Investigating postharvest chilling injury in tomato (Solanum lycopersicum L.) fruit using magnetic resonance imaging and 5-azacytidine, a hypomethylation agent
Tomato, like most species from tropical and subtropical regions, exhibits postharvest chilling injury (PCI) when stored at low temperatures. Because of its economic importance and the functional genomics tools available, we used tomato to investigate aspects of fruit PCI development. We asked two questions: First, are there spatial-temporal differences in the development of PCI that can be detected by magnetic resonance imaging (MRI)? Here, the aim was to use a non-invasive method to study PCI progression in vivo. At mature green and breaker, the pericarp, locular tissue and columella produced distinct D-values while in contrast, there was no such differentiation in riper fruit. Although the pericarp is where most PCI symptoms are visible, this tissue showed less dynamism upon cold exposure, compared to the inner tissues as detected by MRI. This suggests the occurrence of distinct, independently modulated mechanisms contributing to the development of PCI-symptomatology. Collectively our data showed that the MRI could detect fruit ripening, its attenuation by cold, and fruit tissue-specific responses to chilling stress. The second question we asked was if epigenetic modification of the tomato genome or transcriptome influences PCI response. We examined PCI severity in fruit injected with a demethylating agent, 5- azacytidine (AZA). Two tomato genotypes exposed to varying severities of cold-stress were studied. Results suggested that AZA was able to moderate PCI in 'Micro-Tom' after 3 weeks at 2.5°C, while different patterns were observed in 'Sun Cherry' across various cold treatments. Inclination-Independent Galaxy Classification
We present a new method to classify galaxies from large surveys like the
Sloan Digital Sky Survey using inclination-corrected concentration,
inclination-corrected location on the color-magnitude diagram, and apparent
axis ratio. Explicitly accounting for inclination tightens the distribution of
each of these parameters and enables simple boundaries to be drawn that
delineate three different galaxy populations: Early-type galaxies, which are
red, highly concentrated, and round; Late-type galaxies, which are blue, have
low concentrations, and are disk dominated; and Intermediate-type galaxies,
which are red, have intermediate concentrations, and have disks. We have
validated our method by comparing to visual classifications of high-quality
imaging data from the Millennium Galaxy Catalogue. The inclination correction
is crucial to unveiling the previously unrecognized Intermediate class.
Intermediate-type galaxies, roughly corresponding to lenticulars and early
spirals, lie on the red sequence. The red sequence is therefore composed of two
distinct morphological types, suggesting that there are two distinct mechanisms
for transiting to the red sequence. We propose that Intermediate-type galaxies
are those that have lost their cold gas via strangulation, while Early-type
galaxies are those that have experienced a major merger that either consumed
their cold gas, or whose merger progenitors were already devoid of cold gas

Starburst-driven superwinds from infrared galaxies
New data is presented that indicate that strong far infrared galaxies commonly have largescale emission line nebulae whose properties are suggestive of mass outflows (superwinds), presumably driven by the high supernova rate associated with the central starburst. These data include longslit spectra of M82 which show that the radial variation of the gas pressure in the emission line nebula is in excellent agreement with a previous wind model. The M82 nebula also has a LINER spectrum, consistent with shock heating. Morphologically and spectroscopically similar emission line nebulae were found in NGC253, and Arp 220 and NGC6240. A longslit spectroscopic investigation was conducted of 20 additional very powerful far-infrared galaxies and found that they generally have spatially extended emission line nebulae whose spectra closely resemble that of the M82 nebula. If the superwind interpretation is correct, it could have many important consequences in extragalactic astronomy
Physical activity patterns in a nationally representative sample of adults in Ireland

