Proton decoupling and recoupling under double-nutation irradiation in solid-state NMR.
The effect of (1)H decoupling in magic-angle spinning solid-state NMR is studied under radiofrequency irradiation causing simultaneous nutations around a pair of orthogonal axes. Double-nutation with an arbitrary pair of nutation frequencies is implemented through modulation of the amplitude, phase, and frequency of the transmitting pulses. Similarity and difference of double-nutation decoupling and two-pulse phase-modulation decoupling schemes [A. E. Bennett, C. M. Rienstra, M. Auger, K. V. Lakshmi, and R. G. Griffin, J. Chem. Phys. 103, 6951-6958 (1995) and I. Scholz, P. Hodgkinson, B. H. Meier, and M. Ernst, J. Chem. Phys. 130, 114510 (2009)] are discussed. The structure of recoupling bands caused by interference of the (1)H spin nutation with sample spinning is studied by both experiments and numerical simulations
Terminal and bridging fluorine ligands in TiF₄ as studied by ¹⁹F NMR in solids
To examine bonding nature of fluorine ligands in a metal coordinated system, ¹⁹F high-resolution solid-state NMR has been applied to TiF₄, which bears both bridging and terminal fluorines. Observed 12 isotropic signals are assigned to 12 crystallographically different fluorines (6 terminal and 6 bridging fluorines) in TiF₄ by referring to the calculated isotropic shifts using density functional theory (DFT). The isotropic chemical shift (δiso) for terminal F (FT) appears at high frequency (420–480 ppm from δ(CCl3F) = 0 ppm) with large shielding anisotropy Δσ ∼ 850 ppm. Whereas the δiso and Δσ values for bridging F (FB) are moderate; δiso ∼ 0–25 ppm and Δσ ∼ 250 ppm. The origin of the observed high-frequency shift for FT is ascribed to the second-order paramagnetic shift with increased covalency, shorter Ti–F bonds, and smaller energy difference between the occupied and vacant orbitals. Examination of the orientation of the shielding tensor relative to the molecular structure shows that the most deshielded component of the shielding tensor is oriented along the Ti–F bond. The characteristic orientation is consistent with a Ti–F σ bond formed by dYZ of Ti and pz of F. Further, we show that the selectively observed spinning sideband patterns and the theoretical patterns with the calculated Δσ and η (shielding asymmetry) values are not consistent with each other for FB, indicating deficiency of the present DFT calculation in evaluating Δσ
On the cohomology of pseudoeffective line bundles
The goal of this survey is to present various results concerning the
cohomology of pseudoeffective line bundles on compact K{\"a}hler manifolds, and
related properties of their multiplier ideal sheaves. In case the curvature is
strictly positive, the prototype is the well known Nadel vanishing theorem,
which is itself a generalized analytic version of the fundamental
Kawamata-Viehweg vanishing theorem of algebraic geometry. We are interested
here in the case where the curvature is merely semipositive in the sense of
currents, and the base manifold is not necessarily projective. In this
situation, one can still obtain interesting information on cohomology, e.g. a
Hard Lefschetz theorem with pseudoeffective coefficients, in the form of a
surjectivity statement for the Lefschetz map. More recently, Junyan Cao, in his
PhD thesis defended in Grenoble, obtained a general K{\"a}hler vanishing
theorem that depends on the concept of numerical dimension of a given
pseudoeffective line bundle. The proof of these results depends in a crucial
way on a general approximation result for closed (1,1)-currents, based on the
use of Bergman kernels, and the related intersection theory of currents.
Another important ingredient is the recent proof by Guan and Zhou of the strong
openness conjecture. As an application, we discuss a structure theorem for
compact K{\"a}hler threefolds without nontrivial subvarieties, following a
joint work with F.Campana and M.Verbitsky. We hope that these notes will serve
as a useful guide to the more detailed and more technical papers in the
literature; in some cases, we provide here substantially simplified proofs and

Site-specific perturbations of alpha-synuclein fibril structure by the Parkinson's disease associated mutations A53T and E46K.

