    Interviewing the Interpretive Researcher: An Impressionist Tale

    In this manuscript, we describe the use of debriefing interviews for interviewing the interpretive researcher. Further, we demonstrate the value of using debriefing questions as part of a qualitative research study, specifically, one doctoral student’s dissertation study. We describe the reflexivity process of the student in her study and the debriefing data that were coded via qualitative coding techniques. Thus, we provide an exemplar of the debriefing process and the findings that emerged as a result. We believe that our exemplar of interviewing the interpretive researcher provides evidence of an effective strategy for addressing the crises of representation and legitimation for researchers and instructors of qualitative methods courses alike

    A Critical Dialectical Pluralistic Examination of the Lived Experience of Select Women Doctoral Students

    The purpose of this study was to explore and to understand the daily life experiences of 8 women doctoral students who were in pursuit of their doctorates. A partially mixed concurrent dominant status design was utilized in this study embedded within a mixed methods phenomenological research lens and driven by a critical dialectical pluralistic philosophical stance. Specifically, these 8 students were interviewed individually to examine their lived experiences as doctoral students. The interview responses then were subjected to a sequential mixed analysis that was characterized by 2 qualitative analyses (i.e., constant comparison analysis, classical content analysis) and 1 quantitative analysis (i.e., correspondence analysis). The 2 qualitative analyses revealed the following 3 metathemes: adjustment (how these doctoral students made necessary accommodations with regard to all aspects of their lives), which comprised the themes of time management, interaction, belief, and lifestyle; encouragement (circumstances that motivated them to pursue their doctoral degrees), which comprised the themes of intrinsic motivation and extrinsic motivation; and discouragement (circumstances that demotivated them from pursuing their doctoral degrees), which comprised the themes of internal discouragement and external discouragement. The correspondence analysis revealed a fourth metatheme, namely, marital status (separating the single students from the married/divorced students), which comprised the themes of locus of motivation and locus of discouragement. Seven of these women doctoral students struggled to balance either dual roles (i.e., as doctoral students and wives/mothers, or as doctoral students and professionals) or triple roles (i.e., as doctoral students, wives/mothers, and professionals). Implications of the findings are discussed

    The purpose of this paper is to describe the systemic strategies used in marriage and family therapy relevant to interviews, via what we call the therapeutic interview process, that expand the meaning of a research study for both the counselor researcher and the participant(s). We outline the therapeutic interview process for conducting transformative - based interviews via similar strategies from a family systems perspective conceptualized by CharlΓ©s (2007). The central core of the interview process is the therapeutic conversation itself that involves the systemic whole.     A stealth health approach to dietary fibre

    Average dietary fibre intakes have increased little in the past twenty years in many countries, including the USA1. Multi-million-dollar campaigns promoting fruits, vegetables, whole grains and other foods high in fibre have delivered only small changes in diets2, and consumers have not changed from traditional staples to whole-grain options3. UK millers report that consumption of whole-wheat bread has actually declined over the past decade (P. Shewry, personal communication). In the US, white flour, which is lower in fibre than whole-wheat flour, accounts for nearly 40% of the fibre intake4. We believe that as motivating consumers to change food choices has proven difficult, changing food itself β€” a so-called stealth health approach β€” could be a useful strategy to increase fibre in the foods people choose to eat

    In Vivo Expression of MHC Class I Genes Depends on the Presence of a Downstream Barrier Element

    Regulation of MHC class I gene expression is critical to achieve proper immune surveillance. In this work, we identify elements downstream of the MHC class I promoter that are necessary for appropriate in vivo regulation: a novel barrier element that protects the MHC class I gene from silencing and elements within the first two introns that contribute to tissue specific transcription. The barrier element is located in intergenic sequences 3β€² to the polyA addition site. It is necessary for stable expression in vivo, but has no effect in transient transfection assays. Accordingly, in both transgenic mice and stably transfected cell lines, truncation of the barrier resulted in transcriptional gene silencing, increased nucleosomal density and decreased histone H3K9/K14 acetylation and H3K4 di-methylation across the gene. Significantly, distinct sequences within the barrier element govern anti-silencing and chromatin modifications. Thus, this novel barrier element functions to maintain transcriptionally permissive chromatin organization and prevent transcriptional silencing of the MHC class I gene, ensuring it is poised to respond to immune signaling

    Three Novel Downstream Promoter Elements Regulate MHC Class I Promoter Activity in Mammalian Cells

    BACKGROUND: MHC CLASS I TRANSCRIPTION IS REGULATED BY TWO DISTINCT TYPES OF REGULATORY PATHWAYS: 1) tissue-specific pathways that establish constitutive levels of expression within a given tissue and 2) dynamically modulated pathways that increase or decrease expression within that tissue in response to hormonal or cytokine mediated stimuli. These sets of pathways target distinct upstream regulatory elements, have distinct basal transcription factor requirements, and utilize discrete sets of transcription start sites within an extended core promoter. METHODOLOGY/PRINCIPAL FINDINGS: We studied regulatory elements within the MHC class I promoter by cellular transfection and in vitro transcription assays in HeLa, HeLa/CIITA, and tsBN462 of various promoter constructs. We have identified three novel MHC class I regulatory elements (GLE, DPE-L1 and DPE-L2), located downstream of the major transcription start sites, that contribute to the regulation of both constitutive and activated MHC class I expression. These elements located at the 3' end of the core promoter preferentially regulate the multiple transcription start sites clustered at the 5' end of the core promoter. CONCLUSIONS/SIGNIFICANCE: Three novel downstream elements (GLE, DPE-L1, DPE-L2), located between +1 and +32 bp, regulate both constitutive and activated MHC class I gene expression by selectively increasing usage of transcription start sites clustered at the 5' end of the core promoter upstream of +1 bp. Results indicate that the downstream elements preferentially regulate TAF1-dependent, relative to TAF1-independent, transcription
