    Neighbourhood Socioeconomic Disparity in Exposure to Preterm Birth Risk During the Pandemic: Secondary Analysis of Pregnancy During the Pandemic Cohort

    Background: Preterm birth (PTB) is defined as a live birth before 37 weeks of gestation and remains a major public health concern, affecting an estimated 15 million births annually with a global prevalence of 11%. Socioeconomic disparities play a crucial role in PTB rates, with both individual- and neighborhood-level factors contributing. Stress response pathway has been identified as a key mechanism in the relationship between neighborhood socioeconomic status (nSES) and PTB risk. As such, the new challenges added due to the COVID-19 pandemic, such as disruption to the support networks and increased psychosocial distress for pregnant individuals, were expected to increase PTB rates. However, studies found lack of change or even decrease in the incidence during this period. It is suggested that such counter intuitive findings are due to lack of consideration for the differential exposure to the pandemic-related hardships based on nSES. Therefore, the primary aim of the present study is to test whether a measure of objective pandemic hardship and psychological distress mediate the relationship between nSES and PTB. Methods: Present study is a secondary analysis of the data collected from a prospective longitudinal cohort study, Pregnancy during the Pandemic (PdP). Two serial mediation path models with a measure of baseline objective pandemic hardship and psychological distress included as mediators between nSES and PTB/gestational age (GA) were tested. Results: In both models, the main indirect pathway of interest from nSES to pandemic objective hardship, psychological distress, then PTB/GA was non-significant with minimal effect. Secondary indirect pathway of interest from pandemic objective hardship to psychological distress then PTB/GA was significant in both models while controlling for months into the pandemic at birth and baseline sociodemographic characteristics. Discussion: The present paper is the first to test a comprehensive model of the role of nSES on PTB risk with an explicit measure of pandemic objective hardship and psychological distress included as mediators. While the main indirect serial mediation pathway was non-significant, partial support for the proposed mechanism was observed where increase in pandemic-related hardship elevated psychological distress, which then heightened the risk of PTB risk and shortened gestational age at birth

    Elevated intracellular cAMP exacerbates vulnerability to oxidative stress in optic nerve head astrocytes.

    Glaucoma is characterized by a progressive loss of retinal ganglion cells and their axons, but the underlying biological basis for the accompanying neurodegeneration is not known. Accumulating evidence indicates that structural and functional abnormalities of astrocytes within the optic nerve head (ONH) have a role. However, whether the activation of cyclic adenosine 3',5'-monophosphate (cAMP) signaling pathway is associated with astrocyte dysfunction in the ONH remains unknown. We report here that the cAMP/protein kinase A (PKA) pathway is critical to ONH astrocyte dysfunction, leading to caspase-3 activation and cell death via the AKT/Bim/Bax signaling pathway. Furthermore, elevated intracellular cAMP exacerbates vulnerability to oxidative stress in ONH astrocytes, and this may contribute to axonal damage in glaucomatous neurodegeneration. Inhibition of intracellular cAMP/PKA signaling activation protects ONH astrocytes by increasing AKT phosphorylation against oxidative stress. These results strongly indicate that activation of cAMP/PKA pathway has an important role in astrocyte dysfunction, and suggest that modulating cAMP/PKA pathway has therapeutic potential for glaucomatous ONH degeneration

