    Posttraumatic Stress Disorder in women after Hurricane Katrina: predictors and symptom endorsement

    Hurricane Katrina devastated areas of New Orleans and caused the evacuation of most of the city’s residents. Many people were exposed to dangerous storms and flooding and lost many of their possessions. One of the most common psychological disorders following a disaster is Posttraumatic Stress Disorder. This study describes the PTSD symptom endorsement of a sample of women who experienced Hurricane Katrina. In addition, many of these women had previous trauma histories which are also described. Participants included 287 women from New Orleans, Jefferson and East Baton Rouge Parish recruited for a larger study on mother’s and children’s psychological functioning in the aftermath of Katrina. Participants completed the Posttraumatic Stress Diagnostic Scale and a hurricane exposure questionnaire at 4-7 months (T1) and 14-17 months (T2) post-Katrina. Trauma history, hurricane exposure and demographic variables such as race, income and education were entered into a regression analysis to predict PTSD symptom severity at Time 1. These variables and PTSD symptom severity at T1 were entered into a second regression analysis to predict PTSD symptom severity at T2. At T1, hurricane exposure, trauma history and education predicted T1 PTSD symptom severity. At T2, only T1 PTSD symptom severity was significantly predictive of T2 PTSD symptom severity. Results of the analyses and the description of symptom endorsement are discussed in light of current criticisms about the conceptualization of PTSD

    Support for Instructional Leadership: Supervision, Mentoring, and Professional Development for U.S. School Leaders - Findings from the American School Leader Panel

    With school leadership second only to teaching among school-related influences on student learning, principals can play an important role in school success. But how do their districts promote their effectiveness, especially in improving teaching? Based on a survey of the American School Leader Panel, a representative sample of principals from across the United States, this report explores the prevalence and quality of three important on-the-job supports for school leaders: supervision, mentorship and professional development (as defined by at least a day focused on principals). The good news is that two-thirds of principals report receiving some support. The bad news is that more than two thirds (68 percent) report that they don't receive all three sources of help. Mentoring, for example, is typically available only to first- or second-year principals or those encountering difficulties on the job, and only a minority of principals report that their districts require mentoring, even for first-year (49 percent of respondents) or struggling principals (21 percent). Also, the prevalence of support a principal receives may depend on the size of his or her school district. Both mentoring and professional development are more readily available in larger and medium-sized districts than smaller ones. The value principals place on the support they receive is linked to whether the support emphasizes the key aspect of principals' jobβ€”improving teachers' instruction. For example, all of the principals (100 percent) who reported that their mentors focused on instruction to a great extent also said that they prized the mentoring. That compares with a minority (40 percent) of principals who said their mentors devoted little to no time to instruction

    In situ apparatus for the study of clathrate hydrates relevant to solar system bodies using synchrotron X-ray diffraction and Raman spectroscopy

    Clathrate hydrates are believed to play a significant role in various solar system environments, e.g. comets, and the surfaces and interiors of icy satellites, however the structural factors governing their formation and dissociation are poorly understood. We demonstrate the use of a high pressure gas cell, combined with variable temperature cooling and time-resolved data collection, to the in situ study of clathrate hydrates under conditions relevant to solar system environments. Clathrates formed and processed within the cell are monitored in situ using synchrotron X-ray powder diffraction and Raman spectroscopy. X-ray diffraction allows the formation of clathrate hydrates to be observed as CO2 gas is applied to ice formed within the cell. Complete conversion is obtained by annealing at temperatures just below the ice melting point. A subsequent rise in the quantity of clathrate is observed as the cell is thermally cycled. Four regions between 100-5000cm-1 are present in the Raman spectra that carry features characteristic of both ice and clathrate formation. This novel experimental arrangement is well suited to studying clathrate hydrates over a range of temperature (80-500K) and pressure (1-100bar) conditions and can be used with a variety of different gases and starting aqueous compositions. We propose the increase in clathrate formation observed during thermal cycling may be due to the formation of a quasi liquid-like phase that forms at temperatures below the ice melting point, but which allows easier formation of new clathrate cages, or the retention and delocalisation of previously formed clathrate structures, possibly as amorphous clathrate. The structural similarities between hexagonal ice, the quasi liquid-like phase, and crystalline CO2 hydrate mean that differences in the Raman spectrum are subtle; however, all features out to 5000cm-1 are diagnostic of clathrate structure.Comment: Astronomy & Astrophysics, in press. 6 page

