    PROTECTIVE EFFECT OF SAPONIN OF MOMORDICA CYMBALARIA FENZL ON HIGH-GLUCOSE INDUCED NEUROPATHY IN NB-41A3 MOUSE NEUROBLASTOMA CELLS

    Objective: The natural product Momordica cymbalaria Fenzl has been in use considerably because of its antidiabetic activity. In this work, we have investigated the protective action of one of its phytoconstituent, a saponin, in peripheral neuropathy–a diabetic complication.Methods: The saponin was isolated and identified by preparative high-performance liquid chromatography (preparative HPLC) and thin layer chromatography TLC). High glucose (56 mM)-mediated neuropathy was induced in NB-41A3 mouse neuroblastoma cells and the neuroprotective activity of the saponin was assessed by evaluating its effect on the two polyol pathway enzymes–aldose reductase (AR) and sorbitol dehydrogenase (SDH). The accumulation of intracellular sorbitol, activity of Na+K+-ATPase and production of pro-inflammatory cytokines IL-6, IL-1β and TNF-α were also investigated.Results: The results showed a significant reduction in AR activity and intracellular accumulation of sorbitol on saponin treatment. Improvement in Na+K+-ATPase activity was evident with a noteworthy reduction in IL-6, IL-1β and TNF-α production.Conclusion: These results suggest that the saponin possesses neuroprotective activity in diabetic peripheral neuropathy.Keywords: Diabetic peripheral neuropathy (DPN), Saponin of Momordica cymbalaria (SMC), Neuroblastoma cells, Aldose reductase, Sorbitol dehydrogenase, Na+K+-ATPas

    Antiangiogenic and anticancer activity of saponins of Momordica cymbalaria

    Background: The aim of the current study was to evaluate invitro anticancer activity of saponins of MC on EAC cells by using cytotoxicity (MTT) assay. To evaluate in vivo antiangiogenic potential of saponins of MC on rat air sac angiogenesis, EAC induced peritoneal angiogenesis, CAM angiogenesis.Methods: MTT assay was carried out at different concentrations of saponins of MC in 12 microliter plates containing media with EAC cells. In rat air sac angiogenesis, carrageenin was injected (s.c.) into the air sac. Dexamethasone, indomethacin, saponins of MC was administered to identify the angiogenic activity. In EAC induced angiogenesis in peritoneum, EAC cells were administered through i.p in mice peritoneum. 5-fluoro uracil, (i.p) and saponins of MC (orally) was given to identify angiogenic activity. In CAM angiogenesis, erythropoietin was given to eggs on 8th day of incubation. saponins were given on the 12th day for two days to observe the antiangiogenic activity.Results: The observed cytotoxic effects of saponins of MC on EAC cells find statistically significant. There is significant reduction in vascular branching in rat air sac model; EAC induced peritoneal angiogenesis, CAM model by the saponins of MC.Conclusions: Due to lack of certain records, it is envisaged that the change of medicine both discontinuation as well as addition was done because of blood glucose control, cost factor [in case of pioglitazone] as well as patient’s compliance

    Beclin-1 Expression is a Predictor of Clinical Outcome in Patients with Esophageal Squamous Cell Carcinoma and Correlated to Hypoxia-Inducible Factor (HIF)-1α Expression

    In the present study, we examined the relationship between Beclin-1 expression and HIF-1α expression in esophageal squamous cell carcinoma(ESCC). There was a loss of Beclin-1 protein expression in 33% of ESCCs. Beclin-1 expression significantly correlated with depth of invasion, lymph node metastasis and clinical stage. Among the 54 patients, The survival rate of the Beclin-1-positive group was better than that of the Beclin-1-negative group. Twenty-five of the 54 (46%) tumor specimens showed high levels of HIF-1α immunoreactivity. Beclin-1 expression was associated with HIF-1α expression. The survival rate of patients with Beclin-1-positive and HIF-1α-low tumors was significantly higher than that of the other groups. These results suggest that Beclin-1 and HIF-1α expression are important determinants of survival in ESCCs
