    Reinforced carbon-carbon oxidation behavior in convective and radiative environments

    Reinforced carbon-carbon, which is used as thermal protection on the space shuttle orbiter wing leading edges and nose cap, was tested in both radiant and plasma arcjet heating test facilities. The test series was conducted at varying temperatures and pressures. Samples tested in the plasma arcjet facility had consistently higher mass loss than those samples tested in the radiant facility. A method using the mass loss data is suggested for predicting mission mass loss for specific locations on the Orbiter

    The Deductibility of Punitive Damages as an Ordinary and Necessary Business Expense: Reviving the Public Policy Doctrine

    Under the judicially created public policy doctrine, the deductibility as a business expense of a payment imposed for a statutory violation turned on whether the payment was penal, or whether it was compensatory. If penal, it was nondeductible, but if compensatory, it was deductible. Congress codified this judicial result in 1969. This Comment argues that Congress should have also codified this penal-compensatory test of deductibility in the case of payments imposed in ordinary civil suits. That is, a payment of punitive damages should not be deductible as a business expense

    Synthetic Turing protocells: vesicle self-reproduction through symmetry-breaking instabilities

    The reproduction of a living cell requires a repeatable set of chemical events to be properly coordinated. Such events define a replication cycle, coupling the growth and shape change of the cell membrane with internal metabolic reactions. Although the logic of such process is determined by potentially simple physico-chemical laws, the modeling of a full, self-maintained cell cycle is not trivial. Here we present a novel approach to the problem which makes use of so called symmetry breaking instabilities as the engine of cell growth and division. It is shown that the process occurs as a consequence of the breaking of spatial symmetry and provides a reliable mechanism of vesicle growth and reproduction. Our model opens the possibility of a synthetic protocell lacking information but displaying self-reproduction under a very simple set of chemical reactions

    Aerothermal test results from the first flight of the Pegasus air-launched space booster

    A survey of temperature measurements at speeds through Mach 8.0 on the first flight of the Pegasus air-launched booster system is discussed. In addition, heating rates were derived from the temperature data obtained on the fuselage in the vicinity of the wing shock interaction. Sensors were distributed on the wing surfaces, leading edge, and on the wing-body fairing or fillet. Side-by-side evaluations were obtained for a variety of sensor installations. Details of the trajectory reconstruction through first-stage separation are provided. Given here are indepth descriptions of the sensor installations, temperature measurements, and derived heating rates along with interpretations of the results

    Study of cost/benefit tradeoffs for reducing the energy consumption of the commercial air transportation system

    Economic studies were conducted for three general fuel conserving options: (1) improving fuel consumption characteristics of existing aircraft via retrofit modifications; (2) introducing fuel efficient derivations of existing production aircraft and/or introducing fuel efficient, current state-of-the-art new aircraft; and (3) introducing an advanced state-of-the-art turboprop airplane. These studies were designed to produce an optimum airline fleet mix for the years 1980, 1985 and 1990. The fleet selected accommodated a normal growth market by introducing somewhat larger aircraft while solving for maximum departure frequencies and a minimum load factor corresponding to a 15% investment hurdle rate. Fuel burnt per available-seat-mile flown would drop 22% from 1980 to 1990 due to the use of more fuel efficient aircraft designs, larger average aircraft size, and increased seating density.     An ABC transporter containing a forkhead-associated domain interacts with a serine-threonine protein kinase and is required for growth of Mycobacterium tuberculosis in mice

    Forkhead-associated (FHA) domains are modular phosphopeptide recognition motifs with a striking preference for phosphothreonine-containing epitopes. FHA domains have been best characterized in eukaryotic signaling pathways but have been identified in six proteins in Mycobacterium tuberculosis, the causative organism of tuberculosis. One of these, coded by gene Rv1747, is an ABC transporter and the only one to contain two such modules. A deletion mutant of Rv1747 is attenuated in a mouse intravenous injection model of tuberculosis where the bacterial load of the mutant is 10-fold lower than that of the wild type in both lungs and spleen. In addition, growth of the mutant in mouse bone marrow-derived macrophages and dendritic cells is significantly impaired. In contrast, growth of this mutant in vitro was indistinguishable from that of the wild type. The mutant phenotype was lost when the mutation was complemented by the wild-type allele, confirming that it was due to mutation of Rv1747. Using yeast two-hybrid analysis, we have shown that the Rv1747 protein interacts with the serine-threonine protein kinase PknF. This interaction appears to be phospho-dependent since it is abrogated in a kinase-dead mutant and by mutations in the presumed activation loop of PknF and in the first FHA domain of Rv1747. These results demonstrate that the protein coded by Rv1747 is required for normal virulent infection by M. tuberculosis in mice and, since it interacts with a serine-threonine protein kinase in a kinase-dependent manner, indicate that it forms part of an important phospho-dependent signaling pathway

    Structural Insights into Differences in Drug-binding Selectivity between Two Forms of Human α1-Acid Glycoprotein Genetic Variants, the A and F1*S Forms

    Human α1-acid glycoprotein (hAGP) in serum functions as a carrier of basic drugs. In most individuals, hAGP exists as a mixture of two genetic variants, the F1*S and A variants, which bind drugs with different selectivities. We prepared a mutant of the A variant, C149R, and showed that its drug-binding properties were indistinguishable from those of the wild type. In this study, we determined the crystal structures of this mutant hAGP alone and complexed with disopyramide (DSP), amitriptyline (AMT), and the nonspecific drug chlorpromazine (CPZ). The crystal structures revealed that the drug-binding pocket on the A variant is located within an eight-stranded β-barrel, similar to that found in the F1*S variant and other lipocalin family proteins. However, the binding region of the A variant is narrower than that of the F1*S variant. In the crystal structures of complexes with DSP and AMT, the two aromatic rings of each drug interact with Phe-49 and Phe-112 at the bottom of the binding pocket. Although the structure of CPZ is similar to those of DSP and AMT, its fused aromatic ring system, which is extended in length by the addition of a chlorine atom, appears to dictate an alternative mode of binding, which explains its nonselective binding to the F1*S and A variant hAGPs. Modeling experiments based on the co-crystal structures suggest that, in complexes of DSP, AMT, or CPZ with the F1*S variant, Phe-114 sterically hinders interactions with DSP and AMT, but not CPZ. © 2011 by The American Society for Biochemistry and Molecular Biology, Inc