    Non-aqueous formation of the calcium carbonate polymorph vaterite: astrophysical implications

    We study the formation of calcium carbonate, through the solid-gas interaction of amorphous Ca-silicate with gaseous CO2, at elevated pressures, and link this to the possible presence of calcium carbonate in a number of circumstellar and planetary environments. We use in-situ synchrotron X-Ray powder diffraction to obtain detailed structural data pertaining to the formation of the crystalline calcium carbonate phase vaterite and its evolution with temperature. We found that the metastable calcium carbonate phase vaterite was formed alongside calcite, at elevated CO2 pressure, at room temperature and subsequently remained stable over a large range of temperature and pressure. We report the formation of the calcium carbonate mineral vaterite whilst attempting to simulate carbonate dust grain formation in astrophysical environments. This suggests that vaterite could be a mineral component of carbonate dust and also presents a possible method of formation for vaterite and its polymorphs on planetary surfaces.Comment: Astronomy and Astrophysics, in pres

    Routine handling methods affect behaviour of three-spined sticklebacks in a novel test of anxiety

    Fish are increasingly popular subjects in behavioural and neurobiological research. It is therefore important that they are housed and handled appropriately to ensure good welfare and reliable scientific findings, and that species-appropriate behavioural tests (e.g. of cognitive/affective states) are developed. Routine handling of captive animals may cause physiological stress responses that lead to anxiety-like states (e.g. increased perception of danger). In fish, these may be particularly pronounced when handling during tank-to-tank transfer involves removal from water into air. Here we develop and use a new combined scototaxis (preference for dark over light areas) and novel-tank-diving test, alongside conventional open-field and novel-object tests, to measure the effects of transferring three-spined sticklebacks (Gasterosteus aculeatus) between tanks using a box or net (in and out of water respectively). Preference tests for dark over light areas confirmed the presence of scototaxis in this species. Open-field and novel-object tests failed to detect any significant differences between net and box-handled fish. However, the combined diving and scototaxis detected consistent differences between the treatments. Net-handled fish spent less time on the dark side of the tank, less time in the bottom third, and kept a greater distance from the β€˜safe’ bottom dark area than box-handled fish. Possible explanations for this reduction in anxiety-like behaviour in net-handled fish are discussed. The combined diving and scototaxis test may be a sensitive and taxon-appropriate method for measuring anxiety-like states in fish

    Widespread mRNA Association with Cytoskeletal Motor Proteins and Identification and Dynamics of Myosin-Associated mRNAs in S. cerevisiae

    Programmed mRNA localization to specific subcellular compartments for localized translation is a fundamental mechanism of post-transcriptional regulation that affects many, and possibly all, mRNAs in eukaryotes. We describe her e a systematic approach to identify the RNA cargoes associated with the cytoskeletal motor proteins of Saccharomyces cerevisiae in combination with live-cell 3D super-localization microscopy of endogenously tagged mRNAs. Our analysis identified widespread association of mRNAs with cytoskeletal motor proteins, including association of Myo3 with mRNAs encoding key regulators of actin branching and endocytosis such as WASP and WIP. Using conventional fluorescence microscopy and expression of MS2-tagged mRNAs from endogenous loci, we observed a strong bias for actin patch nucleator mRNAs to localize to the cell cortex and the actin patch in a Myo3- and F-actin dependent manner. Use of a double-helix point spread function (DH-PSF) microscope allowed super-localization measurements of single mRNPs at a spatial precision of 25 nm in x and y and 50 nm in z in live cells with 50 ms exposure times, allowing quantitative profiling of mRNP dynamics. The actin patch mRNA exhibited distinct and characteristic diffusion coefficients when compared to a control mRNA. In addition, disruption of F-actin significantly expanded the 3D confinement radius of an actin patch nucleator mRNA, providing a quantitative assessment of the contribution of the actin cytoskeleton to mRNP dynamic localization. Our results provide evidence for specific association of mRNAs with cytoskeletal motor proteins in yeast, suggest that different mRNPs have distinct and characteristic dynamics, and lend insight into the mechanism of actin patch nucleator mRNA localization to actin patches