    Quantitative measurements of C-reactive protein using silicon nanowire arrays

    A silicon nanowire-based sensor for biological application showed highly desirable electrical responses to either pH changes or receptor-ligand interactions such as protein disease markers, viruses, and DNA hybridization. Furthermore, because the silicon nanowire can display results in real-time, it may possess superior characteristics for biosensing than those demonstrated in previously studied methods. However, despite its promising potential and advantages, certain process-related limitations of the device, due to its size and material characteristics, need to be addressed. In this article, we suggest possible solutions. We fabricated silicon nanowire using a top-down and low cost micromachining method, and evaluate the sensing of molecules after transfer and surface modifications. Our newly designed method can be used to attach highly ordered nanowires to various substrates, to form a nanowire array device, which needs to follow a series of repetitive steps in conventional fabrication technology based on a vapor-liquid-solid (VLS) method. For evaluation, we demonstrated that our newly fabricated silicon nanowire arrays could detect pH changes as well as streptavidin-biotin binding events. As well as the initial proof-of-principle studies, C-reactive protein binding was measured: electrical signals were changed in a linear fashion with the concentration (1 fM to 1 nM) in PBS containing 1.37 mM of salts. Finally, to address the effects of Debye length, silicon nanowires coupled with antigen proteins underwent electrical signal changes as the salt concentration changed

    Cancer Statistics in Korea: Incidence, Mortality, Survival, and Prevalence in 2008

    PURPOSE: This paper overviews the nationwide cancer statistics including incidence, mortality, survival and prevalence, and their trends in Korea based on the year 2008 cancer incidence data. MATERIALS AND METHODS: Incidence data from 1993 to 2008 were obtained from the Korea National Cancer Incidence Database, and the vital status was followed through December 31, 2009. Mortality data from 1983 to 2008 were obtained from the Korea National Statistics Office. Crude rates and age-standardized rates for incidence, mortality, prevalence and relative survival were calculated. RESULTS: There were 178,816 cancer cases and 68,912 cancer deaths observed during year 2008 and 724,663 10-year cancer prevalent cases as of January 1, 2009 in Korea. The incidence rate for all cancer combined showed an annual increase of 3.1% from 1999 to 2008. CONCLUSION: With significantly increasing cancer incidence, Korea faces a large cancer burden and efficient cancer control programs are essential.ope

    The small GTPase ARF6 regulates GABAergic synapse development

    ADP ribosylation factors (ARFs) are a family of small GTPases composed of six members (ARF1-6) that control various cellular functions, including membrane trafficking and actin cytoskeletal rearrangement, in eukaryotic cells. Among them, ARF1 and ARF6 are the most studied in neurons, particularly at glutamatergic synapses, but their roles at GABAergic synapses have not been investigated. Here, we show that a subset of ARF6 protein is localized at GABAergic synapses in cultured hippocampal neurons. In addition, we found that knockdown (KD) of ARF6, but not ARF1, triggered a reduction in the number of GABAergic synaptic puncta in mature cultured neurons in an ARF activity-dependent manner. ARF6 KD also reduced GABAergic synaptic density in the mouse hippocampal dentate gyrus (DG) region. Furthermore, ARF6 KD in the DG increased seizure susceptibility in an induced epilepsy model. Viewed together, our results suggest that modulating ARF6 and its regulators could be a therapeutic strategy against brain pathologies involving hippocampal network dysfunction, such as epilepsy. © 2020 The Author(s).1

    Cancer Statistics in Korea: Incidence, Mortality and Survival in 2006-2007

    Cancer has been the leading cause of death in Korea. Korea is facing a very rapid change and increase in cancer incidence, which draws much attention in public health. This paper overviews the nationwide cancer statistics, including incidence, mortality, and survival rates, and their trends in Korea based on the cancer incidence data from The Korea Central Cancer Registry (KCCR) in year 2006 and 2007. In Korea, there were 153,237 cancer cases and 65,519, cancer deaths observed in 2006, and 161,920 cancer cases and 67,561 cancer deaths in 2007, respectively. The incidence rate for all cancer combined showed an annual increase of 2.8% from 1999 to 2007. Specifically, there was significant increase in the incidence of colorectal, thyroid, female breast, and prostate cancers. The number of cancer deaths has increased over the past two decades, due mostly to population aging, while the age-standardized mortality rates have decreased in both men and women since 2002. Notable improvement has been observed in the 5-yr relative survival rates for most major cancers and for all cancer combined, with the exception of pancreatic cancer. The nationwide cancer statistics in this paper will provide essential data for cancer research and evidence-based health policy in Korea