These intracellular inclusions are largely composed of misfolded α-synuclein (AS), a neuronal protein that is abundant in the vertebrate brain. Point mutations in AS are associated with rare, early-onset forms of PD, although aggregation of the wild-type (WT) protein is observed in the more common sporadic forms of the disease. Here, we employed multidimensional solid-state NMR experiments to assess A53T and E46K mutant fibrils, in comparison to our recent description of WT AS fibrils. We made de novo chemical shift assignments for the mutants, and used these chemical shifts to empirically determine secondary structures. We observe significant perturbations in secondary structure throughout the fibril core for the E46K fibril, while the A53T fibril exhibits more localized perturbations near the mutation site. Overall, these results demonstrate that the secondary structure of A53T has some small differences from the WT and the secondary structure of E46K has significant differences, which may alter the overall structural arrangement of the fibrils
Predictive values of immune indicators on respiratory failure in the early phase of COVID-19 due to Delta and precedent variants
BackgroundImmune response indicators in the early phase of COVID-19, including interferon and neutralizing responses against SARS-CoV-2, which predict hypoxemia remains unclear.MethodsThis prospective observational study recruited patients hospitalized with COVID-19 (before emergence of omicron variant). As the immune indicators, we assessed the serum levels of IFN-I/III, IL-6, CXCL10 and VEGF, using an ELISA at within 5 days after the onset of symptoms, and serum neutralizing responses using a pseudovirus assay. We also assessed SARS-CoV-2 viral load by qPCR using nasal-swab specimens and serum, to assess the association of indicators and viral distribution.ResultsThe study enrolled 117 patients with COVID-19, of which 28 patients developed hypoxemia. None received vaccine before admission. Serum IFN-I levels (IFN-α and IFN-β), IL-6, CXCL10, LDH and CRP were significantly higher in patients who developed hypoxemia. A significant association with nasopharyngeal viral load was observed only for IFN-I. The serum levels of IFN-α, IL-6, CXCL10 were significantly associated with the presence of RNAemia. Multivariable analysis showed higher odds ratio of IFN-α, with cut-off value of 107 pg/ml, in regard to hypoxemia (Odds ratio [OR]=17.5; 95% confidence interval [CI], 4.7-85; p<0.001), compared to those of IL-6, >17.9 pg/ml (OR=10.5; 95% CI, 2.9-46; p<0.001).ConclusionsThis study demonstrated that serum IFN-α levels in the early phase of SARS-CoV-2 infection strongly predict hypoxemic respiratory failure in a manner different from that of the other indicators including IL-6 or humoral immune response, and instead sensitively reflect innate immune response against SARS-CoV-2 invasion
Fractional deuteration applied to biomolecular solid-state NMR spectroscopy
Solid-state Nuclear Magnetic Resonance can provide detailed insight into structural and dynamical aspects of complex biomolecules. With increasing molecular size, advanced approaches for spectral simplification and the detection of medium to long-range contacts become of critical relevance. We have analyzed the protonation pattern of a membrane-embedded ion channel that was obtained from bacterial expression using protonated precursors and D2O medium. We find an overall reduction of 50% in protein protonation. High levels of deuteration at Hα and Hβ positions reduce spectral congestion in (1H,13C,15N) correlation experiments and generate a transfer profile in longitudinal mixing schemes that can be tuned to specific resonance frequencies. At the same time, residual protons are predominantly found at amino-acid side-chain positions enhancing the prospects for obtaining side-chain resonance assignments and for detecting medium to long-range contacts. Fractional deuteration thus provides a powerful means to aid the structural analysis of complex biomolecules by solid-state NMR
Postmortem Analyses Unveil the Poor Efficacy of Decontamination, Anti-Inflammatory and Immunosuppressive Therapies in Paraquat Human Intoxications
studies resulting from human PQ poisonings have assessed the relationship of these therapeutic measures with PQ toxicokinetics and related histopathological lesions, these being the aims of the present study.For that purpose, during 2008, we collected human fluids and tissues from five forensic autopsies following fatal PQ poisonings. PQ levels were measured by gas chromatography-ion trap mass spectrometry. Structural inflammatory lesions were evaluated by histological and immunohistochemistry analysis. The samples of cardiac blood, urine, gastric and duodenal wall, liver, lung, kidney, heart and diaphragm, showed quantifiable levels of PQ even at 6 days post-intoxication. Structural analysis showed diffused necrotic areas, intense macrophage activation and leukocyte infiltration in all analyzed tissues. By immunohistochemistry it was possible to observe a strong nuclear factor kappa-B (NF-κB) activation and excessive collagen deposition.Considering the observed PQ levels in all analyzed tissues and the expressive inflammatory reaction that ultimately leads to fibrosis, we conclude that the therapeutic protocol usually performed needs to be reviewed, in order to increase the efficacy of PQ elimination from the body as well as to diminish the inflammatory process