    Local structure and site occupancy of Cd and Hg substitutions in CeTIn5 (T=Co, Rh, Ir)

    The CeTIn5 superconductors (T=Co, Rh, or Ir) have generated great interest due to their relatively Tc's, NFL behavior, and their proximity to AF order and quantum critical points. In contrast to small changes with the T-species, electron doping in CeT(In{1-x}Mx)5 with M=Sn and hole doping with Cd or Hg have a dramatic effect on the electronic properties at very low concentrations. The present work reports EXAFS measurements that address the substituent atom distribution as a function of T, M, and x, near the superconducting phase. Together with previous measurements for M=Sn, the proportion of the M atom residing on the In(1) site, f{In(1)}, increases in the order M=Cd, Sn, and Hg, ranging from about 40% to 70%, showing a strong preference for these substituents to occupy the In(1) site (random=20%). In addition, f{In(1)} ranges from 70% to 100% for M=Hg in the order T=Co, Rh, and Ir. These fractions track the changes in the atomic radii of the various species, and help explain the sharp dependence of Tc on substituting into the In site. However, it is difficult to reconcile the small concentrations of M with the dramatic changes in the ground state in the hole-doped materials with only an impurity scattering model. These results therefore indicate that while such substitutions have interesting local atomic structures with important electronic and magnetic consequences, other local changes in the electronic and magnetic structure are equally important in determining the bulk properties of these materials.Comment: 10 pages, 7 figures, to appear in PR

    High-Throughput Screening Data Interpretation in the Context of In Vivo Transcriptomic Responses to Oral Cr(VI) Exposure

    The toxicity of hexavalent chromium [Cr(VI)] in drinking water has been studied extensively, and available in vivo and in vitro studies provide a robust dataset for application of advanced toxicological tools to inform the mode of action (MOA). This study aimed to contribute to the understanding of Cr(VI) MOA by evaluating high-throughput screening (HTS) data and other in vitro data relevant to Cr(VI), and comparing these findings to robust in vivo data, including transcriptomic profiles in target tissues. Evaluation of Tox21 HTS data for Cr(VI) identified 11 active assay endpoints relevant to the Ten Key Characteristics of Carcinogens (TKCCs) that have been proposed by other investigators. Four of these endpoints were related to TP53 (tumor protein 53) activation mapping to genotoxicity (KCC#2), and four were related to cell death/proliferation (KCC#10). HTS results were consistent with other in vitro data from the Comparative Toxicogenomics Database. In vitro responses were compared to in vivo transcriptomic responses in the most sensitive target tissue, the duodenum, of mice exposed to ≀ 180 ppm Cr(VI) for 7 and 90 days. Pathways that were altered both in vitro and in vivo included those relevant to cell death/proliferation. In contrast, pathways relevant to p53/DNA damage were identified in vitro but not in vivo. Benchmark dose modeling and phenotypic anchoring of in vivo transcriptomic responses strengthened the finding that Cr(VI) causes cell stress/injury followed by proliferation in the mouse duodenum at high doses. These findings contribute to the body of evidence supporting a non-mutagenic MOA for Cr(VI)-induced intestinal cancer
