    Drawing-Based Automatic Dementia Screening Using Gaussian Process Markov Chains

    Screening tests play an important role for early detection of dementia. Among those widely used screening tests, drawing tests have gained much attention in clinical psychology. Traditional evaluation of drawing tests totally relies on the appearance of drawn picture, but does not consider any time-dependent behaviour. We demonstrated that the processing speed and direction can reflect the decline of cognitive function, and thus may be useful for disease screening. We proposed a model of Gaussian process Markov chains (GPMC) to study the complex associations within the drawing data. Specifically, we modeled the process of drawing in a state-space form, where a drawing state is composed of drawing direction and velocity with consideration of the processing time. For temporal modeling, our scope focused more on discrete-time Markov chains on continuous state space. Because of the short processing time of picture drawing, we applied higher-order of Markov chains to model long-term temporal correlation across drawing states. Gaussian process regression was used for universal function approximation to flexibly infer the state transition function. With Gaussian process prior to the distribution of function space, we could encode high-level function properties such as noisiness, smoothness and periodicity. We also derived an efficient training mechanism for complex Gaussian process regression on bivariate Markov chains. With GPMC, we present an optimal decision rule based on Bayesian decision theory. We applied our proposed method to a drawing test for dementia screening, i.e. interlocking pentagon-drawing test. We tested our models with 256 subjects who are aged from 65 to 95. Finally, comparing to the traditional methods, our models showed remarkable improvement in drawing test for dementia screening

    Dose-dependent and combined effects of N-methyl-D-aspartate receptor antagonist MK-801 and nitric oxide synthase inhibitor nitro-L-arginine on the survival of retinal ganglion cells in adult hamsters

    This study investigated the effects of daily intraperitoneal injections of N-methyl-D-aspartate receptor antagonist MK-801 and nitric oxide synthase inhibitor nitro-L-arginine (L-NA) on the survival of retinal ganglion cells (RGCs) at 1 and 2 weeks after unilateral optic nerve transection in adult hamsters. The left optic nerves of all animals were transected intraorbitally 1 mm from the optic disc and RGCs were retrogradely labeled with Fluorogold before they received different daily dosages of single MK-801 or L-NA as well as daily combinational treatments of these two chemicals. All experimental and control animals survived for 1 or 2 weeks after optic nerve transection. Our results revealed that the mean numbers of surviving RGCs increased and then decreased when the dosage of MK-801 (1.0, 3.0 and 4.5 mg/kg) and L-NA (1.5, 3.0, 4.5 and 6.0 mg/kg) increased at both 1 and 2 weeks survival time points. Daily combinational use of 1.0 mg/kg MK-801 and 1.5 mg/kg L-NA lead to a highest RGC number that was even higher than the sum of the RGC numbers in 1.0 mg/kg MK-801 and 1.5 mg/kg L-NA subgroups at 2 weeks. These findings indicated that both MK-801 and L-NA can protect axotomized RGCs in a dose-dependent manner and combinational treatment of these chemicals possesses a potentiative and protective effect.published_or_final_versio

    Characterization of Chlorinated Aliphatic Hydrocarbons and Environmental Variables in a Shallow Groundwater in Shanghai Using Kriging Interpolation and Multifactorial Analysis

    CAHs, as a cleaning solvent, widely contaminated shallow groundwater with the development of manufacturing in China's Yangtze River Delta. This study focused on the distribution of CAHs, and correlations between CAHs and environmental variables in a shallow groundwater in Shanghai, using kriging interpolation and multifactorial analysis. The results showed that the overall CAHs plume area (above DIV) was approximately 9,000 m(2) and located in the 2-4 m underground, DNAPL was accumulated at an area of approximately 1,400 m(2) and located in the 6-8m sandy silt layer on the top of the muddy silty clay. Heat-map of PPC for CAHs and environmental variables showed that the correlation between 'Fe2+' and most CAHs such as '1,1,1-TCA', '1,1-DCA', '1,1-DCE' and '% TCA' were significantly positive (p<0.001), but '% CA' and/or '% VC' was not, and 'Cl-' was significantly positive correlated with '1,1-DCA' and '1,1-DCE' (p<0.001). The PCA demonstrated that the relative proportions of CAHs in groundwater were mostly controlled by the sources and the natural attenuation. In conclusion, the combination of geographical and chemometrics was helpful to establishing an aerial perspective of CAHs and identifying reasons for the accumulation of toxic dechlorination intermediates, and could become a useful tool for characterizing contaminated sites in general.published_or_final_versio

