    Active Flow Control at Low Reynolds Numbers on a NACA 0015 Airfoil

    Results from a low Reynolds number wind tunnel experiment on a NACA 0015 airfoil with a 30% chord trailing edge flap tested at deflection angles of 0, 20, and 40 are presented and discussed. Zero net mass flux periodic excitation was applied at the ap shoulder to control flow separation for flap deflections larger than 0. The primary objective of the experiment was to compare force and moment data obtained from integrating surface pressures to data obtained from a 5-component strain-gage balance in preparation for additional three-dimensional testing of the model. To achieve this objective, active flow control is applied at an angle of attack of 6 where published results indicate that oscillatory momentum coefficients exceeding 1% are required to delay separation. Periodic excitation with an oscillatory momentum coefficient of 1.5% and a reduced frequency of 0.71 caused a significant delay of separation on the airfoil with a flap deflection of 20. Higher momentum coefficients at the same reduced frequency were required to achieve a similar level of flow attachment on the airfoil with a flap deflection of 40. There was a favorable comparison between the balance and integrated pressure force and moment results

    Engaging schools and local communities through Raspberry Shake seismometers: a citizen science approach around the geothermal research infrastructure DeepStor in Karlsruhe, Germany

    While there is a general consensus on the importance of the energy transition, geothermal energy projects have faced challenges in recent years linked to unfavorable societal perception. The concerns often center around the perceived environmental risks, specifically focusing on potential impacts on groundwater and the likelihood of induced seismicity and subsequent damage to buildings. The controversy may arise due to insufficient information and ineffective communication strategies between project developers and local communities. This context applies to the region of Karlsruhe (Germany), where the DeepStor research infrastructure wants to explore the feasibility of High-Temperature Aquifer Thermal Energy Storage (HT-ATES). The infrastructure, which is planned at the Karlsruhe Institute of Technology Campus, holds potential in meeting climate protection goals. We present a participatory approach developed in the frame of the DeepStor project and that aims at addressing these societal challenges. The strategy is based on dialogue and transparency. It promotes collaboration between researchers and individuals from surrounding communities to engaging the citizens and foster an active involvement during infrastructure developments. A significant focus involves deploying Raspberry Shake seismometers, known for their ease-of-use and affordability, across various settings, including educational institutions, KIT research labs, and private residences. The approach seeks to increase public understanding of science and foster scientific literacy among non-specialists by allowing a direct interaction between citizens and scientists through cutting-edge technologies. In parrallel, the sensor deployment aims at increasing the density of seismic stations and enhance the spatial coverage of data collection. The experiences and lessons learned from this project provide valuable insights into participatory approaches for research infrastructures and industry projects

    Structure and activity of the Streptococcus pyogenes family GH1 6-phospho β-glycosidase, Spy1599

    The group A streptococcus Streptococcus pyogenes is the causative agent of a wide spectrum of invasive infections, including necrotizing fasciitis, scarlet fever and toxic shock syndrome. In the context of its carbohydrate chemistry, it is interesting that S. pyogenes (in this work strain M1 GAS SF370) displays a spectrum of oligosaccharide-processing enzymes that are located in close proximity on the genome but that the in vivo function of these proteins remains unknown. These proteins include different sugar transporters (SPy1593 and SPy1595), both GH125 -1,6- and GH38 -1,3-mannosidases (SPy1603 and SPy1604), a GH84 -hexosaminidase (SPy1600) and a putative GH2 -galactosidase (SPy1586), as well as SPy1599, a family GH1 `putative -glucosidase'. Here, the solution of the three-dimensional structure of SPy1599 in a number of crystal forms complicated by unusual crystallographic twinning is reported. The structure is a classical (/)8-barrel, consistent with CAZy family GH1 and other members of the GH-A clan. SPy1599 has been annotated in sequence depositions as a -glucosidase (EC 3.2.1.21), but no such activity could be found; instead, three-dimensional structural overlaps with other enzymes of known function suggested that SPy1599 contains a phosphate-binding pocket in the active site and has possible 6-phospho--glycosidase activity. Subsequent kinetic analysis indeed showed that SPy1599 has 6-phospho--glucosidase (EC 3.2.1.86) activity. These data suggest that SPy1599 is involved in the intracellular degradation of 6-phosphoglycosides, which are likely to originate from import through one of the organism's many phosphoenolpyruvate phosphotransfer systems (PEP-PTSs)
