Influência da concentração de NaCl e pH na extração de ricina em torta de mamona (Ricinus communis L.) e sua caracterização por eletroforese.
A mamoneira (Ricinus communis L.) é uma oleaginosa de alto valor econômico pelo fato de apresentar um mercado bem definido para o óleo extraído de suas sementes. A torta, que é um resíduo desta extração, se destaca pelo alto teor em proteínas. Dentre as proteínas encontradas na torta destaca-se a ricina, uma citotoxina, que inviabiliza sua utilização como fonte protéica alternativa para alimentação animal. O presente trabalho tem como objetivo identificar um melhor tratamento experimental para a extração de ricina da torta de mamona, visando futuros estudos de perda de integridade da ricina, o que garantiria a inocuidade do produto. Para tanto, buscou-se identificar a solução de maior capacidade de extração de proteínas, empregando a metodologia de superfície de resposta. Um delineamento composto central rotacional foi elaborado a fim de verificar o melhor pH e concentração de NaCl para a extração. Dos cinco diferentes valores de pH (4,0; 4,6; 6,0; 7,4; 8,0) e concentração de NaCl (0,0M; 0,3M; 1,0M; 1,7M; 2,0M) utilizados, o tratamento associando fosfato de potássio 0,2M/NaCl 1,7M pH 7,4 foi escolhido como o melhor. A concentração de proteína extraída neste tratamento chegou a valores quatro vezes maiores que o encontrado no de mínima extração de proteína. Pela evidenciação do gel de eletroforese não houve extração preferencial de ricina nos tratamentos testados, entretanto etapas de purificação usando diálise e precipitação com sulfato de amônio, permitiram uma evidenciação melhor das duas cadeias polipeptídicas de ricina
Constructive and destructive use of compilers in elliptic curve cryptography
Although cryptographic software implementation is often performed by expert programmers, the range of performance and security driven options, as well as more mundane software engineering issues, still make it a challenge. The use of domain specific language and compiler techniques to assist in description and optimisation of cryptographic software is an interesting research challenge. In this paper we investigate two aspects of such techniques, focusing on Elliptic Curve Cryptography (ECC) in particular. Our constructive results show that a suitable language allows description of ECC based software in a manner close to the original mathematics; the corresponding compiler allows automatic production of an executable whose performance is competitive with that of a hand-optimised implementation. In contrast, we study the worrying potential for naïve compiler driven optimisation to render cryptographic software insecure. Both aspects of our work are set within the context of CACE, an ongoing EU funded project on this general topic

Use of response surface methodology to optimization of extraction of enzymes from pineapple pulp.
The enzymatic reactions are very important for food. They are responsíble for the formation of highly desirable cornpound, but can cause disorders and undesirable consequences. The physiological behaviour of vegetables and fruits is already known, so that most of its deterioration changes in flavour, colour and nutritional value are caused by enzymes of the Oxidoreductases group, mainly peroxidases (PER) and polyphenol oxidase (PPO); The degradation of pectic polysaccharides involves two important enzymatic processed whose action is due to pectin methytesterase (PME) and po/ygalacturonase (PG) enzymes which affect the consistency and texture of fruit during ripening and postharvest handling. Pineapp/e is a tropical fruit very appreciated by consumers, but its physical-chemical and biochemical composition varies according to the time and place of production, and maturity stage. In order to optimize the extraction process of enzymes PER, PPO, PME and PG from pineapple pulp (ev. Peróla) the methodology of response surface was used (STATISTICA 7.0) to study the effect of pH (from 4.0 to 8.0) and NaCI concentration (from 0.0 to 2.0 M) of lhe buffer solulion used in lhe extraction of lhe enzymes. The results showed that the best conditions for enzymes extraction were 1.0 M NaCl, pH 6.0 for PER (4752.9 UI g), 1.0 M NaCI and pH 3.5 for PME (5.8 UI g), 2.0 M NaCI and pH 7.4 for PG (0,000034 Ulg.h·l) and 2.0 M NaCI and pH 4.0 for PPO (81.6 UI g).Disponível em: Acesso em: 16 fev. 2011. Elongational Viscosity of Monodisperse and Bidisperse Polystyrene Melts
Submitted to J. Rheol.The startup and steady uniaxial elongational viscosity have been measured for two monodisperse polystyrene melts with molecular weights of 52 kg/mole and 103 kg/mole, and for three bidisperse polystyrene melts. The monodisperse melts show a maximum in the steady elongational viscosity vs. the elongational rate, ε, of about two times 3η_0 whereas the bidisperse melts have a maximum of up to a factor of seven times the Trouton limit of 3η_0. The Wiest model which incorporates anisotropic drag and finite extensibility may be used to interpret the results in molecular terms.Class of 1960 Fellowship Fund

Purification, characterization and structural determination of UDP-N-acetylglucosamine pyrophosphorylase produced by Moniliophthora perniciosa
The enzyme UDP-N-acetylglucosamine pyrophosphorylase (PyroMp) from Moniliophthora perniciosa (CCMB 0257), a pathogenic fungal strain and the causative agent of the witches' broom disease in Theobroma cacao, was partially purified by precipitation with ammonium sulfate and gel filtration on Sephacryl S-200. The buffer for enzyme extraction was sodium phosphate, 0.050 mol L-1, pH 7.0, containing 1.0 mol L-1 NaCl. Response surface methodology (RSM) was used to determine the optimum pH and temperature conditions. Four different isoenzymes (PyroMp I, PyroMp II, PyroMp III and PyroMp IV) were obtained with optimal pH ranging from 6.9-8.4 and optimum temperature ranging from 28 to 68 °C. The 3D structure of pyrophosphorylase of M. perniciosa was determined by comparative modeling. The model obtained showed a good quality, possessing 78.6% of amino acids in energetically allowed regions. The model was then submitted for DM simulation and showed a good geometric quality (91.1% Ramachandran plot). The active site of the enzyme was found to be extremely well conserved.
