    Ascaris and Parascaris are important parasites in the family Ascarididae, large, ubiquitous intestinal-dwelling nematodes infecting all classes of vertebrates. Parasitic nematode drug resistance in veterinary medicine and drug recalcitrance in human medicine are increasing worldwide, with few if any new therapeutic classes on the horizon. Some of these parasites are zoonotic, e.g., Ascaris is passed from humans to pigs and vice versa. The development of new therapies against this family of parasites would have major implications for both human and livestock health. Here we tested the therapeutic ability of a paraprobiotic or dead probiotic that expresses the Bacillus thuringiensis Cry5B protein with known anthelmintic properties, against zoonotic Ascaris suum and Parascaris spp. This paraprobiotic, known as IBaCC, intoxicated A. suum larvae in vitro and was highly effective in vivo against intestinal A. suum infections in a new mouse model for this parasite. Fermentation was scaled up to 350 l to treat pigs and horses. Single dose Cry5B IBaCC nearly completely cleared A. suum infections in pigs. Furthermore, single dose Cry5B IBaCC drove fecal egg counts in Parascaris-infected foals to zero, showing at least parity with, and potential superiority to, current efficacy of anthelmintics used against this parasite. Cry5B IBaCC therefore represents a new, paraprobiotic One Health approach towards targeting Ascarididae that is safe, effective, massively scalable, stable, and useful in human and veterinary medicine in both the developed and developing regions of the world

    Ion exchanges in apatites. Effects on composition and properties

    The modification of the composition of apatites materials can be made by several processes corresponding to ion exchange reactions which can conveniently be adapted to current coatings and ceramics and are an alternative to the set up of new synthesis methods. In addition to high temperature thermal treatments, which allow to virtually replace partly or totally monovalent OH- anion of stoichiometric hydroxyapatite by any halogen ion or carbonate, aqueous processes corresponding to dissolution-reprecipitation reactions have also been proposed and used. The most interesting possibilities are however provided by aqueous ion exchange reactions involving nanocrystalline apatites. These apatites are characterised by the existence on the crystal surface of a hydrated layer of loosely bound mineral ions which can be easily exchanged in solution. This layer offers a possibility to trap mineral ions and possibly active molecules which can modify the apatite properties. Such processes are involved in mineralised tissues and could be used in biomaterials for the release of active mineral species

    A Behavioral Change Perspective of Maroon Soil Fertility Management in Traditional Shifting Cultivation in Suriname

    In Suriname, the Maroons have practiced shifting cultivation for generations, but now the increasing influence of modern society is causing a trend of decreasing fallow periods with potentially adverse effects for the vulnerable tropical soils. Adoption of appropriate soil fertility management (SFM) practices is currently slow. Combining methods from cultural ecology and environmental psychology, this study identifies two groups with divergent behavioral intentions which we term semi-permanent cultivators and shifting cultivators. Semi-permanent cultivators intend to practice more permanent agriculture and experiment individually with plot-level SFM. Shifting cultivators rely on traditional knowledge that is not adequate for their reduced fallow periods, but perceive constraints that prevent them practicing more permanent agriculture. Semi-permanent cultivators act as a strong reference group setting a subjective norm, yet feel no need to exchange knowledge with shifting cultivators who are in danger of feeling marginalized. Drawing on a political ecology perspective, we conclude that cultural ecological knowledge declined due to negative perceptions of external actors setting a strong subjective norm. Semi-permanent cultivators who wish to enter the market economy are most likely to adopt SFM. We conclude that any future SFM intervention must be based on an in-depth understanding of each group’s behavior, in order to avoid exacerbating processes of marginalization

    A novel theatre-based behaviour change approach for influencing community uptake of schistosomiasis control measures

    Background: Appropriate behaviour change with regard to safe water contact practices will facilitate the elimination of schistosomiasis as a public health concern. Various approaches to effecting this change have been trialled in the field but with limited sustainable outcomes. Our case study assessed the effectiveness of a novel theatre-based behaviour change technique (BCT), in combination with cohort awareness raising and capacity training intervention workshops. Methodology: Our study was carried out in Mwanza, Tanzania and Kemise, Ethiopia. We adapted the Risk, Attitude, Norms, Ability, and Self-regulation (RANAS) framework and four phases using a mixed methods approach. Participatory project phase engagement an11 qualitative formative data were used to guide the design of an acceptable, holistic intervention. Initial baseline (BL) data was collected using quantitative questionnaire surveys with 804 participants in Tanzania and 617 in Ethiopia, followed by the theatre-based BCT and capacity training intervention workshops. Post-intervention (PI) survey was carried out after six months, with a participant return rate of 65% in Tanzania and 60% in Ethiopia. Results: The intervention achieved a significant improvement in the knowledge of schistosomiasis transmission being associated with poorly managed sanitation and risky water contact. Participants in Tanzania increased their uptake of preventive chemotherapy (Male: BL:56%; PI:73%, Female: BL:43%; PI:50%). There was a significant increase in the selection of sanitation (Tanzania: BL:13%; PI:21%, Ethiopia: BL:63%; PI:90%), safe water and avoiding/minimising contact with infested waters as prevention methods in Tanzania and Ethiopia. Some of the participants in Tanzania followed on from the study by building their own latrines. Conclusions: This study showed substantial positive behaviour changes in schistosomiasis control can be achieved using theatre-based BCT intervention and disease awareness training. With appropriate sensitisation, education and stakeholder engagement approaches, community members were more open to minimising risk-associated contact with contaminated water sources and were mobilised to implement preventive measures

    Diffusion-Weighted PFGNMR Study of Molecular Level Interactions of Loops and Direct Bridges of HEURs on Latex Particles

    Viscosity building in latex coatings to provide desirable shear thinning rheological properties is a key property commercially achieved with hydrophobically modified ethylene oxide urethane (HEUR) rheology modifiers (RMs). Prior studies focused on the aqueous solution properties of HEURs, resulting in the well-known transient network model that describes solution rheology reasonably well. Relatively fewer studies have probed the molecular level interactions between the hydrophobe groups of HEUR and latex surfaces under conditions of realistic latex volume fractions and HEUR concentration. The presence of ubiquitous surfactant and oligomer molecules in the latex aqueous phase makes it difficult to detect these interactions directly for any off-the-shelf (industrial) materials. In this work, we outline the use of pulsed field gradient (PFG) NMR spectroscopy as diffusion-weighted filter to remove the signals of low molecular weight species in order to detect hydrophobe end groups and urethane linkers. This in situ approach does not have any perturbation issues that are inherent in prior methods involving centrifugation and avoids the questions raised by the use of custom pyrene hydrophobes in fluorescence spectroscopy. From this study we conclude that there are no HEUR transient network structures present in HEUR–latex composites with less than about 2% HEUR and 30% latex relevant for coatings applications. Our results explain the shear thinning rheology of latex–HEUR composites based on molecular level interactions between hydrophobe end groups and urethane linkers of HEURs and latex particles to produce HEUR PEO loops on latex and direct bridges between pairs of latex particles
