    An Empirical Study of Civil Servants’ Lifelong E-Learning Continuance Intention

    E-learning is an effective way for job-training and continuous education. In recognizing the need for civil servants to embrace the lifelong learning to sustain competitiveness, many countries around the world have created policies to develop e-learning. This study is focus on civil servants’ e-learning continuance intention and through e-learning experience to achieve lifelong learning. Based on Information system (IS) success model proposed by Seddon (1997) and adding organizational factors (Incentive, Supervisor Support, and Technical Support) to survey civil servants’ e-learning behaviour. The sample for the study was taken from the civil servants in Taiwan who have the experience of using the lifelong e-learning websites. The results also support Seddon’s IS success model. Finally, the implications and limitations of the study are discussed

    Unmasking stem-specific broadly neutralizing epitopes by abolishing N-linked glycosylation sites for vaccine design

    Targeting highly conserved HA stem regions has been proposed as a useful strategy for designing universal influenza vaccines. The influenza virus HA stem region, consisting of a HA1 N-terminal part and full HA2 part, contains several potential sites for the addition of N-glycans. We expressed a series of recombinant HA (rHA) mutant proteins with deleted N-linked glycosylation sites in the HA1-stem and HA2-stem regions of H5N1 and pH1N1 viruses. Unmasking N-glycans in the HA2-stem region (rH5HA N484A and rH1HA N503A) did not affect the trimeric structure of HA. Immunizations using rH5HA N484A and rH1HA N503A elicited more potent neutralizing antibody titers against homologous, heterologous and heterosubtypic viruses. Unmasking the HA2-stem N-glycans of rH5HA N484A induced higher levels of stem-specific CR6261-like and FI6v3-like antibodies, improved the ability of stem-specific anti-fusion antibodies, enhanced H5 stem helix A epitope-specific B and T cell responses in splenocytes, and provided better protection against both homologous and heterosubtypic virus challenges. These findings suggest that HA2-stem N-glycan unmasking holds potential as a useful design strategy for developing more broadly protective influenza vaccines

    EBV-positive Hodgkin lymphoma is associated with suppression of p21cip1/waf1 and a worse prognosis

    <p>Abstract</p> <p>Background</p> <p>About 30-50% of Hodgkin lymphomas (HLs) harbor the Epstein-Barr virus (EBV), but the impact of EBV infection on clinical outcomes has been unclear. EBV-encoded small RNAs (<it>EBER</it>s) are presented in all EBV-infected cells, but their functions are still less understood.</p> <p>Results</p> <p><it>EBER1 </it>was transfected into two HL cell lines, KMH2 and L428, and microarrays were used to screen for <it>EBER1</it>-induced changes. We found that <it>EBER1 </it>suppressed <it>p21</it><sup>cip1/waf1 </sup>transcription in HL cell lines. In addition, positive regulators of <it>p21</it><sup>cip1/waf1 </sup>transcription, such as p53, EGR1, and STAT1, were decreased. Suppression of <it>p21</it><sup>cip1/waf1 </sup>in the <it>EBER1</it><sup>+ </sup>HL cell lines was associated with increased resistance to histone deacetylase inhibitors or proteasome inhibitors, drugs known to cause apoptosis by increasing p21<sup>cip1/waf1 </sup>levels. On biopsy specimens, EBV<sup>+ </sup>HLs had weaker expression of both p21<sup>cip1/waf1 </sup>and active caspase 3. Clinically, suppression of p21<sup>cip1/waf1 </sup>in EBV<sup>+ </sup>HLs was associated with a worse 2-year disease-free survival rate (45% for EBV<sup>+ </sup>HLs <it>vs</it>. 77% for EBV<sup>- </sup>HLs, <it>p </it>= 0.002).</p> <p>Conclusion</p> <p>Although the underlying mechanisms are still relatively unclear, <it>EBER1 </it>inhibits <it>p21</it><sup>cip1/waf1 </sup>transcription and prevents apoptosis through down-regulation of p53, EGR1, and STAT1. The anti-apoptotic activity of <it>EBER1 </it>may be important in the rescue of Reed-Sternberg cells from drug-induced apoptosis and in the clinical behaviors of EBV<sup>+ </sup>HLs.</p

    Implementation of academic study on the execution of service learning : an example of Cosmetics and Healthcare Department, Cardinal Tien Junior College Of Health Care And Management

    The Ministry of Education has developed Service Learning aiming to combine the context of service and learning so that students can be enlightened of care and the learning to acquire knowledge and skills through practical services. The 2011 academic statistics show that over 139 colleges and universities in Taiwan has set up service learning units and 135 of them have incorporated service learning courses into official course credits (Ministry of Education, 2012). The purpose of this study is to investigate the professional curriculum of Cosmetics and Healthcare Department into Service Learning . The paper takes the example from the Cosmetics and Healthcare Department, Cardinal Tien Junior College of Health Care and Management to conduct qualitative research literature review and interview. The paper clarifies the implications of service learning and applies informal conversational interview to analyze the implementation and effectiveness of integrating cosmetics vocational education with service learning on the 3 teachers, 51 students and 5members of service agencies participating in service learning, and participant observations to accomplish data collection

    Natural orifice transluminal endoscopic surgery: A transtracheal approach for the thoracic cavity in a live canine model

    BackgroundThe present study aimed to evaluate the performance of transtracheal thoracic exploration and pericardial window creation in a canine survival model.MethodsTransthoracic exploration was performed in 14 dogs. Under general anesthesia, after an incision in the right lateral wall of the middle–lower portion of the trachea was made, a 9-mm metal tube was advanced into the thoracic cavity. For thoracic cavity exploration and pericardial window creation, a flexible bronchoscope was introduced through the metal tube into the thoracic cavity. After thoracoscopy, a Dumon stent (Novatech, Grasse, France) was used to cover the tracheal incision site and facilitate healing. Animals were evaluated by endoscopy 1 and 2 weeks later. Animals were humanely killed, and necropsy was performed 2 weeks after the transtracheal natural orifice transluminal endoscopic surgery.ResultsFourteen dogs underwent transtracheal thoracic exploration lasting for an average of 110 minutes (range, 80–150), with 3 perioperative deaths. At 2 weeks after pericardial window creation, endoscopy revealed normal healing of the tracheal incision sites in all 11 surviving animals. Necropsy on the 11 animals at 2 weeks showed 9 adhesions around the pericardial window and 5 adhesions around the tracheal incision region. No mediastinitis or abscesses could be identified.ConclusionsTranstracheal thoracic exploration is technically feasible. Increasing surgical experience together with improvement in endoscopic techniques will further facilitate the development of natural orifice transluminal endoscopic surgery for thoracic diseases

    Variability of the giant X-ray bump in GRB 121027A and possible origin

    The particular giant X-ray bump of GRB 121027A triggered by \emph{Swift} is quite different from the typical X-ray flares in gamma-ray bursts. There exhibit four parts of the observed structural variabilities in the rise and decay phase of the bump. Considering the quality of four parts of the data, we can only analyze the data from about 5300 s to about 6100 s in the bump using the stepwise filter correlation method (Gao et al. 2012), and find that the 869.4+5.9 s86^{+5.9}_{-9.4}~\rm s periodic oscillation may exist, which is confirmed by the Lomb-Scargle method (Scargle 1982). Furthermore, a jet precession model (Liu et al. 2010) is proposed to account for such a variability.Comment: 5 pages, 3 figures, accepted for publication in MNRA
