    The effect of developmental vitamin D deficiency in male and female Sprague-Dawley rats on decision-making using a rodent gambling task

    Developmental vitamin D (DVD) deficiency is a plausible risk factor for schizophrenia that has been associated with behavioural alterations including disruptions in latent inhibition and response inhibition. The rodent gambling task (rGT) assesses risk-based decision-making, which is a key cognitive deficit observed in schizophrenia patients. The primary aim of this study was to examine risk-based decision-making in DVD-deficient and control rats on the rGT. We also evaluated the performance of female Sprague–Dawley rats on the rGT for the first time. Adult male and female Sprague–Dawley rats from control and vitamin D deficient dams were trained to perform the rGT in standard operant chambers and their performance and choice-preferences were assessed. Female rats were significantly faster to reach rGT training criteria compared with male rats and DVD-deficient rats were faster to reach training criteria than control animals. After reaching stable performance on the rGT DVD-deficient and control rats showed a significant preference for the optimal choice-option in the rGT, but there were no significant effects of sex or diet on these responses. DVD deficiency did not alter the decision-making processes on the rGT because no significant changes in choice-preferences were evident. This is the first study to demonstrate that once established, the performance of females is comparable to male Sprague–Dawley rats on the rGT

    Evaluating the propagator by the usual time-sliced manner, we use it to compute the second virial coefficient of an anyon gas interacting through the repulsive potential of the form g/r2(g>0)g/r^2 (g > 0). All the cusps for the unpolarized spin-1/2 as well as spinless cases disappear in the ω→0\omega \to 0 limit, where ω\omega is a frequency of harmonic oscillator which is introduced as a regularization method.     ADRIC: Adverse Drug Reactions In Children - a programme of research using mixed methods

    Aims To comprehensively investigate the incidence, nature and risk factors of adverse drug reactions (ADRs) in a hospital-based population of children, with rigorous assessment of causality, severity and avoidability, and to assess the consequent impact on children and families. We aimed to improve the assessment of ADRs by development of new tools to assess causality and avoidability, and to minimise the impact on families by developing better strategies for communication. Review methods Two prospective observational studies, each over 1 year, were conducted to assess ADRs in children associated with admission to hospital, and those occurring in children who were in hospital for longer than 48 hours. We conducted a comprehensive systematic review of ADRs in children. We used the findings from these studies to develop and validate tools to assess causality and avoidability of ADRs, and conducted interviews with parents and children who had experienced ADRs, using these findings to develop a leaflet for parents to inform a communication strategy about ADRs. Results The estimated incidence of ADRs detected in children on admission to hospital was 2.9% [95% confidence interval (CI) 2.5% to 3.3%]. Of the reactions, 22.1% (95% CI 17% to 28%) were either definitely or possibly avoidable. Prescriptions originating in the community accounted for 44 out of 249 (17.7%) of ADRs, the remainder originating from hospital. A total of 120 out of 249 (48.2%) reactions resulted from treatment for malignancies. Off-label and/or unlicensed (OLUL) medicines were more likely to be implicated in an ADR than authorised medicines [relative risk (RR) 1.67, 95% CI 1.38 to 2.02; p  48 hours, the overall incidence of definite and probable ADRs based on all admissions was 15.9% (95% CI 15.0 to 16.8). Opiate analgesic drugs and drugs used in general anaesthesia (GA) accounted for > 50% of all drugs implicated in ADRs. The odds ratio of an OLUL drug being implicated in an ADR compared with an authorised drug was 2.25 (95% CI 1.95 to 2.59; p < 0.001). Risk factors identified were exposure to a GA, age, oncology treatment and number of medicines. The systematic review estimated that the incidence rates for ADRs causing hospital admission ranged from 0.4% to 10.3% of all children [pooled estimate of 2.9% (95% CI 2.6% to 3.1%)] and from 0.6% to 16.8% of all children exposed to a drug during hospital stay. New tools to assess causality and avoidability of ADRs have been developed and validated. Many parents described being dissatisfied with clinician communication about ADRs, whereas parents of children with cancer emphasised confidence in clinician management of ADRs and the way clinicians communicated about medicines. The accounts of children and young people largely reflected parents’ accounts. Clinicians described using all of the features of communication that parents wanted to see, but made active decisions about when and what to communicate to families about suspected ADRs, which meant that communication may not always match families’ needs and expectations. We developed a leaflet to assist clinicians in communicating ADRs to parents. Conclusion The Adverse Drug Reactions In Children (ADRIC) programme has provided the most comprehensive assessment, to date, of the size and nature of ADRs in children presenting to, and cared for in, hospital, and the outputs that have resulted will improve the management and understanding of ADRs in children and adults within the NHS. Recommendations for future research: assess the values that parents and children place on the use of different medicines and the risks that they will find acceptable within these contexts; focusing on high-risk drugs identified in ADRIC, determine the optimum drug dose for children through the development of a gold standard practice for the extrapolation of adult drug doses, alongside targeted pharmacokinetic/pharmacodynamic studies; assess the research and clinical applications of the Liverpool Causality Assessment Tool and the Liverpool Avoidability Assessment Tool; evaluate, in more detail, morbidities associated with anaesthesia and surgery in children, including follow-up in the community and in the home setting and an assessment of the most appropriate treatment regimens to prevent pain, vomiting and other postoperative complications; further evaluate strategies for communication with families, children and young people about ADRs; and quantify ADRs in other settings, for example critical care and neonatology