DOI: 10.1079/PHN2001192Objective To evaluate habitual levels of physical activity in a nationally representative sample of adults in Ireland. Design Cross-sectional survey using a self-administered questionnaire. Usual levels of work, recreational and household activities were evaluated in relation to anthropometric, demographic and socio-economic characteristics. The amount and intensity of all activities were quantified by assigning metabolic equivalents (METS) to each activity. Setting Republic of Ireland and Northern Ireland, 1997–1999. Subjects Random sample of 1379 adults aged 18–64 years. Results Men were approximately twice as active in work and recreational activity (139.7 ± 83.9 METS) as women (68.5 ± 49.8 METS; P 28kg m−2) or obese (BMI > 30kg m−2). Fewer obese subjects reported higher levels of work and leisure activities. However, a higher percentage of obese women reported participation in the higher levels of household activities. Participation rates in recreational activities were low. Walking was the most important leisure activity of both men (41%) and women (60%). In terms of hours per week spent in vigorous physical activity, men were more active than women, professional and skilled non-manual women were more active than women in other social classes, and younger subjects (aged 18–35 years) were more active than older subjects. Conclusions The holistic approach used in the assessment of physical activity in this study has revealed important and subtle differences in the activity patterns of men and women. Failure to fully characterise the respective activity patterns of men and women could lead to ill-informed public health policy aimed at promoting and sustaining lifetime habits of physical activity. The results suggest that simple population-focused programmes to promote physical activity are unlikely to offer the same chance of long-term success as more sensitive and individualised strategies.Peer reviewe
miR-375 gene dosage in pancreatic β-cells: implications for regulation of β-cell mass and biomarker development
MicroRNAs play a crucial role in the regulation of cell growth and differentiation. Mice with genetic deletion of miR-375 exhibit impaired glycemic control due to decreased β-cell and increased α-cell mass and function. The relative importance of these processes for the overall phenotype of miR-375KO mice is unknown. Here, we show that mice overexpressing miR-375 exhibit normal β-cell mass and function. Selective re-expression of miR-375 in β-cells of miR-375KO mice normalizes both, α- and β-cell phenotypes as well as glucose metabolism. Using this model, we also analyzed the contribution of β-cells to the total plasma miR-375 levels. Only a small proportion (≈1 %) of circulating miR-375 originates from β-cells. Furthermore, acute and profound β-cell destruction is sufficient to detect elevations of miR-375 levels in the blood. These findings are supported by higher miR-375 levels in the circulation of type 1 diabetes (T1D) subjects but not mature onset diabetes of the young (MODY) and type 2 diabetes (T2D) patients. Together, our data support an essential role for miR-375 in the maintenance of β-cell mass and provide in vivo evidence for release of miRNAs from pancreatic β-cells. The small contribution of β-cells to total plasma miR-375 levels make this miRNA an unlikely biomarker for β-cell function but suggests a utility for the detection of acute β-cell death for autoimmune diabetes
Dust-forming molecules in VY Canis Majoris (and Betelgeuse)
The formation of inorganic dust in circumstellar environments of evolved
stars is poorly understood. Spectra of molecules thought to be most important
for the nucleation, i.e. AlO, TiO, and TiO2, have been recently detected in the
red supergiant VY CMa. These molecules are effectively formed in VY CMa and the
observations suggest that non-equilibrium chemistry must be involved in their
formation and nucleation into dust. In addition to exploring the recent
observations of VY CMa, we briefly discuss the possibility of detecting these

Pure rotational spectra of TiO and TiO_2 in VY Canis Majoris
We report the first detection of pure rotational transitions of TiO and TiO_2
at (sub-)millimeter wavelengths towards the red supergiant VY CMa. A rotational
temperature, T_rot, of about 250 K was derived for TiO_2. Although T_rot was
not well constrained for TiO, it is likely somewhat higher than that of TiO_2.
The detection of the Ti oxides confirms that they are formed in the
circumstellar envelopes of cool oxygen-rich stars and may be the "seeds" of
inorganic-dust formation, but alternative explanations for our observation of
TiO and TiO_2 in the cooler regions of the envelope cannot be ruled out at this
time. The observations suggest that a significant fraction of the oxides is not
converted to dust, but instead remains in the gas phase throughout the outflow.Comment: to appear in Astronomy and Astrophysic
