Genome-wide gene expression profiling suggests distinct radiation susceptibilities in sporadic and post-Chernobyl papillary thyroid cancers
Papillary thyroid cancers (PTCs) incidence dramatically increased in the vicinity of Chernobyl. The cancer-initiating role of radiation elsewhere is debated. Therefore, we searched for a signature distinguishing radio-induced from sporadic cancers. Using microarrays, we compared the expression profiles of PTCs from the Chernobyl Tissue Bank (CTB, n=12) and from French patients with no history of exposure to ionising radiations (n=14). We also compared the transcriptional responses of human lymphocytes to the presumed aetiological agents initiating these tumours, γ-radiation and H2O2. On a global scale, the transcriptomes of CTB and French tumours are indistinguishable, and the transcriptional responses to γ-radiation and H2O2 are similar. On a finer scale, a 118 genes signature discriminated the γ-radiation and H2O2 responses. This signature could be used to classify the tumours as CTB or French with an error of 15–27%. Similar results were obtained with an independent signature of 13 genes involved in homologous recombination. Although sporadic and radio-induced PTCs represent the same disease, they are distinguishable with molecular signatures reflecting specific responses to γ-radiation and H2O2. These signatures in PTCs could reflect the susceptibility profiles of the patients, suggesting the feasibility of a radiation susceptibility test
Cerebral gene expression in response to single or combined gestational exposure to methylmercury and selenium through the maternal diet
Controversy remains regarding the safety of consuming certain types of seafood, particularly during pregnancy. While seafood is rich in vital nutrients, it may also be an important source of environmental contaminants such as methylmercury (MeHg). Selenium (Se) is one essential element present in seafood, hypothesised to ameliorate MeHg toxicity. The aim of the present study was to ascertain the impact of Se on MeHg-induced cerebral gene expression in a mammalian model. Microarray analysis was performed on brain tissue from 15-day-old mice that had been exposed to MeHg throughout development via the maternal diet. The results from the microarray analysis were validated using qPCR. The exposure groups included: MeHg alone (2.6 mg kg−1), Se alone (1.3 mg kg−1), and MeHg + Se. MeHg was presented in a cysteinate form, and Se as Se–methionine, one of the elemental species occurring naturally in seafood. Eight genes responded to Se exposure alone, five were specific to MeHg, and 63 were regulated under the concurrent exposure of MeHg and Se. Significantly enriched functional classes relating to the immune system and cell adhesion were identified, highlighting potential ameliorating mechanisms of Se on MeHg toxicity. Key developmental genes, such as Wnt3 and Sparcl1, were also identified as putative ameliorative targets. This study, utilising environmentally realistic forms of toxicants, delivered through the natural route of exposure, in association with the power of transcriptomics, highlights significant novel information regarding putative pathways of selenium and MeHg interaction in the mammalian brain
Desempenho produtivo de tambacus alimentados com dietas suplementadas com vitamina E
O objetivo deste trabalho foi avaliar os efeitos da suplementação da dieta de tambacus com concentrações de vitamina E sobre variáveis de desempenho e hematológicas, bem como determinar a relação da atividade da glutationa peroxidase com a inclusão de vitamina E e selênio na dieta. Foram utilizados 250 juvenis de tambacu, divididos em 25 aquários. A dieta basal constituiu-se de ração peletizada com 32% de proteÃna bruta e 3.300 kcal kg-1 de energia digestÃvel, e inclusão de 0,40 mg kg-1 de Se. As dietas-teste - isoprotéicas e isoenergéticas - foram compostas da dieta basal com diferentes concentrações de vitamina E (0, 100, 200, 300, 400 mg kg-1), em delineamento inteiramente ao acaso, com cinco tratamentos e cinco repetições. Não foi observado efeito significativo da suplementação com vitamina E sobre o comprimento total, volume corpuscular médio, concentração de hemoglobina corpuscular média e número de eritrócitos, nem sobre a atividade da glutationa peroxidase. Peso final e conversão alimentar aparente, no entanto, sofreram efeito dos tratamentos suplementados. O teor de 400 mg kg-1 de vitamina E melhorou o comprimento padrão e ganho de peso. A suplementação da dieta com vitamina E resulta em melhor desempenho produtivo de tambacus
