    Pneumocephalus after posterior fossa exploration in the sitting position

    Entrainment of air following exploration of posterior cranial fossa in the sitting position was studied in five patients. Intracranial pressure was monitored through a ventriculostomy catheter after closure of the dura. In three patients nitrous oxide was added to the breathing mixture only after the baseline intracranial pressure had stabilised following closure of the dura. A marked rise in intracranial pressure was observed immediately. A rapid decrease in intracranial pressure occurred when nitrous oxide administration was stopped. Two patients were given nitrous oxide from the beginning. No change in intracranial pressure was noted.     Flight Programs and X-ray Optics Development at MSFC

    The X-ray astronomy group at the Marshall Space Flight Center is developing electroformed nickel/cobalt x-ray optics for suborbital and orbital experiments. Suborbital instruments include the Focusing X-ray Solar Imager (FOXSI) and Micro-X sounding rocket experiments and the HERO balloon payload. Our current orbital program is the fabrication of a series of mirror modules for the Astronomical Roentgen Telescope (ART) to be launched on board the Russian-German Spectrum Roentgen Gamma Mission (SRG.) The details and status of these various programs are presented. A second component of our work is the development of fabrication techniques and optical metrology to improve the angular resolution of thin shell optics to the arcsecond-level. The status of these x-ray optics technology developments is also presented

    Genetic Linkage Map Construction in Pearl Millet Using SSR Markers

    For pearl millet production in the semi-arid tropics, Sclerospora graminicola is a major biotic constraint causes Downy mildew. The pathogen is heterothallic and frequent recombination leads to evolution of new virulent populations. Identification of resistance to new virulent isolates is a prerequisite for resistance breeding. A very good understanding of ability to manipulate oligogenic and quantitative traits is offered to plant breeders by recent advances in genetic marker technology. In the present investigation genotypic data generated for a total of 88 marker loci (39 Genomic SSRs and 49 EST SSRs) were used to construct a linkage map of the pearl millet mapping population of 188 F8 RIL progenies based on the cross ICMB 89111-P6 x ICMB 90111-P6. A skeleton linkage map of seven linkage groups with a total map length of 725.5 cM (Haldane units) was constructed using data from 74 marker loci for 188 RILs using JoinMap at LOD threshold value of 5.0 and MapMaker/Exp version 3.0b and map was drawn using Map Chart 2.2. The map length of individual linkage groups ranged from a minimum of 32.1cM (LG3) to a maximum of 140.2 cM (LG1). The average inter marker distance was 9.8 cM, with an average density of 0.102 markers/cM. The total number of mapped loci per linkage group (LG) ranged from 5 on LG3 to 23 on LG1

    Epigenetic and genetic variation at SKA2 predict suicidal behavior and post-traumatic stress disorder

    Traumatic stress results in hypothalamic pituitary adrenal (HPA) axis abnormalities and an increased risk to both suicidal behaviors and post-traumatic stress disorder (PTSD). Previous work out of our laboratory identified SKA2 DNA methylation associations with suicidal behavior in the blood and brain of multiple cohorts. Interaction of SKA2 with stress predicted suicidal behavior with ~80% accuracy. SKA2 is hypothesized to reduce the ability to suppress cortisol following stress, which is of potentially high relevance in traumatized populations. Our objective was to investigate the interaction of SKA2 and trauma exposure on HPA axis function, suicide attempt and PTSD. SKA2 DNA methylation at Illumina HM450 probe cg13989295 was assessed for association with suicidal behavior and PTSD metrics in the context of Child Trauma Questionnaire (CTQ) scores in 421 blood and 61 saliva samples from the Grady Trauma Project (GTP) cohort. Dexamethasone suppression test (DST) data were evaluated for a subset of 209 GTP subjects. SKA2 methylation interacted with CTQ scores to predict lifetime suicide attempt in saliva and blood with areas under the receiver operator characteristic curve (AUCs) of 0.76 and 0.73 (95% confidence interval (CI): 0.6–0.92, P=0.003, and CI: 0.65–0.78, P<0.0001) and to mediate the suppression of cortisol following DST (β=0.5±0.19, F=1.51, degrees of freedom (df)=12/167, P=0.0096). Cumulatively, the data suggest that epigenetic variation at SKA2 mediates vulnerability to suicidal behaviors and PTSD through dysregulation of the HPA axis in response to stress

