    Optimization of the ultrasonic processing in a melt flow

    Ultrasonic cavitation treatment of melt significantly improves the downstream properties and quality of conventional and advanced metallic materials. However, the transfer of this technology to treating large melt volumes has been hindered by a lack of fundamental knowledge, allowing for the ultrasonic processing in the melt flow. In this study, we present the results of experimental validation of an advanced numerical model applied to the acoustic cavitation treatment of liquid aluminum during continuous flow [1]. This was achieved by using a calibrated high-temperature cavitometer. The acoustic spectrum was analyzed at various points across the launder while acoustic pressures were calculated at the frequencies of interest.This work is performed within the Ultramelt Project supported by the EPSRC Grants EP/K005804/1 and EP/K00588X/1

    Analysis of the Effects of Polymorphism on Pollen Profilin Structural Functionality and the Generation of Conformational, T- and B-Cell Epitopes

    An extensive polymorphism analysis of pollen profilin, a fundamental regulator of the actin cytoskeleton dynamics, has been performed with a major focus in 3D-folding maintenance, changes in the 2-D structural elements, surface residues involved in ligands-profilin interactions and functionality, and the generation of conformational and lineal B- and T-cell epitopes variability. Our results revealed that while the general fold is conserved among profilins, substantial structural differences were found, particularly affecting the special distribution and length of different 2-D structural elements (i.e. cysteine residues), characteristic loops and coils, and numerous micro-heterogeneities present in fundamental residues directly involved in the interacting motifs, and to some extension these residues nearby to the ligand-interacting areas. Differential changes as result of polymorphism might contribute to generate functional variability among the plethora of profilin isoforms present in the olive pollen from different genetic background (olive cultivars), and between plant species, since biochemical interacting properties and binding affinities to natural ligands may be affected, particularly the interactions with different actin isoforms and phosphoinositides lipids species. Furthermore, conspicuous variability in lineal and conformational epitopes was found between profilins belonging to the same olive cultivar, and among different cultivars as direct implication of sequences polymorphism. The variability of the residues taking part of IgE-binding epitopes might be the final responsible of the differences in cross-reactivity among olive pollen cultivars, among pollen and plant-derived food allergens, as well as between distantly related pollen species, leading to a variable range of allergy reactions among atopic patients. Identification and analysis of commonly shared and specific epitopes in profilin isoforms is essential to gain knowledge about the interacting surface of these epitopes, and for a better understanding of immune responses, helping design and development of rational and effective immunotherapy strategies for the treatment of allergy diseases.     New Pharmacological Agents to Aid Smoking Cessation and Tobacco Harm Reduction: What has been Investigated and What is in the Pipeline?

    A wide range of support is available to help smokers to quit and aid attempts at harm reduction, including three first-line smoking cessation medications: nicotine replacement therapy, varenicline and bupropion. Despite the efficacy of these, there is a continual need to diversify the range of medications so that the needs of tobacco users are met. This paper compares the first-line smoking cessation medications to: 1) two variants of these existing products: new galenic formulations of varenicline and novel nicotine delivery devices; and 2) twenty-four alternative products: cytisine (novel outside of central and eastern Europe), nortriptyline, other tricyclic antidepressants, electronic cigarettes, clonidine (an anxiolytic), other anxiolytics (e.g. buspirone), selective 5-hydroxytryptamine (5-HT) reuptake inhibitors, supplements (e.g. St John’s wort), silver acetate, nicobrevin, modafinil, venlafaxine, monoamine oxidase inhibitors (MAOI), opioid antagonist, nicotinic acetylcholine receptors (nAChR) antagonists, glucose tablets, selective cannabinoid type 1 receptor antagonists, nicotine vaccines, drugs that affect gamma-aminobutyric acid (GABA) transmission, drugs that affect N-methyl-D-aspartate receptors (NMDA), dopamine agonists (e.g. levodopa), pioglitazone (Actos; OMS405), noradrenaline reuptake inhibitors, and the weight management drug lorcaserin. Six criteria are used: relative efficacy, relative safety, relative cost, relative use (overall impact of effective medication use), relative scope (ability to serve new groups of patients), and relative ease of use (ESCUSE). Many of these products are in the early stages of clinical trials, however, cytisine looks most promising in having established efficacy and safety and being of low cost. Electronic cigarettes have become very popular, appear to be efficacious and are safer than smoking, but issues of continued dependence and possible harms need to be considered

    Pre-operative assessment of relationship between inferior dental nerve canal and mandibular impacted third molar in Saudi population

    OBJECTIVE: To study the correlation between the position of the inferior dental (ID) nerve canal and the angulation of impacted mandibular third molars using dental cone beam computed tomography (CBCT). MATERIALS AND METHODS: The study considered 100 impactions in 85 patients (60 males, 25 females), for whom an initial panoramic radiographic assessment had revealed that the ID canal and the lower 3rd molar were in close proximity. A CBCT scan of each patient was carried out to assess how the ID nerve canal position influenced the class and position of impaction, angulation of impaction, and bone contact. RESULTS: Class I position B impactions were found in the majority of cases, where the position of the ID canal was approximate to the lingual plate and inferior to the 3rd molar (85.7%). The results were statistically significant (p = 0.001). 96% of the ID canals showed bone contact. Of these, 77.1% of ID canals exhibited lingual bone contact, inferior to impaction. The results were statistically significant (p = 0.001). Horizontally angulated impactions were most common in the mandible, and significantly associated with lingual and inferior positioning of the ID canal (76.2%). CONCLUSIONS: Our sample population most commonly exhibited horizontally angulated class I position B impactions of the mandible.     Conceptualising and measuring levels of risk by immigration status for children in the UK

    Extensive evidence exists on how characteristics and circumstances of children shape their lifepaths and outcomes, and on the scale of resulting need. However, little research exists assessing the numbers of children who may be at risk of harm or disadvantage due to their immigration status. In this paper, we sought to establish the degree to which it is possible to monitor the aggregate vulnerability to risk of children in the UK by virtue of immigration status. First, we developed an observable set of immigration risk and vulnerability factors through workshop consultations that were analysed to produce a core set of variables that might be measured to assess aggregate need by virtue of immigration status. Second, we assessed through an administrative data review what is known statistically about the numbers of children at risk by virtue of immigration status in the UK. This research indicates a considerable gap in statistical knowledge of the level of vulnerability of children in the UK by virtue of immigration status. The approach we have developed provides a framework for future statistical work that might address this gap
