbance of bacterial growth curves. Growth inhibition was seen in the broth but hardly at all in whey. The isolates of E. coli and CNS did not grow sufficiently well in whey to draw any conclusions. The most effective inhibitory activity of Lf was seen against E. coli and P. aeruginosa. All 5 E. coli isolates had similar growth patterns. Inhibition of growth by Lf was concentration-dependent. The concentration of 0.67 mg/ml in broth and whey was generally too low for a significant inhibitory effect

    The construction and validation of a short form of the developmental, diagnostic and dimensional interview

    We aimed to construct and validate a shortened form of the developmental, diagnostic and dimensional interview (3Di), a parent report interview for assessing and diagnosing autistic spectrum disorders (ASDs). Data from 879 children and young people were used. In half of the sample (n = 440) reliability analysis was used to identify 3Di items that best measured each dimension of the autism triad. This informed the construction of a shortened (53 item) 3Di, which was then validated on subjects not used in the reliability analysis (n = 439). This involved comparison with scores from the original 3Di algorithm and, in a subsample (n = 29), with the autism diagnostic interview-revised (ADI-R). Agreement of the new shortened 3Di with the 3Di’s original algorithm was excellent in both dimensional and categorical terms. Agreement on caseness (27 out of 29) with the ADI-R was also strong. The new 3Di short version is less than half as long as the original version and outputs very similar scores. It will be useful to clinicians and researchers for obtaining a dimensional autism assessment in less than 45 minutes

    Power line mapping technique using all-terrain mobile laser scanning

    Power line mapping using remote sensing can automate the traditionally labor-intensive power line corridor inspection. Land-based mobile laser scanning (MLS) can be a good choice for the power line mapping if an aerial inspection is impossible, too costly or slow, unsafe, prohibited by regulations, or if more detailed information on the power line corridor is needed. The mapping of the power lines using MLS was studied in a rural environment outside the road network for the first time. An automatic power line extraction algorithm was developed. The algorithm first found power line candidate points based on the shape and orientation of the local neighborhood of a point using principal component analysis. Power lines were retrieved from the candidates using random sample consensus (Ransac) and a new power line labeling method, which takes into account the three-dimensional shape of the power lines. The new labeling method was able to find the power lines and remove false detections, which were found, for example, from the forest. The algorithm was tested in forested and open field (arable land) areas, outside the road environment using two different platforms of MLS, namely, personal backpack and all-terrain vehicle. The recall and precision of the power line extraction were 93.3% and 93.6%, respectively, using 10 cm as a distance criterion for a successful detection. Drifting of the positioning solution of the scanner was the largest error source, being the (contributory) cause for 60–70% of the errors. The platform did not have a significant effect on the power line extraction accuracy. The accuracy was higher in the open field compared to the forest, because the one-dimensional point density along the power line was inhomogeneous and GNSS (global navigation satellite system) signal was weak in the forest. The results suggest that the power lines can be mapped accurately enough for inspection purposes using MLS in a rural environment outside the road network.</p

    Prescribing antimicrobial agents for dogs and cats via University Pharmacies in Finland : Patterns and quality of information

    The aim of our study was to evaluate antimicrobial use in dogs and cats in Finland. Information on veterinary prescriptions was gathered from University Pharmacies (n = 17) over a one-month period, April 2001. A total of 2719 prescriptions for veterinary use were delivered, of which the majority were for dogs (70%, n = 1898) and cats (14%, n = 384). The most prescribed therapy group was per-oral antimicrobial agents (53%, n = 1449), of which 16% (n = 237) were medicines approved for humans. The most commonly used substances for dogs and cats were betalactams, 66% and 78%, respectively. The proportion of fluoroquinolones was 3–5%. The average duration of the treatment periods was 10 days with the exception of treatment of cats with macrolide lincosamides, where the mean period was 20 days. Indication was mentioned only in 37% of the prescriptions

    Alk2/ACVR1 and Alk3/BMPR1A Provide Essential Function for Bone Morphogenetic Protein–Induced Retinal AngiogenesisHighlights

