    Systematic design of cell membrane coating to improve tumor targeting of nanoparticles

    Cell membrane (CM) coating technology is increasingly being applied in nanomedicine, but the entire coating procedure including adsorption, rupture, and fusion is not completely understood. Previously, we showed that the majority of biomimetic nanoparticles (NPs) were only partially coated, but the mechanism underlying this partial coating remains unclear, which hinders the further improvement of the coating technique. Here, we show that partial coating is an intermediate state due to the adsorption of CM fragments or CM vesicles, the latter of which could eventually be ruptured under external force. Such partial coating is difficult to self-repair to achieve full coating due to the limited membrane fluidity. Building on our understanding of the detailed coating process, we develop a general approach for fixing the partial CM coating: external phospholipid is introduced as a helper to increase CM fluidity, promoting the final fusion of lipid patches. The NPs coated with this approach have a high ratio of full coating (similar to 23%) and exhibit enhanced tumor targeting ability in comparison to the NPs coated traditionally (full coating ratio of similar to 6%). Our results provide a mechanistic basis for fixing partial CM coating towards enhancing tumor accumulation.Peer reviewe

    Grains - a major source of sustainable protein for health

    Cereal grains are the main dietary source of energy, carbohydrates, and plant proteins world-wide. Currently, only 41% of grains are used for human consumption, and up to 35% are used for animal feed. Cereals have been overlooked as a source of environmentally sustainable and healthy plant proteins and could play a major role in transitioning towards a more sustainable food system for healthy diets. Cereal plant proteins are of good nutritional quality, but lysine is often the limiting amino acid. When consumed as whole grains, cereals provide health-protecting components such as dietary fiber and phytochemicals. Shifting grain use from feed to traditional foods and conceptually new foods and ingredients could improve protein security and alleviate climate change. Rapid development of new grain-based food ingredients and use of grains in new food contexts, such as dairy replacements and meat analogues, could accelerate the transition. This review discusses recent developments and outlines future perspectives for cereal grain use

    Increasing epidemiological and experimental data now emphasize that a diet rich in vegetables and fruits confers many health benefits. Functional products containing elevated levels of bioactive compounds are attracting considerable attention due to their potential to lower the risk of chronic diseases and their associated huge healthcare costs. On a global scale, there is an increasing demand for berries and fruits, since they are natural polyphenol-rich raw material to be incorporated into functional foods, nutraceuticals and pharmaceuticals. This is a major challenge for both industry and horticultural experts, because the content of health-promoting compounds in plants varies widely not only in different plant species, but also between cultivars. The content is also significantly affected by harvesting, storage and processing factors. This review summarizes the recent data and clarifies the main contributors of harvesting time, various storage conditions and post-harvest procedures, such as temperature management, controlled atmosphere, 1-MCP, calcium and plant activators, as ways to influence health-promoting compounds in fruits. Furthermore, the ways processing factors, e.g., enzymatic treatment, pressing, clarification, temperature, pressure and fermentation, can influence the levels of polyphenols and vitamins in berries and soft fruits will be discussed. Finally, strategies for preventing the decline of health-promoting compounds in fruits during long-term storage will be assessed in light of recent scientific progress and modern methods, which preserve the levels of polyphenols, will be highlighted

    High Pressure Processing Has Variable Effects on Protein-Related and Sensory Properties of Cold and Hot Smoked Rainbow Trout

    High pressure processing (HPP) is a state-of-the-art food preservation method that can be applied to many types of food items, including fish products. HPP has been found to affect fish protein conformation, with potentially positive effects on protein digestibility, but detrimental effects on sensory quality. In this study, cold and hot smoked rainbow trout samples (CSRT and HSRT, respectively) were treated with 200, 400, and 600 MPa at 4 °C for 3 min and tested for solubility of proteins (SP) and water holding capacity (WHC). Level of proteolysis (LP) was tested in an in vitro digestion model. A consumer panel evaluated sensory properties with the Rate-All-That-Apply method. Treatment at 200 MPa positively affected LP in HSRT, while SP and WHC were reduced in CSRT with increasing pressure. HPP also significantly affected texture, appearance, and appeal, especially of CSRT samples. Reduction in SP and WHC contributed to colour changes in CSRT treated with 600 MPa. It was concluded that HPP had more pronounced effects on physicochemical and sensory properties of CSRT than HSRT. The results highlight the importance of optimising the HPP parameters for each product type, in order to minimise the potentially negative effects on protein-related properties and sensory quality

    Intestinal Exposure to Food-Derived Protease Inhibitors: Digestion Physiology- and Gut Health-Related Effects

    Plant-derived protease inhibitors (PI), such as Bowman-Birk inhibitors and Kunitz-type inhibitors, have been suggested to negatively affect dietary protein digestion by blocking the activity of trypsin and chymotrypsin in the human gastrointestinal system. In addition, some PIs may possess proinflammatory activities. However, there is also scientific evidence on some beneficial effects of PIs, for example, gut-related anti-inflammatory and chemopreventive activities in vitro and in vivo. Some PIs are sensitive to processing and digestion; thus, their survival is an important aspect when considering their positive and negative bioactivities. The aim of this review was to evaluate the relevance of PIs in protein digestion in humans and to discuss the potential of PIs from whole foods and as purified compounds in decreasing symptoms of bowel-related conditions. Based on the reviewed literature, we concluded that while the complex interactions affecting plant protein digestibility and bioavailability remain unclear, PI supplements could be considered for targeted purposes to mitigate inflammation and gastric pain

    Oxygen-18 and carbon-13 isotopes in eCO(2) and erythrocytes carbonic anhydrase activity of Finnish prediabetic population

    Complex human physiological processes create the stable isotopic composition of exhaled carbon dioxide (eCO(2)), measurable with noninvasive breath tests. Recently, isotope-selective breath tests utilizing natural fluctuation in O-18/O-16 isotope ratio in eCO(2) have been proposed for screening prediabetic (PD) individuals. It has been suggested that O-18/O-16 fractionation patterns reflect shifts in the activity of carbonic anhydrase (CA), an enzyme involved in the metabolic changes in the PD state. To evaluate the applicability of the breath sampling method in Finnish PD individuals, breath delta values (BDVs, parts per thousand) of O-18/O-16 (delta O-18) were monitored for 120 min in real-time with a high-precision optical isotope ratio spectrometer, both in the fasting state and during a 2 h oral glucose tolerance test (2 h OGTT) with non-labeled glucose. In addition, the BDV of C-13/C-12 (delta C-13) was measured, and total erythrocyte CA activity was determined. delta O-18 and CA did not demonstrate any statistically significant differences between PD and non-diabetic control (NDC) participants. Instead, delta C-13 was significantly lower in PD patients in comparison to NDCs in the fasting state and at time points 90 and 120 min of the 2 h OGTT, thus indicating slightly better potential in identifying Finnish PD individuals. However, overlapping values were measured in PD participants and NDCs, and therefore, delta C-13 cannot be applied as a sole measure in screening prediabetes at an individual level. Thus, because the combination of environmental and lifestyle factors and anthropometric parameters has a greater effect on glucose metabolism and CA activity in comparison to the PD state, O-18/O-16 and C-13/C-12 fractionations or CA activity did not prove to be reliable biomarkers for impaired glucose tolerance in Finnish subjects.