    Diffusivity of the uncinate fasciculus in heroin users relates to their levels of anxiety

    Heroin use is closely associated with emotional dysregulation, which may explain its high comorbidity with disorders such as anxiety and depression. However, the understanding of the neurobiological etiology of the association between heroin use and emotional dysregulation is limited. Previous studies have suggested an impact of heroin on diffusivity in white matter involving the emotional regulatory system, but the specificity of this finding remains to be determined. Therefore, this study investigated the association between heroin use and diffusivity of white matter tracts in heroin users and examined whether the tracts were associated with their elevated anxiety and depression levels. A sample of 26 right-handed male abstinent heroin users (25 to 42 years of age) and 32 matched healthy controls (19 to 55 years of age) was recruited for this study. Diffusion tensor imaging data were collected, and their levels of anxiety and depression were assessed using the Hospital Anxiety and Depression Scale. Our findings indicated that heroin users exhibited higher levels of anxiety and depression, but the heroin use-associated left uncinate fasciculus was only related to their anxiety level, suggesting that association between heroin and anxiety has an incremental organic basis but that for depression could be a threshold issue. This finding improves our understanding of heroin addiction and its comorbid affective disorder and facilitates future therapeutic development.published_or_final_versio

    Hormonal regulation of endometrial olfactomedin expression and its suppressive effect on spheroid attachment onto endometrial epithelial cells

    Background Olfactomedin (Olfm) is a member of a diverse group of extracellular matrix proteins important for neuronal growth. Recent microarray studies identified Olfm as one of the down-regulated transcripts in receptive endometrium at the time of embryo attachment and implantation. However, the underlying molecular mechanisms that govern Olfm expression and its effect on embryo attachment and implantation remain unknown. Methods The expression of Olfm in the human endometrium was investigated by real-time PCR, western blotting and immunohistochemistry on human endometrial biopsies from natural and ovarian stimulated cycles. To investigate the function of Olfm in trophoblastendometrial cell attachment, an in vitro spheroid-endometrial cell co-culture study was performed. Results Human endometrial Olfactomedin-1 and -2(Olfm-1 and -2) transcripts decreased significantly from the proliferative to the secretory phases of the menstrual cycle. Olfm protein was strongly expressed in the luminal and glandular epithelium and moderately in the stromal cells of human endometria. Ovarian stimulation significantly decreased (P < 0.05) the expression of endometrial Olfm-1 and -2 transcripts in patients receiving IVF treatment when compared with those in the natural cycle. Importantly, recombinant Olfm-1 suppressed JAr spheroid attachment onto Ishikawa cells and this was not associated with changes of β-catenin and E-cadherin expression in trophoblast and endometrial cells.     Reinforcement learning based routing for energy sensitive wireless mesh IoT networks

    With the huge growth of the Internet of Things (IoT) in manufacturing, agricultural and numerous other applications, connectivity solutions have become increasingly important especially for those covering wide remote area in the scale of kilometre squares. Although many low-power wide-area network (LPWAN) technologies such as Long Range are supposed to support long-range low-power wireless communication, the underneath star topology limits the scalability of the networks due to the need of a central hub. To provide connectivity to a wider area, the authors propose to build the mesh topology upon these LPWAN technologies. One of the challenges of meshing these networks is the routing mechanism originally designed for star networks is not energy sensitive. In this Letter, the authors address this issue by proposing a distributed as well as energy-efficient reinforcement learning based routing algorithm for the wide area wireless mesh IoT networks. They evaluate the failure rate, spectrum and power efficiencies of the proposed algorithm by simulations, which resemble the long-range IoT networks, by comparing it to that of a random routing with loop-detection algorithm and a centralised pre-programmed routing algorithm which represents the ideal scenario. They also present a progressive study to demonstrate how the learning in the algorithm reduces the power consumption of the entire network
