    Requirement of the FATC domain of protein kinase Tel1 for localization to DNA ends and target protein recognition

    Two large phosphatidylinositol 3-kinase-related protein kinases (PIKKs), ATM and ATR, play a central role in the DNA damage response pathway. PIKKs contain a highly conserved extreme C-terminus called the FRAP-ATM-TRRAP-C-terminal (FATC) domain. In budding yeast, ATM and ATR correspond to Tel1 and Mec1, respectively. In this study, we characterized functions of the FATC domain of Tel1 by introducing substitution or truncation mutations. One substitution mutation, termed tel1-21, and a truncation mutation, called tel1- Î"C, did not significantly affect the expression level. The tel1-21 mutation impaired the cellular response to DNA damage and conferred moderate telomere maintenance defect. In contrast, the tel1-Î"C mutation behaved like a null mutation, conferring defects in both DNA damage response and telomere maintenance. Tel1-21 protein localized to DNA ends as effectively as wild-type Tel1 protein, whereas Tel1-Î"C protein failed. Introduction of a hyperactive TEL1-hy mutation suppressed the tel1-21 mutation but not the tel1-Î"C mutation. In vitro analyses revealed that both Tel1-21 and Tel1-Î"C proteins undergo efficient autophosphorylation but exhibit decreased kinase activities toward the exogenous substrate protein, Rad53. Our results show that the FATC domain of Tel1 mediates localization to DNA ends and contributes to phosphorylation of target proteins. © 2015 Ogi, Goto, Ghosh, et al

    The purpose of this study was to determine the effect of transformational leadership style and organizational culture on employee performance PT. Bank Rakyat Indonesia (Persero), Tbk. Regional Office Pekanbaru. This research was conducted at PT. Bank Rakyat Indonesia (Persero), Tbk. Pekanbaru Regional Office located in the Sudirman Block E Plot No.1, RT.01 / RW.01, Simpang Tiga village, subdistrict Bukit Raya.In this research method used is the method of statistical tests with SPSS, where samples were used that employees of PT. Bank Rakyat Indonesia (Persero), Tbk. Pekanbaru Regional Office and as an informant to interview is with the HRD of PT. Bank Rakyat Indonesia (Persero), Tbk. Regional Office Pekanbaru. To determine the sample using census techniques and data collection techniques with interviews and questionnaires.From the results of data analysis include validity, reliability test, t-test, f, and multiple linear regression showed that transformational leadership style and organizational culture has an influence on the performance of employees at PT. Bank Rakyat Indonesia (Persero), Tbk. Pekanbaru Regional Office with a value of 0.777 means that the influence of variable transformational leadership style and organizational culture on employee performance variables was 77.7%.     Suppression of Brillouin oscillation in transparent free-standing diamond thin films in picosecond ultrasound

    Brillouin oscillation appears in picosecond ultrasonics for a transparent specimen because of backward light scattering by moving strain pulse. Its amplitude is comparable with those of other responses, such as pulse-echo signals and through-thickness resonance, obscuring these non-Brillouin-oscillation responses. We here find that Brillouin oscillation can be suppressed in a transparent free-standing film by coating both sides with a metallic thin film of appropriate thickness and that this peculiar behavior is caused by strain pulses generated on both sides with a slight phase difference. This phenomenon allowed us to fabricate a Brillouin-oscillation-free diamond free-standing film, which showed a high capability for sensor applications.H. K. Weng, A. Nagakubo, H. Watanabe, and H.     Comparison of different methods of temporomandibular joint disc reconstruction - An animal model

Article first published online: 12 MAR 2008The optimum method of temporomandibular joint (TMJ) reconstruction has not been defined despite numerous surgical treatments and several well controlled clinical trials. Animal models offer an experimental method allowing direct comparison of standardized surgical techniques. Advanced osteoarthrosis was induced bilaterally in 12 mature merino sheep. Three months later unilateral surgical reconstruction was performed. Four sheep had discectomy alone, four discectomy with fresh TMJ disc grafts, and four discectomy with fresh auricular grafts. All three surgical methods resulted in some reversal and repair of the osteoarthritic process, with the best result being from the auricular graft, next discectomy alone, next the disc graft, with the untreated osteoarthritic joint showing the most advanced pathosis.     Detection of a climatological short break in the polar night jet in early winter and its relation to cooling over Siberia

    The polar night jet (PNJ) is a strong stratospheric westerly circumpolar wind at around 65°&thinsp;N in winter, and the strength of the climatological PNJ is widely recognized to increase from October through late December. Remarkably, the climatological PNJ temporarily stops increasing during late November. We examined this short break in terms of the atmospheric dynamical balance and the climatological seasonal march. We found that it results from an increase in the upward propagation of climatological planetary waves from the troposphere to the stratosphere in late November, which coincides with a maximum of the climatological Eliassen–Palm (EP) flux convergence in the lower stratosphere. The upward propagation of planetary waves at 100&thinsp;hPa, which is strongest over Siberia, is related to the climatological strengthening of the tropospheric trough over Siberia. We suggest that longitudinally asymmetric forcing by land–sea heating contrasts caused by their different heat capacities can account for the strengthening of the trough.</p

    Biomanufacture of nano-Pd(0) by Escherichia coli and electrochemical activity of bio-Pd(0) made at the expense of H2 and formate as electron donors

    Objective: Palladised cells of Desulfovibrio desulfuricans and Shewanella oneidensis have been reported as fuel cell electrocatalysts but growth at scale may be unattractive/costly; we have evaluated the potential of using E. coli, using H2/formate for Pd-nanoparticle manufacture. Results: Using ‘bio-Pd’ made under H2 (20 wt%) cyclic voltammograms suggested electrochemical activity of bio-NPs in a native state, attributed to proton adsorption/desorption. Bio-Pd prepared using formate as the electron donor gave smaller, well separated NPs; this material showed no electrochemical properties, and hence little potential for fuel cell use using a simple preparation technique. Bio-Pd on S. oneidensis gave similar results to those obtained using E. coli. Conclusion: Bio-Pd is sufficiently conductive to make an E. coli-derived electrochemically active material on intact, unprocessed bacterial cells if prepared at the expense of H2, showing potential for fuel cell applications using a simple one-step preparation method

    Ultrasonic Response in Nd-Fe-B Sintered Material During Elastic Deformation

    The rare earth material based on Nd-Fe-B alloy shows remarkable magnetic properties in the energy product (BH)max and coercive force. It is manufactured by pressing in a unidirectional magnetic field to align the easy axes, and then sintering, leading to a large magnetic and mechanical anisotropy between the normal and parallel directions. The material is first developed by Croat [1] and Sagawa [2] independently in 1984. Following them, many researchers [3–6] have studied the material. They concentrated the efforts on the investigation of the manufacturing process to have better magnetic properties as well as on the observation of the microstructure to understand the mechanism realizing the high coercive force. The previous studies showed that the material consists of the major phase of Nd2Fe14B grains and the boundary phase of the Nd-rich alloys. It is considered that their different melting points and different thermal expansion coefficients introduce microcracks during the cooling process after sintering
