Hepcidin secretion was not directly proportional to intracellular iron-loading in recombinant-TfR1 HepG2 cells: short communication
Hepcidin is the master regulator of systemic iron homeostasis and its dysregulation is observed in several chronic liver diseases. Unlike the extracellular iron-sensing mechanisms, the intracellular iron-sensing mechanisms in the hepatocytes that lead to hepcidin induction and secretion are incompletely understood. Here, we aimed to understand the direct role of intracellular iron-loading on hepcidin mRNA and peptide secretion using our previously characterised recombinant HepG2 cells that over-express the cell-surface iron-importer protein transferrin receptor-1. Gene expression of hepcidin (HAMP) was determined by real-time PCR. Intracellular iron levels and secreted hepcidin peptide levels were measured by ferrozine assay and immunoassay, respectively. These measurements were compared in the recombinant and wild-type HepG2 cells under basal conditions at 30 min, 2 h, 4 h and 24 h. Data showed that in the recombinant cells, intracellular iron content was higher than wild-type cells at 30 min (3.1-fold, p<0.01), 2 h (4.6-fold, p<0.01), 4 h (4.6-fold, p<0.01) and 24 h (1.9-fold, p<0.01). Hepcidin (HAMP) mRNA expression was higher than wild-type cells at 30 min (5.9-fold; p=0.05) and 24 h (6.1-fold; p<0.03), but at 4 h, the expression was lower than that in wild-type cells (p<0.05). However, hepcidin secretion levels in the recombinant cells were similar to those in wild-type cells at all time-points, except at 4 h, when the level was lower than wild-type cells (p<0.01). Automated Markerless Extraction of Walking People Using Deformable Contour Models
We develop a new automated markerless motion capture system for the analysis of walking people. We employ global evidence gathering techniques guided by biomechanical analysis to robustly extract articulated motion. This forms a basis for new deformable contour models, using local image cues to capture shape and motion at a more detailed level. We extend the greedy snake formulation to include temporal constraints and occlusion modelling, increasing the capability of this technique when dealing with cluttered and self-occluding extraction targets. This approach is evaluated on a large database of indoor and outdoor video data, demonstrating fast and autonomous motion capture for walking people

Ammonia removal in anaerobic digestion by biogas stripping: an evaluation of process alternatives using a first order rate model based on experimental findings
The feasibility of biogas stripping to remove ammonia in the anaerobic digestion of source segregated food waste was investigated. It was found in batch experiments that ammonia could be removed from digestate and that the removal followed 1st order kinetics with respect to total ammonia nitrogen concentration. Increasing temperature, biogas flow rate and initial pH all increased removal rates. Using kinetic data gathered in these experiments allowed the integration of ammonia stripping with an anaerobic digestion plant to be modelled for different configurations. Four scenarios were identified: post digestion, in situ, side-stream and pre-digestion ammonia removal relating to where in the process the ammonia stripping was performed. The modelling showed that in situ ammonia removal may be best able to reduce in-digester ammonia concentrations over a wide range of organic loading rates whereas pre-digestion showed most promise in terms of application due to the flexibility to control each part of the process separately. Further experimental work is required into these scenarios to confirm their viability
Configuration control of seven-degree-of-freedom arms
A seven degree of freedom robot arm with a six degree of freedom end effector is controlled by a processor employing a 6 by 7 Jacobian matrix for defining location and orientation of the end effector in terms of the rotation angles of the joints, a 1 (or more) by 7 Jacobian matrix for defining 1 (or more) user specified kinematic functions constraining location or movement of selected portions of the arm in terms of the joint angles, the processor combining the two Jacobian matrices to produce an augmented 7 (or more) by 7 Jacobian matrix, the processor effecting control by computing in accordance with forward kinematics from the augmented 7 by 7 Jacobian matrix and from the seven joint angles of the arm a set of seven desired joint angles for transmittal to the joint servo loops of the arm. One of the kinematic functions constraints the orientation of the elbow plane of the arm. Another one of the kinematic functions minimizes a sum of gravitational torques on the joints. Still another kinematic function constrains the location of the arm to perform collision avoidance. Generically, one kinematic function minimizes a sum of selected mechanical parameters of at least some of the joints associated with weighting coefficients which may be changed during arm movement. The mechanical parameters may be velocity errors or gravity torques associated with individual joints
CONVENIENCE, ACCESSIBILITY, AND THE DEMAND FOR FAST FOOD
This study explores the growth in demand for fast food. A distinguishing characteristic of fast food is its convenience; in today's pervasive marketplace, consumers need not travel far to find a fast food outlet. This greater availability translates into a decrease in the full price of obtaining a meal, which contributes to greater consumption. Market-level data are used to estimate demand equations in two time periods, incorporating changes in availability as well as prices, income, and various demographic characteristics. Our findings show that greater availability has led to increased consumption. Failure to account for these types of marketplace changes could lead to incorrect inferences regarding the factors responsible for the industry growth.Demand and Price Analysis,