    Thermal dependency of RAG1 self-association properties

    <p>Abstract</p> <p>Background</p> <p>Functional immunoglobulin and T cell receptor genes are produced in developing lymphocytes by V(D)J recombination. The initial site-specific DNA cleavage steps in this process are catalyzed by the V(D)J recombinase, consisting of RAG1 and RAG2, which is directed to appropriate DNA cleavage sites by recognition of the conserved recombination signal sequence (RSS). RAG1 contains both the active site and the RSS binding domains, although RAG2 is also required for DNA cleavage activity. An understanding of the physicochemical properties of the RAG proteins, their association, and their interaction with the RSS is not yet well developed.</p> <p>Results</p> <p>Here, we further our investigations into the self-association properties of RAG1 by demonstrating that despite the presence of multiple RAG1 oligomers, only the dimeric form maintains the ability to interact with RAG2 and the RSS. However, facile aggregation of the dimeric form at physiological temperature may render this protein inactive in the absence of RAG2. Upon addition of RAG2 at 37°C, the preferentially stabilized V(D)J recombinase:RSS complex contains a single dimer of RAG1.</p> <p>Conclusion</p> <p>Together these results confirm that the functional form of RAG1 in V(D)J recombination is in the dimeric state, and that its stability under physiological conditions likely requires complex formation with RAG2. Additionally, in future structural and functional studies of RAG1, it will be important to take into account the temperature-dependent self-association properties of RAG1 described in this study.</p

    Quantisation without Gauge Fixing: Avoiding Gribov Ambiguities through the Physical Projector

    The quantisation of gauge invariant systems usually proceeds through some gauge fixing procedure of one type or another. Typically for most cases, such gauge fixings are plagued by Gribov ambiguities, while it is only for an admissible gauge fixing that the correct dynamical description of the system is represented, especially with regards to non perturbative phenomena. However, any gauge fixing procedure whatsoever may be avoided altogether, by using rather a recently proposed new approach based on the projection operator onto physical gauge invariant states only, which is necessarily free on any such issues. These different aspects of gauge invariant systems are explicitely analysed within a solvable U(1) gauge invariant quantum mechanical model related to the dimensional reduction of Yang-Mills theory.Comment: 22 pages, no figures, plain LaTeX fil

    Risky driving or risky drivers? Exploring driving and crash histories of illegal street racing offenders

    Illegal street racing has received increased attention in recent years from road safety professionals and the media as jurisdictions in Australia, Canada, and the United States have implemented laws to address the problem, which primarily involves young male drivers. Although some evidence suggests that the prevalence of illegal street racing is increasing, obtaining accurate estimates of the crash risk of this behavior is difficult because of limitations in official data sources. Although crash risk can be explored by examining the proportion of incidents of street racing that result in crashes, or the proportion of all crashes that involve street racing, this paper reports on the findings of a study that explored the riskiness of involved drivers. The driving histories of 183 male drivers with an illegal street racing conviction in Queensland, Australia, were compared with a random sample of 183 male Queensland drivers with the same age distribution. The offender group was found to have significantly more traffic infringements, license sanctions, and crashes than the comparison group. Drivers in the offender group were more likely than the comparison group to have committed infringements related to street racing, such as speeding, "hooning," and offenses related to vehicle defects or illegal modifications. Insufficient statistical capacity prevented full exploration of group differences in the type and nature of earlier crashes. It was concluded, however, that street racing offenders generally can be considered risky drivers who warrant attention and whose risky behavior cannot be explained by their youth alone

    Fluorescence Quenching at High Quencher Concentrations

    Chemical reactions occurring in dense media at high reactant concentrations can be described by rate ‘‘constants’’ which are actually functions of concentration. We present a theoretical model in which this so‐called rate constant ‘‘renormalization’’ occurs for the specific case of fluorescence quenching in solution. We show that both the quenching and the excitation rate constants can become concentration dependent. We fit our theory to several sets of experimental data—our own and some from the literature—and show that excellent agreement is obtained by varying a single free parameter, namely, the efficiency with which a fluorophore‐quencher collision leads to a quench of the excited state

    Pole structure of the Hamiltonian ζ\zeta-function for a singular potential

    We study the pole structure of the ζ\zeta-function associated to the Hamiltonian HH of a quantum mechanical particle living in the half-line R+\mathbf{R}^+, subject to the singular potential gx−2+x2g x^{-2}+x^2. We show that HH admits nontrivial self-adjoint extensions (SAE) in a given range of values of the parameter gg. The ζ\zeta-functions of these operators present poles which depend on gg and, in general, do not coincide with half an integer (they can even be irrational).     Time-Dependent CP Asymmetries in B0 -> Ks pi0 gamma transitions

    We report measurements of CP violation parameters in B0->Ks pi0 gamma transitions based on a data sample of 535x10^6 BB pairs collected with the Belle detector at the KEKB asymmetric-energy e+ e- collider. One neutral B meson is fully reconstructed in the B0->Ks pi0 gamma mode. The flavor of the accompanying B meson is identified from its decay products. For a Ks pi0 invariant mass up to 1.8 GeV/c^2, we obtain S = -0.10 +- 0.31 +- 0.07 and A = -0.20 +- 0.20 +- 0.06. For a Ks pi0 invariant mass near the K*0(892) resonance, we obtain S = -0.32 +0.36-0.33 +-0.05 and A=-0.20 +-0.24 +-0.05.Comment: 6 pages, 3 figures, accepted by PR