    OBJECTIVE: Increasing evidence suggests that bone morphogenetic protein (BMP) signaling regulates angiogenesis. Here, we aimed to define the function of BMP receptors in regulating early postnatal angiogenesis by analysis of inducible, endothelial-specific deletion of the BMP receptor components Bmpr2 (BMP type 2 receptor), Alk1 (activin receptor-like kinase 1), Alk2, and Alk3 in mouse retinal vessels. APPROACH AND RESULTS: Expression analysis of several BMP ligands showed that proangiogenic BMP ligands are highly expressed in postnatal retinas. Consistently, BMP receptors are also strongly expressed in retina with a distinct pattern. To assess the function of BMP signaling in retinal angiogenesis, we first generated mice carrying an endothelial-specific inducible deletion of Bmpr2. Postnatal deletion of Bmpr2 in endothelial cells substantially decreased the number of angiogenic sprouts at the vascular front and branch points behind the front, leading to attenuated radial expansion. To identify critical BMPR1s (BMP type 1 receptors) associated with BMPR2 in retinal angiogenesis, we generated endothelial-specific inducible deletion of 3 BMPR1s abundantly expressed in endothelial cells and analyzed the respective phenotypes. Among these, endothelial-specific deletion of either Alk2/acvr1 or Alk3/Bmpr1a caused a delay in radial expansion, reminiscent of vascular defects associated with postnatal endothelial-specific deletion of BMPR2, suggesting that ALK2/ACVR1 and ALK3/BMPR1A are likely to be the critical BMPR1s necessary for proangiogenic BMP signaling in retinal vessels. CONCLUSIONS: Our data identify BMP signaling mediated by coordination of ALK2/ACVR1, ALK3/BMPR1A, and BMPR2 as an essential proangiogenic cue for retinal vessels

    Mucin Production by SPOC1 Cells - An Immortalized Rat Tracheal Epithelial Cell Line

    An airway epithelial mucous goblet cell line would be useful towards understanding mechanisms underlying the common problem of respiratory mucus hypersecretion. SPOC1 is a novel rat tracheal epithelial (RTE) cell line that developed cytologie features suggestive of mucous goblet cells when grown in tracheal grafts in vivo (Am. J. Respir. Cell Mol. Biol. 1995; 12:385-395). Our aims were to determine whether SPOC1 cells were capable of mucin synthesis and to directly compare mucin production by SPOC1 cells and RTE cells. Towards this end, we validated the use of monoclonal antibody (mAb) RTE11 (Exp. Lung Res. 1992; 18:323-342) as an immunologie probe for rat airway secretory mucin. Our results strongly suggest that mAb RTE11 detects a carbohydrate antigen that is a sensitive and specific marker for rat tracheobronchial secretory mucin. SPOC1 cells in tracheal grafts in vivo contained granules with ultrastructural features similar to mucous granules in normal rat airway goblet cells and they were strongly stained by mAb RTE11. Retinoic acid (RA) and culture on porous supports are known to profoundly modify airway epithelial cell phenotype in vitro. Expression of several retinoid-responsive proteins was similar in cultured SPOC1 and primary RTE cells, but major differences in mucin production were noted. Primary RTE cells in vitro only made mucin when grown on porous supports in the presence of RA, whereas SPOC1 cells produced mucin when grown on plastic or glass surfaces and even in the absence of RA. Interestingly, RA enhanced mucin secretion by SPOC1 cells during the early plateau stage of culture but there were no differences due to RA late in the culture period. SPOC1 cells are capable of mucin production and will be a useful tool for studying select aspects of airway secretory cell differentiation and function

    Functional redundancy of type I and type II receptors in the regulation of skeletal muscle growth by myostatin and activin A.

    Myostatin (MSTN) is a transforming growth factor-ÎČ (TGF-ÎČ) family member that normally acts to limit muscle growth. The function of MSTN is partially redundant with that of another TGF-ÎČ family member, activin A. MSTN and activin A are capable of signaling through a complex of type II and type I receptors. Here, we investigated the roles of two type II receptors (ACVR2 and ACVR2B) and two type I receptors (ALK4 and ALK5) in the regulation of muscle mass by these ligands by genetically targeting these receptors either alone or in combination specifically in myofibers in mice. We show that targeting signaling in myofibers is sufficient to cause significant increases in muscle mass, showing that myofibers are the direct target for signaling by these ligands in the regulation of muscle growth. Moreover, we show that there is functional redundancy between the two type II receptors as well as between the two type I receptors and that all four type II/type I receptor combinations are utilized in vivo. Targeting signaling specifically in myofibers also led to reductions in overall body fat content and improved glucose metabolism in mice fed either regular chow or a high-fat diet, demonstrating that these metabolic effects are the result of enhanced muscling. We observed no effect, however, on either bone density or muscle regeneration in mice in which signaling was targeted in myofibers. The latter finding implies that MSTN likely signals to other cells, such as satellite cells, in addition to myofibers to regulate muscle homeostasis

    This article applies Dumont’s view of ideology to an Eastern Indonesian society with intense trade connections to other ethnic groups and the larger political economy. In spite of their commercial importance, these connections are framed as long-distance kinship. My question is whether this encompassment of economic by social values is part of a totalizing ideological order. I discuss the values of personhood and exchange to show that long-distance commerce is the source of social differentiation expressed in them. Ultimately, however, the test of Dumont’s methodology is not whether it helps explain the resilience of local social orders, but whether it can deal with historical complexity and change. I argue that Dumont’s answer – hybrid ideology – is a good description for the encompassment of both kin-based totalities and political-economic stratificationPeer reviewe