    Phosphopantetheinyl transferase (Ppt)-mediated biosynthesis of lysine, but not siderophores or DHN melanin, is required for virulence of Zymoseptoria tritici on wheat

    Zymoseptoria tritici is the causal agent of Septoria tritici blotch (STB) disease of wheat. Z. tritici is an apoplastic fungal pathogen, which does not penetrate plant cells at any stage of infection, and has a long initial period of symptomless leaf colonisation. During this phase it is unclear to what extent the fungus can access host plant nutrients or communicate with plant cells. Several important primary and secondary metabolite pathways in fungi are regulated by the post-translational activator phosphopantetheinyl transferase (Ppt) which provides an essential co-factor for lysine biosynthesis and the activities of non-ribosomal peptide synthases (NRPS) and polyketide synthases (PKS). To investigate the relative importance of lysine biosynthesis, NRPS-based siderophore production and PKS-based DHN melanin biosynthesis, we generated deletion mutants of ZtPpt. The ?ZtPpt strains were auxotrophic for lysine and iron, non-melanised and non-pathogenic on wheat. Deletion of the three target genes likely affected by ZtPpt loss of function (Aar- lysine; Nrps1-siderophore and Pks1- melanin), highlighted that lysine auxotrophy was the main contributing factor for loss of virulence, with no reduction caused by loss of siderophore production or melanisation. This reveals Ppt, and the lysine biosynthesis pathway, as potential targets for fungicides effective against Z. tritici

    Bendable X-ray Optics for High Resolution Imaging

    Current state-of the-art for x-ray optics fabrication calls for either the polishing of massive substrates into high-angular-resolution mirrors or the replication of thin, lower-resolution, mirrors from perfectly figured mandrels. Future X-ray Missions will require a change in this optics fabrication paradigm in order to achieve sub-arcsecond resolution in light-weight optics. One possible approach to this is to start with perfectly flat, light-weight surface, bend it into a perfect cone, form the desired mirror figure by material deposition, and insert the resulting mirror into a telescope structure. Such an approach is currently being investigated at MSFC, and a status report will be presented detailing the results of finite element analyses, bending tests and differential deposition experiments

    The HEROES Balloon-Borne Hard X-Ray Telescope

    The High Energy Replicated Optics to Explore the Sun (HEROES) payload flew on a balloon from Ft. Sumner, NM, September 21-22, 2013. HEROES is sensitive from about 20-75 keV and comprises 8 optics modules (HPD approximately 33" as flown), each consisting of 13-14 nickel replicated optics shells and 8 matching Xenon-filled position-sensitive proportional counter detectors (dE/E=0.05 @ 60 keV). Our targets included the Sun, the Crab Nebula and pulsar and the black hole binary GRS 1915+105. HEROES was pointed using a day/night star camera system for astrophysical observations and a newly developed Solar Aspect System for solar observations (with a shutter protecting the star camera.) We have successfully detected the Crab Nebula. Analyses for GRS 1915+105 and the Sun are ongoing. In this presentation, I will describe the HEROES mission, the data analysis pipeline and calibrations, preliminary results, and plans for follow-on missions

    The Marshall Space Flight Center Development of Mirror Modules for the ART-XC Instrument aboard the Spectrum-Roentgen-Gamma Mission

    The Marshall Space Flight Center (MSFC) is developing x-ray mirror modules for the ART-XC instrument on board the Spectrum-Roentgen Gamma Mission under a Reimbursable Agreement between NASA and the Russian Space Research Institute (IKI.) ART-XC will consist of seven co-aligned x-ray mirror modules with seven corresponding CdTe focal plane detectors. Currently, four of the modules are being fabricated by the Marshall Space Flight Center (MSFC.) Each MSFC module consist of 28 nested Ni/Co thin shells giving an effective area of 65 sq cm at 8 keV, response out to 30 keV, and an angular resolution of 45 arcsec or better HPD. Delivery of these modules to the IKI is scheduled for summer 2013. We present a status of the ART x-ray modules development at the MSFC
